The complement pathway is an important host defense system that contributes to both innate and acquired immunity. There are three pathways of complement activation: the classical pathway, lectin pathway and alternative pathway. Complement protein Factor I is a key serine protease that modulates the complement cascade by regulating the levels of C3 convertases. It circulates in plasma as a heavily N-glycosylated heterodimer made up of two disulfide linked chains, each carrying three N-linked oligosaccharide chains that may have both structural and functional roles in the interactions with the natural substrate and the cofactor during catalysis. Factor I is a serine protease with a high degree of specificity for C3b and C4b. It requires protein cofactors for cleavage of these complement proteins; Factor H, CR1 or MCP are required for C3b cleavage, and C4bp or CR1 are required for C4b cleavage.

MEPE is a 525 amino acid extracellular matrix protein. Expressed in osteocytes and brain, MEPE is a regulator of bone metabolism that is thought to mediate mineralisation and demineralisation within the osteocyte microenvironment. MEPE contains an RGD cell-attachment motif and shares molecular similarities with several dentin-bone extracellular matrix RGD-containing phosphoglycoproteins, including OPN (osteopontin) and DSP (dentin sialophosphoprotein). Via its ability to control bone mineralisation, MEPE is associated with various developmental events such as skeletogenesis, bone regeneration and odontogenesis. MEPE is secreted in hypophosphatemic osteomalacia tumors, suggesting a possible role in the pathophysiology of bone-related cancers.

Calcium-dependent secretion activators (CAPS-1 and CAPS-2) are calcium-binding proteins that direct neurotransmitter and neuropeptide-filled vesicles to the cell membrane for secretory granule exocytosis. Both CAPS-1 and CAPS-2 are expressed primarily in the brain where they regulate the secretion of various substances. The CAPS proteins contain a PH domain that is essential for regulation of exocytosis, as well as regulation of phospholipid binding. Through their regulation of neurotrophin release from granule cells, CAPS proteins help to regulate cell fate during neuronal development. CAPS-1 is thought to regulate catecholamine release from neuronal cells, while CAPS-2 is thought to regulate release of both brain-derived neurotrophic factor and neurotrophin-3 from granule cells. Defects in the genes encoding CAPS-1 and CAPS-2 are implicated in impaired cerebral development and autism.
