Beschreibung:
Protein kinase C (PKC) zeta is a member of the PKC family of serine/threonine kinases which are involved in a variety of cellular processes such as proliferation, differentiation and secretion. Unlike the classical PKC isoenzymes which are calcium-dependent, PKC zeta exhibits a kinase activity which is independent of calcium and diacylglycerol but not of phosphatidylserine. Furthermore, it is insensitive to typical PKC inhibitors and cannot be activated by phorbol ester. Unlike the classical PKC isoenzymes, it has only a single zinc finger module. These structural and biochemical properties indicate that the zeta subspecies is related to, but distinct from other isoenzymes of PKC. Non-selective calcium permeant cation channel probably involved in osmotic sensitivity and mechanosensitivity. Activation by exposure to hypotonicity within the physiological range exhibits an outward rectification. Also activated by low pH, citrate and phorbol esters. Increase of intracellular Ca(2+) potentiates currents. Channel activity seems to be regulated by a calmodulin-dependent mechanism with a negative feedback mechanism. Promotes cell-cell junction formation in skin keratinocytes and plays an important role in the formation and/or maintenance of functional intercellular barriers. Acts as a regulator of intracellular Ca(2+) in synoviocytes. Plays an obligatory role as a molecular component in the nonselective cation channel activation induced by 4-alpha-phorbol 12,13-didecanoate and hypotonic stimulation in synoviocytes and also regulates production of IL-8.

Mitogen-activated protein kinase (MAPK) Signalling pathways involve closely related MAP kinases, including extracellular-signal-related kinase 3 (ERK 3, also designated PRKM6 and p97MAPK). Serum, growth factors and phorbol esters can initiate ERK 3 Signalling pathways. Despite lacking a definitive nuclear localisation sequence, ERK 3 constitutively localizes to the nucleus upon activation. p38 pathway activation-dependent upregulation of ERK 3 is independent of the status of p53, Bcl-2 and caspase-3 during cell stress and damage induced by proteasome inhibition, suggesting ERK 3 in part mediates intracellular defense or cell rescue. The human ERK 3 gene maps to chromosome 15q21.2 and encodes a 721 amino acid protein.
