Zinc-finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. The majority of zinc-finger proteins contain a Krüppel-type DNA binding domain and a KRAB domain, which is thought to interact with KAP1, thereby recruiting histone modifying proteins. ZNF81, also known as HFZ20 or MRX45, is a transcriptional regulator belonging to the Krüppel C2H2-type zinc-finger protein family. It localizes to the nucleus and contains 12 C2H2-type zinc fingers and 1 KRAB domain. Mutations in the gene encoding ZNF81 are implicated in nonsyndromic X-linked mental retardation (XLMR).

Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. This MAb is a useful marker for melanocytes and melanomas.

This gene encodes a subunit of the transcription factor complex nuclear factor-kappa-B (NFkB). The NFkB complex is expressed in numerous cell types and functions as a central activator of genes involved in inflammation and immune function. The protein encoded by this gene can function as both a transcriptional activator or repressor depending on its dimerization partner. The p100 full-length protein is co-translationally processed into a p52 active form. Chromosomal rearrangements and translocations of this locus have been observed in B cell lymphomas, some of which may result in the formation of fusion proteins. The Thrombospondin proteins, Thrombospondins 1-4 and Thrombospondin 5 (also designated COMP), compose a family of glycoproteins that are involved in cell-to-cell and cell-to-matrix Signalling. These extracellular, cell-surface proteins form complexes of both homo- and heteromultimers. Spondin-2, or Mindin, is also designated DIL-1 for its differential expression in cancerous and non-cancerous lung cells. Full-length SPON2 cDNA encodes a 331 amino acid protein with a domain arrangement similar to zebrafish F-Spondin and Mindin-1/Mindin-2: an FS1 domain, an FS2 domain, a hydrophobic signal sequence in the N-terminus and a Thrombospondin type I repeat. Immunoblot analysis demonstrates expression of dimers and oligomers in a concentration-dependent manner under nonreducing conditions.

It is expressed on lymphocyte cells and is upregulated during T-cell activation. CD26 is also expressed on activated B cells and natural killer cells and abundantly on epithelia. CD26 is implicated in a variety of biological functions including T-cell activation, cell adhesion with extracellular matrix such as fibronectin or collagens, and in HIV infection. Cross-linking of CD26 using this antibody dramatically enhances the anti-CD3-induced IL-2 production. In Western blotting, this MAb reacts with only glycosylated CD26, but not with the deglycosylated form. It does not prevent ADA binding to CD26.

Parkinson's Disease, the second most common neurodegenerative disease after Alzheimer's Disease, is characterized by the loss of dopaminergic neurons and the presence of Lewy bodies (comprised of alpha synuclein and parkin inclusions). Autosomal Recessive Juvenile Parkinsonism (AR-JP) is a recently described form of Parkinson's Disease that has been linked to a gene that codes for parkin. Parkin, a 52 kDa protein, has a suggested role in the ubiquitin/proteasome pathway for protein degradation. The amino terminus bears sequence homology to ubiquitin while functionally it acts as a RING type ubiquitin protein ligase (E3) that coordinates the transfer of ubiquitin to substrate proteins, thus targeting them for degradation by the proteasome.

In human liver cytosolic fractions, four forms of biliverdin reductase have been identified, including two biliverdin-IX Beta reductases and two biliverdin-IX Alpha reductases, designated isozymes I and II and isozymes III and IV, respectively. Biliverdin reductase A (BLVRA), also designated biliverdin-IX Alpha-reductase, belongs to the GFO/iIDH/MocA family and the biliverdin reductase subfamily. The gene that encodes this cytoplasmic protein maps to chromosome 7p14-cen. BLVRA reduces biliverdin IX (the methene bridge of the open tetrapyrrole) to bilirubin with the concomitant oxidation of an NADH or NADPH cofactor (bilirubin + NADP+ = biliverdin + NADPH). BLVRA is expressed primarily in liver.

Functions both as NADH oxidoreductase and as regulator of apoptosis. In response to apoptotic stimuli, it is released from the mitochondrion intermembrane space into the cytosol and to the nucleus, where it functions as a proapoptotic factor in a caspase-independent pathway.
