G protein-coupled receptors (GPRs), also known as seven transmembrane receptors, heptahelical receptors or 7TM receptors, comprise a superfamily of proteins that play a role in many different stimulus-response pathways. G protein coupled receptors translate extracellular signals into intracellular signals (G protein activation) and they respond to a variety of signaling molecules, such as hormones and neurotransmitters. GPR101 (G protein-coupled receptor 101), also known as GPCR6, is a 508 amino acid multi-pass membrane protein that belongs to the G-protein coupled receptor 1 family. GPR101 functions as an orphan receptor that is thought to play a role in signaling events throughout the cell. Expressed predominantly in brain, GPR101 participates in a wide range of activities in the CNS via modulation of cAMP levels.

Tubulin family members are globular proteins important in the assembly of microtubules. Microtubules are structural components that play important roles in mitosis, cytokinesis and vesicle transport. TPPP (Tubulin polymerization-promoting protein), also known as p24 and p25, is a widely expressed 219 amino acid protein found in the perinuclear region of the cytoplasm. TPPP may form dimers and functions in polymerizing tubulin into double-walled tubules, polymorphic aggregates, or stabilized blocks. TPPP overexpression prevents formation of the mitotic spindle assembly and breakdown of the nuclear envelope. TPPP is phosphorylated by TPK II and is encoded by a gene that maps to human chromosome 5, which contains 181 million base pairs and comprises nearly 6% of the human genome.

BMAL2 is a 636 amino acid protein that localizes to the nucleus and contains one bHLH (basic helix-loop-helix) domain, one PAC (PAS-associated C-terminal) domain and two PAS (PER-ARNT-SIM) domains. Expressed at high levels in placenta and brain and at lower levels in liver, thymus, heart, lung and kidney, BMAL2 functions as a component of the circadian core oscillator, which includes a variety of proteins that work in tandem to activate the transcription of target genes. More specifically, BMAL2, when functioning as a component of the core oscillator, binds to the E-box element (3'-CACGTG-5') of target DNA, thus inducing transcription. Multiple isoforms of BMAL2 exist due to alternative splicing events.

The sodium-coupled neutral amino acid transporters (SNAT) of the SLC38 gene family include System A subtypes SNAT1, SNAT2 and SNAT4 and System N subtypes SNAT3 and SNAT5. The SLC38 transporters are essential for the uptake of nutrients, energy production, metabolism, detoxification, and the cycling of neurotransmitters. SNAT2, also designated ATA2, PRO1068 and SAT2 is encoded by the human gene SLC38A2. The functional role of SNAT2 in the nervous system is unclear. Protein expression is notably enriched in the spinal cord and brain stem nuclei of the auditory system. System A transport proteins are also present in placental tissue. These SNAT proteins may play a significant role in fetal development and inhibition of the transport system has been associated with fetal growth retardation.

Shootin1 is a 631 amino acid protein that belongs to the shootin family. The shootin1 protein contains three coiled-coil domains, a proline-rich region and interacts with RUFY3. Shootin1 is involved in the generation of internal asymmetric signals required for neuronal polarization. The shootin1 protein acts upstream of PI3K (phosphoinositide 3-kinase), by being required for spatially localized PI3K activity. By accumulating asymmetrically in a single neurite before polarization, shootin1 leads to axon induction for polarization, additionally the absence of shootin1 from the nascent axon's siblings by competition prevents the formation of surplus axons. Existing as seven alternatively spliced isoforms, the shootin1 gene is conserved in chimpanzee, dog, mouse, rat, chicken and zebrafish, and maps to human chromosome 10q25.3.

TNRC6B is a 1723 amino acid protein that exists as two alternatively spliced isoforms and is thought to be involved in mRNA cleavage events. Expressed ubiquitously, TNRC6B contains one glycine/tryptophan (GW)-rich N-terminal domain, one central glutamine-rich region and one C-terminal RNA recognition motif and is encoded by a gene that maps to human chromosome 22. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia. Additionally, translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia Chromosome and the subsequent production of the novel fusion protein BCR-Abl, a potent cell proliferation activator found in several types of leukaemias.

The transcriptional enhancer factor-1 (TEF-1) family of transcription factors regulate tissue-specific gene expression in muscle and placenta. The mechanism whereby TEF-1 confers tissue specificity depends largely on the interaction of TEF-1 with tissue-specific cofactors. Transcription cofactor Vgl-4 (vestigial-like protein 4) is a 290 amino acid nuclear protein that interacts with TEF-1 and MEF-2. Vgl-4 is the only member of the vestigial-like family that is expressed in heart. Overexpression of Vgl-4 in cardiac myocytes interferes with basal expression and ?-adrenergic receptor-dependent activation of a TEF-1 dependent skeletal ?actin promoter. This suggests that Vgl-4 counteracts ?-adrenergic activation of gene expression in cardiomyocytes. There are two isoforms of Vgl-4 that are produced as a result of alternative splicing events.

Serine protease that shows proteolytic activity against a non-specific substrate beta-casein. Promotes or induces cell death either by direct binding to and inhibition of BIRC proteins(also called inhibitor of apoptosis proteins, IAPs), leading to an increase in caspase activity, or by a BIRC inhibition-independent, caspase-independent and serine protease activity-dependent mechanism. Isoform 2 seems to be proteolytically inactive. [CATALYTIC ACTIVITY] Cleavage of non-polar aliphatic amino-acids at the P1 position, with a preference for Val, Ile and Met. At the P2 and P3 positions, Arg is selected most strongly with a secondary preference for other hydrophilic residues. [SUBUNIT] Homotrimer. Interacts with MXI2. The mature protein, but not the precursor, binds to BIRC2, BIRC3 and XIAP.

STK31 (Serine/threonine protein kinase 31) is similar to a mouse gene that encodes a putative protein kinase with a tudor domain, and shows testis specific expression. Alternative splicing results in multiple transcript variants encoding different isoforms. Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the g phosphate of ATP, onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. With more than 500 gene products, the protein kinase family is one of the largest families of proteins in eukaryotes.

BZW2, also known as HSPC028 or MSTP017, is a 419 amino acid protein that contains one W2 domain and is thought to be involved in neuronal differentiation. The gene encoding BZW2 maps to human chromosome 7. Chromosome 7 houses over 1,000 genes and comprises nearly 5% of the human genome. Defects in some of the genes localised to chromosome 7 have been linked to Osteogenesis imperfecta, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome. The deletion of a portion of the q arm of chromosome 7 is associated with Williams-Beuren syndrome, a condition characterised by mild mental retardation, an unusual comfort and friendliness with strangers and an elfin appearance. Deletions of portions of the q arm of chromosome 7 are also seen in a number of myeloid disorders, including cases of acute myelogenous leukemia and myelodysplasia.
