Staurosporine is provided in a lyophilised form. It may be reconstituted by dissolving the lyophilised reagent in DMSO (5 mg/ml - clear, colourless solution), DMF, ethyl acetate (5 mg/ml - clear colourless solution) or methanol. SIGIRR Antibody: SIGIRR is a member of the Toll-like receptor-interleukin 1 receptor superfamily. Members of this family are defined by the presence of an intracellular Toll-IL-1R (TIR) domain. The Toll-like receptors (TLRs) are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. SIGIRR was originally identified through database analysis and was shown to have only one Ig domain as opposed to the normal three Ig folds seen in the TIR family. Similar to ST2, another TIR family member, it has been shown to negatively regulate IL-1 receptor and Toll-like receptor signaling. However, SIGIRR inhibits TLR-IL-1R signaling by dimerizing with TLR4, TLR5, TLR9, and IL-1R. It also associates with the down-stream TLR signaling proteins IRAK and TRAF6 in an IL-1-dependent fashion.

Interleukin 34 (IL-34) was originally identified in humans, by large scale screening of secreted proteins; orthologs have also been found in chimpanzee, murine, rat and chicken. Human IL-34 is synthesized as a 242 amino acid (aa) precursor that contains a 20 aa signal sequence and a 222 aa mature chain. IL-34 is secreted as a homodimer and contains one potential N-glycosylation site. IL-34 increases proliferation of immune cells known as monocytes; it elicits its activity by binding the Colony Stimulating Factor 1 Receptor (CSF-1R). Expression of IL-34 is most abundant in the spleen and is expressed in many other tissues including the heart, brain, liver, kidney, thymus, intestine and lung. Human IL-34 contains a C-terminal 6x-polyhistidine (6xHis) tag.

Brefeldin A is a fungal metabolite which is a macrocyclic lactone exhibiting a wide range of antibiotic activity. Produced by Penicillium brefeldianum. Blocks binding of the cytosolic coat protein b-COP and ARF to Golgi membranes mediated by protein G. Also blocks protein transportation into post-Golgi compartments. It activates the sphingomyelin cycle. Brefeldin A mediated apoptosis has been observed in human tumor cells. Brefeldin A reversibly inhibits the intracellular translocation of proteins in eukaryotes, e.g., during transport of proteins to the cell surface for secretion or expression. It has been reported to block the response of cultured cells to cholera toxin. In HepG2 cells, BFA induces two blocks in the secretory pathway; one at the level of the endoplasmic reticulum-Golgi juncture and the other in the trans-Golgi network. Brefeldin A is used in the studies of Brefeldin A-inhibited Guanine Nucleotide-exchange Protein, BIG2, Regulates the Constitutive Release of TNFR1 Exosome-like Vesicles. Brefeldin A (BFA) is a fungal metabolite which disrupts the structure and function of the Golgi apparatus. BFA is an activator of the sphingomyelin cycle. Brefeldin A-mediated apoptosis has been observed in human tumor cells.
