Leptin, the product of the ob (obese) gene, is a 16kD protein consisting of 146 amino acid residues. Leptin is produced in the adipose tissue, and is considered to play an important role in appetite control, fat metabolism and regulation of body weight. It targets the central nervous system, particularly hypothalamus, affecting food intake. Leptin levels are high in most obese individuals. Studies have shown that it may also influence reproductive function.

This protein regulates the activity of many immediate-early genes, for example c-fos, and thereby participates in cell cycle regulation, apoptosis, cell growth, and cell differentiation. This gene is the downstream target of many pathways; for example, the mitogen-activated protein kinase pathway (MAPK) that acts through the ternary complex factors (TCFs). Anaphylatoxin, a chemotactic and inflammatory peptide, is a receptor for C5a anaphylatoxin chemotactic receptor (C5L2), also designated G protein-coupled receptor 77. C5L2 is an integral membrane protein that belongs to the Gprotein-coupled receptor 1 family. C5L2 weakly couples to Gi-mediated signaling pathways and is widely expressed in the hippocampus, hypothalamus, pons, frontal cortex and liver. It is coexpressed with the C5a receptor (C5AR) on polymorphonuclear neutrophils and may modulate the activity of C5AR. The function of the C5a receptors, particularly that of C5L2, is still unclear. Research suggests that C5L2 may be required for the in vitro release of high mobility group box 1 protein (HMGB1).

Defects in G6PD are the cause of chronic non-spherocytic hemolytic anaemia (CNSHA) . Deficiency of G6PD is associated with hemolytic anaemia in two different situations. First, in areas in which malaria has been endemic, G6PD-deficiency alleles have reached high frequencies (1% to 50%) and deficient individuals, though essentially asymptomatic in the steady state, have a high risk of acute hemolytic attacks. Secondly, sporadic cases of G6PD deficiency occur at a very low frequencies, and they usually present a more severe phenotype. Several types of CNSHA are recognized. Class-I variants are associated with severe NSHA; class-II have an activity <10% of normal; class-III have an activity of 10% to 60% of normal; class-IV have near normal activity.

Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA. EF-CBP2, also known as NECAB2 (N-terminal EF-hand calcium binding protein 2), neuronal calcium-binding protein 2 or synaptotagmin-interacting protein 2 (Stip-2), is a 386 amino acid cytoplasmic protein that contains one antibiotic biosynthesis monooxygenase (ABM) domain and two EF-hand domains. Expressed in brain, EF-CBP2 is suggested to bind metabotropic glutamate receptor 5 (mGluR-5) in a calcium-regulated manner. The gene encoding EF-CBP2 maps to human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, which is a gastrointestinal inflammatory condition.

Peroxisomes are single-membrane bound organelles present in virtually all eukaryotic cells. They are involved in numerous catabolic and anabolic pathways, including beta-oxidation of very long chain fatty acids, metabolism of hydrogen peroxide, plasmalogen biosynthesis and bile acid synthesis. The Peroxin gene family, which includes more than 20 members, is required for peroxisome biogenesis. Peroxin 5R, also known as PEX5-related protein or Peroxisome biogenesis factor 5-like, is a 626 amino acid protein that is mainly expressed in brain, with some expression in testis and pancreas. Peroxin 5R contains five TPR repeats, which enable protein-protein interactions and assembly of large multiprotein complexes. There are three isoforms of Peroxin 5R that are produced as a result of alternative splicing events. These isoforms bind C-terminal peroxisome-targeting signals in a similar manner to Peroxin-5. Peroxin 5R interacts with Rab 8b, possibly playing a role in vesicular trafficking and neurotransmitter release.

The product of this gene functions in the NF-kappaB signal transduction pathway. The encoded protein, and the similar and functionally redundant protein MAP3K7IP2/TAB2, forms a ternary complex with the protein kinase MAP3K7/TAK1 and either TRAF2 or TRAF6 in response to stimulation with the pro-inflammatory cytokines TNF or IL-1. Subsequent MAP3K7/TAK1 kinase activity triggers a signaling cascade leading to activation of the NF-kappaB transcription factor. The human genome contains a related pseudogene. Alternatively spliced transcript variants have been described, but their biological validity has not been determined.
