Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. AGPS is a 658 amino acid enzyme that is required for glycerolipid metabolism and ether lipid biosynthesis. localised to the inner aspect of the peroxisomal membrane, AGPS is likely part of a heterotrimeric complex that is also composed of GNPAT and a modified form of GNPAT. Containing one FAD-binding PCMH-type domain, AGPS utilizes FAD as a cofactor in the synthesis of alkyl-glycerone 3-phophate and a long-chain acid anion from 1-acteyl-glyerone 3-phosphate and a long-chain alcohol. Defects in the gene encoding AGPS results in rhizomelic chondrodysplasia punctata type 3, a disease characterised by vertebral disorders, severe mental retardation, cutaneous lesions, cataracts and rhizomelic shortening of the humerus and femur.

SynGAP (for synaptic GTPase activating protein), a novel synaptic RasGAP, is a brain-specific protein abundant in the cortex, hippocampus and olfactory bulb. SynGAP interacts with all three PDZ domains within PSD-95/ SAP 90 proteins. It represents one of three classes of mammalian RasGAPs and is specifically localized to excitatory synapses possessing NMDA receptors. SynGAP may be involved in the regulation of BDNF as well as Ras signaling. Its activity is inhibited by phosphorylation by CaMKII, which could result in the activation of the MAP kinase pathway.

Hip (HSP70-interacting protein), also known as ST13 (suppression of tumorigenicity protein 13), is one of several co-chaperones that regulate activities of the HSP70 chaperone family (1,2). The homo-oligomeric protein Hip cooperates with HSP70 in protein folding by stabilizing the ADP-bound state of HSP70. Hip directly binds to the ATPase domain of HSP70 when it is converted to the ADP-bound state by proteins of the HSP40 family (3). By collaborating with other positive co-factors such as HSP40 and Hop, or competing with negative co-factors such as Bag1, Hip may facilitate the chaperone function of HSP70 in protein folding and repair, and in controlling the activity of regulatory proteins such as steroid receptors and various regulators of proliferation or apoptosis (4-8).

Copine 6 is a 557 amino acid member of the copine family of evolutionarily conserved, soluble, calcium-dependent, membrane-binding proteins. Members of the copine family are involved in signal transduction and membrane trafficking. Arabidopsis thaliana mutants lacking copine proteins exhibit reduced cell number and smaller cell size, effects which may be due to a defect in vesicle fusion or transport. Copine 6 contains two N-terminal C2 domains and one C-terminal VWFA (von Willebrand factor A) domain, which is also referred to as the A domain or the core domain. As is characteristic of the copine family, copine 6 functions in membrane trafficking and is capable of binding phospholipids in a calcium-dependent manner. Copine 6 may also play a role in synaptic plasticity.
