KLHL28 is a 571 amino acid protein similar to the Drosophila kelch protein. KLHL26 contains six kelch repeats and one BTB (POZ) domain. The BTB (broad complex, tramtrack and bric-a-brac) domain, also known as the POZ (Poxvirus and zinc finger) domain, is an N-terminal homodimerization domain that contains multiple copies of kelch repeats and/or C2H2-type zinc fingers. Proteins that contain BTB domains are thought to be involved in transcriptional regulation via control of chromatin structure and function. KLHL28 is expressed as two isoforms produced by alternative splicing.

It also converts bicarbonate in carbon dioxide and water. This reaction occurs spontaneously in aqueous solution, but does so slowly. Carbonic anhydrase can greatly increase the rate of the reaction, with typical catalytic rates of the different forms of this enzyme ranging between 104 and 106 reactions per second. The active site of most carbonic anhydrases contains a zinc ion.

MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Function of gp100 is not known but it is reported to be a useful marker for melanocytes and melanomas. This cocktail of three markers is designed for extremely sensitive labeling of formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation.
