TESK2 is a nuclear protein that belongs to the protein kinase superfamily and is expressed in testis and prostate. Functioning as a dual-specificity protein kinase, TESK2 catalyzes the ATP-dependent phosphorylation of substrates and autophosphorylation on tyrosine and serine/threonine residues, thereby mediating intracellular signal transduction pathways. TESK2 requires magnesium as a cofactor and its catalytic activity is thought to play an important role in meiotic events such as spermatogenesis. TESK2 contains one protein kinase domain that is 65% identical to the kinase domain found in TESK1 (testicular protein kinase 1), suggesting a similar role for these proteins in phosphorylation events. Three isoforms of TESK2 are expressed due to alternative splicing.

Alternative splicing of this gene results in three tissue-specific forms of glucokinase, one found in pancreatic islet beta cells and two found in liver. In contrast to other forms of hexokinase, this enzyme is not inhibited by its product glucose-6-phosphate but remains active while glucose is abundant. Mutations in this gene have been associated with non-insulin dependent diabetes mellitus (NIDDM), maturity onset diabetes of the young, type 2 (MODY2) and persistent hyperinsulinemic hypoglycemia of infancy (PHHI).

RecQ helicases are implicated in DNA recombination and repair helicases and are critical in genome maintenance. RecQL5 is one of five known RecQ homologues in humans, and defects in three of these (BLM, WRN and RecQ4) are associated with autosomal recessive disorders that involve genomic instability and a predisposition to cancer. The exact role of ReqL5 is unclear. It has been shown in mouse embryonic stem (ES) cells that mutations in the Recql5 genes result in a significant increase in the frequency of sister chromatid exchange (SCE), whereas deleting Recql5 lead to an even higher frequency of SCE. MARVELD1, (MARVEL domain-containing 1), is one of the MARVEL (MAL and related proteins for vesicle trafficking and membrane link) domain-containing proteins. It is a nuclear-localized protein that is widely expressed in normal human tissues. While it has been reported to be overexpressed in colon cancer, MARVELD1 protein levels are generally lower in several primary tumors including ovary, breast, kidney, bladder and liver. This lower level of MARVELD1 expression in tumors is thought to be due to a higher methylation status of the MARVELD1 promoter as treatment of MDA-MB-361 with 5-aza-2'-deoxycytidine restores MARVELD1 expression, suggesting that loss of MARVELD1 expression may contribute to tumorgenicity.

This gene is a proto-oncogene that encodes a RING finger E3 ubiquitin ligase. The encoded protein is one of the enzymes required for targeting substrates for degradation by the proteasome. This protein mediates the transfer of ubiquitin from ubiquitin conjugating enzymes (E2) to specific substrates. This protein also contains an N-terminal phosphotyrosine binding domain that allows it to interact with numerous tyrosine-phosphorylated substrates and target them for proteasome degradation. As such it functions as a negative regulator of many signal transduction pathways. This gene has been found to be mutated or translocated in many cancers including acute myeloid leukaemia. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

Mutations in the CLN8 gene cause neuronal ceroid lipofuscinosis 8 and progressive epilepsy with mental retardation (EPMR). Both disorders are forms of neuronal ceroid-lipofuscinose (NCL), a group of progressive neurodegenerative diseases found in children, characterised by failure of psychomotor development, impaired vision, seizures and premature death. The CLN8 protein is one of eight proteins in the CLN family, including CLN1-CLN7, which are associated with NCL.
