CREB3L3 is a 461 amino acid single-pass type II membrane protein that localizes to the endoplasmic reticulum (ER) and, in response to ER stress, is cleaved and translocated to the nucleus. Expressed exclusively in liver, CREB3L3 functions as a transcription factor that, during ER stress, is thought to activate genes that are involved in both the unfolded protein response and the acute phase response (APR). Additionally, CREB3L3 is underexpressed in hepatocellular carcinoma, suggesting a possible role as a tumor suppressor. CREB3L3 functions as a dimer and contains one leucine zipper domain, a KDEL-like sequence and a bZIP domain, through which it conveys its DNA binding ability. Three isoforms of CREB3L3 exist due to alternative splicing events.

Present in eukaryotic proteins that are involved in ubiquitination and protein trafficking pathways, CUE domains can bind monoubiquitin and may be required for ubiquitination of the proteins in which they are found. CUEDC1 (CUE domain-containing protein 1) is a 386 amino acid protein that contains one CUE domain, suggesting a possible role in protein trafficking and degradation pathways. Defects in the gene encoding CUEDC1 may be associated with early stage cervical cancer, implicating CUEDC1 as a potential tumor marker. Two isoforms of CUEDC1 exist due to alternative splicing events.

However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

ZBED1 is a 694 amino acid protein that localizes specifically to granular structures within the nucleus. Expressed ubiquitously at low levels and present at higher levels in heart, placenta, spleen and skeletal muscle, ZBED1 is thought to function as a transcription factor that regulates a number of ribosomal protein (RP) encoding genes, thereby playing a role in the cell cycle and in cell proliferation events. ZBED1 contains one BED-type zinc finger and binds specifically to 5'-TGTCG[CT]GA[CT]A-3' DNA regions found in RP promotors. Additionally, ZBED1 binds strongly to the promotor region of Histone H1 (a protein required for the condensation of nucleosomes into higher order structures), subsequently activating H1 transcription.

Copine 6 is a 557 amino acid member of the copine family of evolutionarily conserved, soluble, calcium-dependent, membrane-binding proteins. Members of the copine family are involved in signal transduction and membrane trafficking. Arabidopsis thaliana mutants lacking copine proteins exhibit reduced cell number and smaller cell size, effects which may be due to a defect in vesicle fusion or transport. Copine 6 contains two N-terminal C2 domains and one C-terminal VWFA (von Willebrand factor A) domain, which is also referred to as the A domain or the core domain. As is characteristic of the copine family, copine 6 functions in membrane trafficking and is capable of binding phospholipids in a calcium-dependent manner. Copine 6 may also play a role in synaptic plasticity.

CMT4J is a recessive demyelinating disorder of the peripheral nervous system and is characterized by reduced motor nerve conduction velocities and axonal degeneration. Unlike CMT4J, ALS11 is characterized by the degeneration of upper motor neurons in the brain and lower neurons in the spinal cord, causing paralysis and, ultimately, death.

It represents one of three classes of mammalian RasGAPs and is specifically localized to excitatory synapses possessing NMDA receptors. Localized to both the cytoplasm and the nucleus, GMEB-1 forms a heterodimer with GMEB-2 (Glucocorticoid modulatory element-binding protein 2) and, once associated with GMEB-2, plays a key role in parvovirus DNA replication. In addition, GMEB-1 functions alone as a trans-acting factor that, by binding to glucocorticoid modulatory elements (GMEs) in TAT (tyrosine aminotransferase) promoters, increases intracellular sensitivity to glucocorticoid concentrations. GMEB-1 also interacts with initiator procaspases and, via this interaction, can inhibit caspase-induced apoptosis. Due to alternative splicing events, GMEB-1 is expressed as two isoforms.
