This gene encodes a member of the NCK family of adaptor proteins. The protein contains three SH3 domains and one SH2 domain. The protein has no known catalytic function but has been shown to bind and recruit various proteins involved in the regulation of receptor protein tyrosine kinases. It is through these regulatory activities that this protein is believed to be involved in cytoskeletal reorganisation. Alternate transcriptional splice variants, encoding different isoforms, have been characterised.

VAX2 is a 290 amino acid homeobox protein that plays a crucial role in development of the eye, particularly in the specification of the ventral optic vesicle and in establishment of a correct dorsoventral pattern. VAX2 acts as a transcription factor with VAX1 to cooperatively regulate retinal differentiation, neuroepithelial cell proliferation and axial polarization in the retina. Together, VAX1 and VAX2 repress transcription of Pax-6, a strong inducer of retinal development. VAX1 and VAX2 localize to the nucleus and contain one homeobox DNA-binding domain each.

HOXA5 upregulates both p53 promoter-reporter constructs and endogenous p53 synthesis, leading to apoptosis. HOXA5 is detectable in approximately one-third of primary tumors. Lack of HOXA5 expression strongly correlates with methylation of its promoter region, suggesting a causal role for methylation in the silencing of HOXA5 gene expression. HOXA5 expression is an important step in tumorigenesis and loss of expression of p53 in human breast cancer may be primarily due to lack of expression of HOXA5.

This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma aminobutyric acid from L glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin dependent diabetes. This gene may also play a role in the stiff man syndrome.

LAIR2 is a member of the immunoglobulin superfamily. It was identified by its similarity to LAIR1, an inhibitory receptor present on mononuclear leukocytes. This gene maps to a region of 19q13.4, termed the leukocyte receptor cluster, which contains 29 genes in the immunoglobulin superfamily, including LAIR1. The function of this protein is unknown, although it is thought to be secreted and may help modulate mucosal tolerance. LAIR2 contains one Ig-like C2-type (immunoglobulin-like) domain. There are two named isoforms.

GPKOW is a potential RNA-binding protein consisting of one central G patch domain and two C-terminal KOW domains. T54 is a 476 amino acid protein belonging to the MOS2 family. It is a mammalian homolog of the Arabidopsis thaliana MOS2 (modifier of SNC1, 2) nuclear protein that is required for innate immunity. Similar to A. thaliana MOS2, T54 localizes to the nucleus and contains G patch and KOW domains, suggesting that T54 may play a similar role in mammalian innate immunity.

Streptavidin is biotin-binding protein that was originally isolated from Streptomyces avidinii. In contrast to avidin, streptavidin has no carbohydrate and has a mildly acidic pI of 5. Streptavidin products use a recombinant form of streptavidin having a mass of 53,000 daltons and a near-neutral pI. Streptavidin is a tetrameric protein, with each subunit binding one molecule of biotin with affinity similar to that of avidin. Guanidinium chloride will dissociate avidin and streptavidin into subunits, but streptavidin is more resistant to dissociation.

Glucosamine 6-phosphate N-acetyltransferase (GNA1), also designated phosphoglucosamine transacetylase or phosphoglucosamine acetylase, belongs to the GNA1 subfamily of the larger acetyltransferase family of proteins. GNA1, a peripheral membrane protein containing one N-acetyltransferase domain, is expressed in the colon and maps to cytoband 14q22.1. The protein localizes to the Golgi apparatus and the endosome. It is important for UDP-GlcNAc biosynthesis pathway. GNA1 catalyzes the synthesis of GlcNAc6P from AcCoA and GlcN6P, a step in the UDP-GlcNAc6P formation pathway.

The activity dependent neuroprotective protein (ADNP) gene is frequently amplified in many neoplasias, including breast, bladder, ovarian, pancreatic, and colon cancers. ADNP mRNA is abundantly expressed in distinct normal tissues, and high expression levels were encountered in malignant cells. ADNP is implicated in maintaining cell survival, perhaps through modulation of p53. The encoded protein contains one homeobox and nine zinc finger domains, suggesting that it functions as a transcription factor.

This gene is a member of the natriuretic peptide family and encodes a secreted protein which functions as a cardiac hormone. The protein undergoes two cleavage events, one within the cell and a second after secretion into the blood. The protein's biological actions include natriuresis, diuresis, vasorelaxation, inhibition of renin and aldosterone secretion, and a key role in cardiovascular homeostasis. A high concentration of this protein in the bloodstream is indicative of heart failure. Mutations in this gene have been associated with postmenopausal osteoporosis.

This gene is one of several genes encoding pulmonary-surfactant associated proteins (SFTPA) located on chromosome 10. Mutations in this gene and a highly similar gene located nearby, which affect the highly conserved carbohydrate recognition domain, are associated with idiopathic pulmonary fibrosis. The current version of the assembly displays only a single centromeric SFTPA gene pair rather than the two gene pairs shown in the previous assembly which were thought to have resulted from a duplication. The NADH-ubiquinone oxidoreductase complex (complex I) of the mitochondrial respiratory chain catalyzes the transfer of electrons from NADH to ubiquinone, and consists of at least 43 subunits. The complex is located in the inner mitochondrial membrane. This gene encodes the 24 kDa subunit of complex I, and is involved in electron transfer. Mutations in this gene are implicated in Parkinson's disease, bipolar disorder, schizophrenia, and have been found in one case of early onset hypertrophic cardiomyopathy and encephalopathy. A non-transcribed pseudogene of this locus is found on chromosome 19. [provided by RefSeq, Oct 2009].

Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Expression of this gene is regulated by steroid hormones and may be useful as a marker for breast cancer. An additional transcript variant has been identified, but its full length sequence has not been determined. [provided by RefSeq, Jul 2008].
