Myosin is a protein which is known to interact with actin in muscle and non-muscle cells. It contains two identical heavy chains and four light chains. Myosin molecules consist of two major regions: tails (rods) and heads. They aggregate into filaments through the tail region and interact with actin and with ATP through the head region. Multiple forms of myosin heavy chains exist for each muscle type-skeletal, cardiac, smooth and non-muscle isomyosin forms exist in different types of skeletal muscle, depending on the physiological function of the muscle. They are designated at type I (slow twitch) and type II (fast twitch).

Alzheimer's disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1;PSEN2) or the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Three alternative splice variants of PSEN1 have been identified.

PATJ associates with Claudin-1, CRB1 (a transmembrane protein that plays a role in epithelial cell polarity and photoreceptor development), and Pals1 (a Lin-7 associated protein). The PATJ/Pals1/CRB1 complex can form a tripartite tight junction in epithelial cells crucial to their integrity.

Mutations in the ALS2 gene result in a number of juvenile recessive motor neuron diseases (MNDs), including juvenile primary lateral sclerosis (JPLS), a recessive form of amyotrophic lateral sclerosis (ALS2); infantile onset ascending hereditary spastic paralysis (IAHSP); and a form of complicated hereditary spastic paraplegia (cHSP). The ALS2 gene encodes the Alsin protein. Alsin acts as a guanine nucleotide exchange factor for Rab5, a modulator of the endocytic pathway. Alsin is a cytosolic protein that is associated with small, punctate membrane structures. Therefore, Alsin may mediate membrane transport events, potentially linking endocytic processes and actin cytoskeleton remodeling. The ALS2 C-terminal-like protein (ALS2CL) also modulates Rab 5 activity.
