Beschreibung:   The immunophilins are a highly conserved family of cis-trans peptidyl-prolyl isomerases that bind to and mediate the effects of immunosuppressive drugs, such as cyclosporin, FK506 and rapamycin. Immunophilins have also been implicated in protein folding and trafficking within the endoplasmic reticulum (ER). FKBP11 (FK506-binding protein 11), also known as FKBP19 or peptidyl-prolyl cis-trans isomerase FKBP11, is a 201 amino acid single-pass membrane protein belonging to the FKBP-type PPIase family, a group of proteins known to catalyze the folding of proline-containing polypeptides. Containing one PPIase FKBP-type domain, FKBP11 is expressed in secretory tissues such as pancreas, pituitary, stomach, lymph node and salivary gland, and is encoded by a gene that maps to human chromosome 12q13.12. FK506 and rapamycin are known to inhibit FKBP11’s peptidyl-prolyl isomerase activity.

Beschreibung:   Protein kinases are enzymes that transfer a phosphate group from a phosphate donor onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. The protein kinase family is one of the largest families of proteins in eukaryotes, classified in 8 major groups based on sequence comparison of their tyrosine (PTK) or serine/threonine (STK) kinase catalytic domains. Epidermal Growth factor receptor (EGFR) is the prototype member of the type 1 receptor tyrosine kinases. EGFR overexpression in tumors indicates poor prognosis and is observed in tumors of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, esophagus, stomach and in squamous cell carcinoma.

Beschreibung:   ATRNL1 is a 1,379 amino acid single-pass type I membrane protein that may play a role in melanocortin signaling pathways that regulate energy homeostasis. The ATRNL1 protein contains a C-type lectin domain, a CUB domain, two EGF-like domains, six Kelch repeats, two laminin EGF-like domains and five PSI domains. ATRNL1 interacts with MC4-R in several regions known to be important in the regulation of energy homeostasis by melanocortins, such as the paraventricular nucleus of hypothalamus and the dorsal motor nucleus of the vagus. The ATRNL1 gene is conserved in dog, cow, mouse, rat, chicken, zebrafish and C. elegans, exists as two alternatively spliced isoforms and maps to human chromosome 10q25.3. Strong evidence of linkage to late-onset Alzheimer disease (LOAD) is linked to chromosome 10, which implicates a wide region and at least one disease-susceptibility locus.

Beschreibung:   Angiotensin II is a potent vasopressor hormone and a primary regulator of aldosterone secretion. It is an important effector controlling blood pressure and volume in the cardiovascular system. It acts through at least two types of receptors. This gene encodes the type 1 receptor which is thought to mediate the major cardiovascular effects of angiotensin II. This gene may play a role in the generation of reperfusion arrhythmias following restoration of blood flow to ischemic or infarcted myocardium. It was previously thought that a related gene, denoted as AGTR1B, existed; however, it is now believed that there is only one type 1 receptor gene in humans. At least five transcript variants have been described for this gene. Additional variants have been described but their full-length nature has not been determined. The entire coding sequence is contained in the terminal exon and is present in all transcript variants. [provided by RefSeq].

Beschreibung:   GRAMD2 is a 354 amino acid single-pass membrane protein that contains one GRAM domain and is encoded by a gene that maps to human chromosome 15q23. Encoding more than 700 genes, chromosome 15 is made up of approximately 106 million base pairs and is about 3% of the human genome. Angelman and Prader-Willi syndromes are associated with loss of function or deletion of genes in the 15q11-q13 region. In the case of Angelman syndrome, this loss is due to inactivity of the maternal 15q11-q13 encoded UBE3A gene in the brain by either chromosomal deletion or mutation. In cases of Prader-Willi syndrome, there is a partial or complete deletion of this region from the paternal copy of chromosome 15. Tay-Sachs disease is a lethal disorder associated with mutations of the HEXA gene, which is encoded by chromosome 15. Marfan syndrome is associated with chromosome 15 through the FBN1 gene.

Beschreibung:   Polycomb group (PcG) proteins are important for maintaining the transcriptionally repressed state of target genes and are thought to function via chromatin modification. PcG proteins assemble into multimeric protein complexes, which are involved in maintaining the transcriptional repressive state of genes over successive cell generations. PcG proteins are also required for normal maturation of myeloid progenitor cells. A putative PcG protein, L3MBTL3 (lethal(3)malignant brain tumor-like protein 3), also known as MBT-1 or KIAA1798, is a 780 amino acid protein containing three MBT repeats and one SAM (sterile alpha motif) domain. Localized to the nucleus, L3MBTL3 interacts with RING1B, another PcG protein that may be involved in the specification of anterior-posterior axis and cell proliferation in early development. L3MBTL3 exists as two isoforms produced by alternative splicing events.

Beschreibung:   Glutathione (GSH) is a tripeptide antioxidant that reduces disulfide bonds between cytoplasmic proteins. The constitutive enzyme glutathione reductase transforms glutathione into its reduced state, which ultimately can provide a measure of cellular toxicity. GSTCD (glutathione S-transferase, C-terminal domain containing) is a 633 amino acid protein belonging to the GSTCD family and contains one GST C-terminal domain. The gene encoding GSTCD maps to human chromosome 4, which represents approximately 6% of the human genome and contains nearly 900 genes. Notably, the Huntingtin gene, which is found to encode an expanded glutamine tract in cases of Huntington's disease, is on chromosome 4. FGFR-3 is also encoded on chromosome 4 and has been associated with thanatophoric dwarfism, achondroplasia, Muenke syndrome and bladder cancer. Chromosome 4 is also tied to Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease.

Beschreibung:   MAP (mitogen-activated protein) kinases play a significant role in many biological processes, including cell adhesion and spreading, cell differentiation and apoptosis. p38 alpha, p38 beta and p38 gamma, also known as MAPK14, MAPK11 and MAPK12, respectively, each contain one protein kinase domain and belong to the MAP kinase family. Expressed in different areas throughout the body with common expression patterns in heart, p38 proteins use magnesium as a cofactor to catalyze the ATP-dependent phosphorylation of target proteins. Via their catalytic activity, p38 alpha, p38 beta and p38 gamma are involved in a variety of events throughout the cell, including signal transduction pathways, cytokine production and cell proliferation and differentiation. The p38 proteins are subject to phosphoryation on Thr and Tyr residues, an event which is thought to activate the phosphorylated protein.

Beschreibung:   Recognizes the -chain of CD3, which consists of five different polypeptide chains (designated as γ, δ, ε, ζ, and η) with MW ranging from 16-28 kDa. The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. In cortical thymocytes, CD3 is predominantly intra-cytoplasmic. However, in medullary thymocytes, it appears on the T cell surface. CD3 antigen is a highly specific marker for T cells, and is present in majority of T cell neoplasms.

Beschreibung:   This gene is a proto-oncogene that encodes a RING finger E3 ubiquitin ligase. The encoded protein is one of the enzymes required for targeting substrates for degradation by the proteasome. This protein mediates the transfer of ubiquitin from ubiquitin conjugating enzymes (E2) to specific substrates. This protein also contains an N-terminal phosphotyrosine binding domain that allows it to interact with numerous tyrosine-phosphorylated substrates and target them for proteasome degradation. As such it functions as a negative regulator of many signal transduction pathways. This gene has been found to be mutated or translocated in many cancers including acute myeloid leukaemia. Mutations in this gene are also the cause of Noonan syndrome-like disorder.
