Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This mAb recognises all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.

The protein that coordinates these activities is transcription factor IID (TFIID), which binds to the core promoter to position the polymerase properly, serves as the scaffold for assembly of the remainder of the transcription complex, and acts as a channel for regulatory signals. TFIID is composed of the TATA-binding protein (TBP) and a group of evolutionarily conserved proteins known as TBP-associated factors or TAFs. TBPL1 encodes a protein that serves the same function as TBP and substitutes for TBP at some promoters that are not recognized by TFIID. It is essential for spermiogenesis and believed to be important in expression of developmentally regulated genes.

The CD4 molecule is a major receptor for human immunodeficiency virus (HIV), binding directly to the envelope glycoprotein gp120 on HIV, with the co-receptors being CCR5 or CXCR4. It has been shown that the V-like domains are critical for binding with HIV envelope gp120.

Mouse Legumain,also known as LGMN, is a cysteine protease belonging to peptidase family C13 and is expressed in kidney and placenta abundantly. LGMN has a strict specificity for hydrolysis of asparaginyl bonds. It can also cleave aspartyl bonds slowly, especially under acidic conditions. The mammalian legumain is involved in the processing of proteins for MHC class II antigen presentation in the lysosomal/endosomal system. It plays a role in the regulation of cell proliferation via its role in EGFR degradation. Legumain deficiency causes the accumulation of pro-Cathepsins B, H and L, another group of lysosomal cysteine proteases. Overexpression of Legumain in tumours is significant for invasion/metastasis. Mammalian legumain is inhibited by iodoacetamide and maleimides. Legumain activation appears to be autocatalytic and can be triggered by acidic pH.

Insulin-like growth factor-binding protein 4 (IGFBP-4) inhibits the actions of IGF-I and IGF-II. The distribution of IGFBP-4 in tissues is dependent on its glycosylation state, however it preferentially targets connective tissue (Firth and Baxter; Qin <i>et al.</i>). IGFBP-4 is also produced in liver, adrenal glands, Leydig cells, developing embryos, spinal cord, and thymic cortex. IGFBP-4 inhibits WNT/β-catenin signaling, which downregulates prostate cancer proliferation (Zhu <i>et al.</i>). In vitro studies suggest that IGFBP-4 inhibits proliferation of neuroblastoma and glioma, while promoting bone loss in osteosarcoma and multiple myeloma (Durai <i>et al.</i>).

Receptors for the Fc region of IgG (Fc gamma R) are members of the Ig superfamily that function in the activation or inhibition of immune responses. Human Fc gamma Rs are divided into three classes designated Fc gamma RI (CD64), Fc gamma RII (CD32), and Fc gamma RIII (CD16), which generate multiple isoforms, are recognized. The activating type receptor either has or associates noncovalently with an accessory subunit that has an immunoreceptor tyrosinebased activation motif (ITAM) in its cytoplasmic domain. Fc gamma RI binds IgG with high affinity and functions during early immune responses, whereas Fc gamma RII and RIII are low affinity receptors that recognize IgG as aggregates surrounding multivalent antigens during late immune responses. Three genes for human Fc gamma RII (A, B, and C) and one for mouse (Fc gamma RIIB), encoding type I transmembrane proteins with ITAM motifs (Fc gamma RII A and C) or ITIM motifs (Fc gamma RIIB) in their cytoplasmic domains, have been identified. Human CD32, also known as Low affinity immunoglobulin gamma Fc region receptor II-a (IgG Fc receptor II-a), Fc gamma RII A or FCGR2A Protein, is expressed on cells of both myeloid and lymphoid lineages as well as on cells of non-hematopoietic origin. Associated with an ITAM-bearing adapter subunit, FcR gamma , CD32a (Fc gamma RII A) delivers an activating signal upon ligand binding, and results in the initiation of inflammatory responses including cytolysis, phagocytosis, degranulation, and cytokine production. The responses can be modulated by signals from the co-expressed inhibitory receptors such as Fc gamma RII B, and the strength of the signal is dependent on the ratio of expression of the activating and inhibitory receptors.

SOCS1 Antibody: The Suppressor of cytokine signaling (SOCS) and cytokine-inducible SH2 proteins are a family of intracellular proteins which regulate the immune cell responses to cytokines. SOCS1 acts to suppress dendritic cell (DC) as well as T cell hyperactivation following cytokine signaling by inhibiting JAK tyrosine kinase, a kinase necessary for type I and II cytokine receptors to initiate signaling, by directly binding to the catalytic domain of the kinase. SOCS1 also possesses E3 ubiquitin protein ligase activity that results in the polyubiquitination of its target proteins and subsequent degradation by the proteosome. It is through this method that SOCS1 negatively regulates signaling by Toll-like receptors TLR2 and TLR4 by mediating the degradation of the TLR signaling adaptor protein TIRAP.

ZIP13 is a multipass membrane protein that belongs to the ZIP transporter subfamily LIV-1. Mutations in ZIP13 have recently been shown to cause a spondylocheiro dysplastic form of Ehlers-Danlos syndrome (SCD-EDS), a generalized skeletal dysplasia involving mainly the spine with clinical abnormalities of the hands in addition to EDS-like features. Other experiments have shown that ZIP13 is required for proper connective tissue development and is involved in BMP/TGF- signaling pathways.
