FBXO29, also designated F-box/WD repeat-containing protein 8 (FBXW8), is a 598 amino acid protein that contains one 40 amino acid F-box region, making it a member of the F-box family. FBXO29 also contains five WD repeats. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and recruitment for ubiquitination. F-box proteins are members of a large family that regulates cell cycle, immune response, signaling cascades and developmental programs by targeting proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkB-å and ∫-catenin, for degradation by the proteasome after ubiquitination. Functioning as a component of the SCF complex, FBXO29 is thought to recognize and bind to select phosphorylated proteins, thereby promoting their ubiquitination and subsequent degradation. FBXO29 exists as two isoforms as a result of alternative splicing events.

The leucine-rich (LRR) repeat is a 20-30 amino acid motif that forms a hydrophobic å/∫ horseshoe fold, allowing it to accommodate several leucine residues within a tightly packed core. The primary function of these motifs is to provide a versatile structural framework to mediate the formation of protein-protein interactions. LRRs are present in a variety of proteins with diverse structure and function, including innate immunity and nervous system development. Several human diseases are associated with mutations in genes encoding LRR-containing proteins. The leucine-rich repeat-containing protein 39 (LRRC39) is a 335 amino acid protein that contains nine LRR repeats and exists as two alternatively spliced isoforms.

The endothelin (ET) family of proteins, which includes ET-1 (endothelin-1), ET-2 (endothelin-2) and ET-3 (endothelin-3), are vasoactive peptides that are involved in various functions throughout the body. Endothelins can affect the central nervous system and neuronal excitability, and they elicit potent vasoconstrictor action. While ET-1 is a potent, 21-amino acid vasoconstrictor peptide, ET-2 has the most potent vasoconstrictor activity. ET-3 functions as a ligand for endothelin receptor type B (ETBR) and, through this interaction, mediates the maturation of enteric neurons and melanocytes. Although ET-3 is expressed as a 238 amino acid peptide, it is post-translationally modified to produce a short active isoform and a long inactive isoform. Calcipressin-3, also known as RCAN3 and DSCR1L2 (Down syndrome candidate region 1-like protein 2), is a 241 amino acid protein that potentially is involved in central nervous system development. Calcipressin-3 interacts with cardiac troponin I, suggesting that it may play a role in cardiac contraction events.

Tyrosine hydroxylase is an excellent marker for dopaminergic and noradrenergic neurons. Tyrosine hydroxylase (a.k.a. tyrosine 3-monooxygenase) is the enzyme responsible for catalyzing the conversion of the amino acid L-tyrosine to L-3,4-dihydroxyphenylalanine (L-DOPA). L-DOPA is a precursor for dopamine, which, in turn, is a precursor for the important neurotransmitters norepinephrine (noradrenaline) and epinephrine (adrenaline). Tyrosine hydroxylase catalyzes the rate limiting step in this synthesis of catecholamines. In humans, tyrosine hydroxylase is encoded by the TH gene, and the enzyme is present in the central nervous system (CNS), peripheral symphatic neurons and the adrenal medulla. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing.
