Gentamicin is an aminoglycoside antibiotic complex produced by fermentation of Micromonospora purpurea or M. echinospora. It is a mixture of 3 major components designated as C1, C1a, and C2. Gentamicin is used as the sulfate salt. Each component consists of five basic nitrogens and requires five equivalents of sulfuric acid per mole of gentamicin base.
Mode of action: Gentamicin causes codon misreading by binding to the 30S ribosomal subunit, blocking the translocation of peptidyl-tRNA from the acceptor site to the donor site. The bactericidal effect of gentamicin on Pseudomonas aeruginosa is exerted by the binding of gentamicin to the outer membrane, where it displaces natural cations, destabilizes the membrane, and forms holes in the cell surface.Antimicrobial spectrum: Gram-negative bacteria, Staphylococcus aureus and other Gram-positive bacteria.
Gentamicin sulfate is a broad spectrum antibiotic. It inhibits the growth of a wide variety of Gram-positive and Gram-negative microorganisms, including strains resistant to tetracycline, chloramphenicol, kanamycin, and colistin, particularly strains of Pseudomonas, Proteus, Staphylococcus, and Streptococcus. Gentamicin sulfate inhibits bacterial protein biosynthesis by binding to the 30S subunit of the ribosome.

Interleukins (ILs) are a large group of cytokines that are produced mainly by leukocytes, although some are produced by certain phagocytes and auxiliary cells. ILs have a variety of functions, but most function to direct other immune cells to divide and differentiate. Each IL acts on a specific, limited group of cells through a receptor specific for that IL. Human IL10 is a non glycosylated polypeptide consisting of 178 amino acids. There is 73% homology between the human and mouse IL10 proteins, however, the human IL10 acts on both human and mouse target cells, while the mouse IL10 has species specific activity. The cellular sources of IL10 are CD4+ T cells and T cell clones, thymocytes, B cells and B cell lymphomas, macrophages, mast cell lines and keratinocytes. IL10 will stimulate the growth of stem cells, mast cells and thymocytes. IL10 enhances cytotoxic T cell development, and costimulates B cell differentiation and immunoglobulin secretion. IL10 inhibits cytokine production by macrophages and suppresses macrophage class II MHC expression. The human IL10 gene is on human chromosome

Adaptor proteins are usually required for this activation, possibly to acetylate and destabilize nucleosomes, thereby relieving chromatin constraints at the promoter. The protein encoded by this gene is a transcriptional activator adaptor and has been found to be part of the PCAF histone acetylase complex. Initiation of transcription by RNA polymerase II requires the activities of more than 70 polypeptides. The protein that coordinates these activities is transcription factor IID (TFIID), which binds to the core promoter to position the polymerase properly, serves as the scaffold for assembly of the remainder of the transcription complex, and acts as a channel for regulatory signals. TFIID is composed of the TATA-binding protein (TBP) and a group of evolutionarily conserved proteins known as TBP-associated factors or TAFs. TAFs may participate in basal transcription, serve as coactivators, function in promoter recognition or modify general transcription factors (GTFs) to facilitate complex assembly and transcription initiation. This gene encodes TBP, the TATA-binding protein. A distinctive feature of TBP is a long string of glutamines in the N-terminal. This region of the protein modulates the DNA binding activity of the C terminus, and modulation of DNA binding affects the rate of transcription complex formation and initiation of transcription. Mutations that expand the number of CAG repeats encoding this polyglutamine tract, and thus increase the length of the polyglutamine string, are associated with spinocerebellar ataxia 17, a neurodegenerative disorder classified as a polyglutamine disease.

Renin is also known as REN and angiotensinogenase, is a circulating enzyme that participates in the body's renin-angiotensin system (RAS), and plays an essential role in the elevation of arterial blood pressure and increased sodium retention by the kidney. Renin activates the renin-angiotensin system by cleaving angiotensinogen, produced by the liver, to yield angiotensin I, which is further converted into angiotensin II by ACE, the angiotensin-converting enzyme primarily within the capillaries of the lungs. Renin is secreted from kidney cells, which are activated via signaling from the macula densa, which responds to the rate of fluid flow through the distal tubule, by decreases in renal perfusion pressure (through stretch receptors in the vascular wall), and by sympathetic nervous stimulation, mainly through beta-1 receptor activation. Renin can bind to ATP6AP2, which results in a fourfold increase in the conversion of angiotensinogen to angiotensin I over that shown by soluble renin. In addition, renin binding results in phosphorylation of serine and tyrosine residues of ATP6AP2. The level of renin mRNA appears to be modulated by the binding of HADHB, HuR and CP1 to a regulatory region in the 3' UTR. An over-active renin-angiotension system leads to vasoconstriction and retention of sodium and water. These effects lead to hypertension. Therefore, renin inhibitors can be used for the treatment of hypertension.

This mAb recognises a protein of 41 kDa, which is identified as TDP2, or ETS1 associated protein II. It is a member of a superfamily of divalent cation-dependent phosphodiesterases. The encoded protein associates with CD40, tumor necrosis factor (TNF) receptor-75 and TNF receptor associated factors (TRAFs), and inhibits nuclear factor-kappa-B activation. This protein has sequence and structural similarities with APE1 endonuclease, which is involved in both DNA repair and the activation of transcription factors. DNA repair enzyme that can remove a variety of covalent adducts from DNA through hydrolysis of a 5'-phosphodiester bond, giving rise to DNA with a free 5' phosphate. catalyses the hydrolysis of dead-end complexes between DNA and the topoisomerase 2 (TOP2) active site tyrosine residue. Hydrolyzes 5'-phosphoglycolates on protruding 5' ends on DNA double-strand breaks (DSBs) due to DNA damage by radiation and free radicals. The 5'-tyrosyl DNA phosphodiesterase activity can enable the repair of TOP2-induced DSBs without the need for nuclease activity, creating a 'clean' DSB with 5'-phosphate termini that are ready for ligation. Has also 3'-tyrosyl DNA phosphodiesterase activity, but less efficiently and much slower than TDP1. May also act as a negative regulator of ETS1 and may inhibit nuclear factor-kappa-B activation.

Oxidized Low-Density Lipoprotein Receptor 1 (Ox-LDL Receptor 1) is a secreted, single-pass type II membrane protein which belongs to the C-type lectin superfamily. Ox-LDL Receptor 1 is expressed at high levels in endothelial cells and vascular-rich organs such as placenta, lung, liver, brain, aortic intima, bone marrow, spinal cord and substantia nigra. The expression of Ox-LDL Receptor 1 is induced by inflammatory cytokines such as TNF, IFNG and IL6 by pathological conditions, such as hyperlipidemia, hypertension and diabetes mellitus. Ox-LDL Receptor 1 mediates the recognition, internalization and degradation of oxidatively modified low density lipoprotein (OxLDL) by vascular endothelial cells. Ox-LDL Receptor 1 association with oxLDL induces the activation of NF-kappa-B through an increased production of intracellular reactive oxygen and a variety of pro-atherogenic cellular responses including a reduction of nitric oxide (NO) release, monocyte adhesion and apoptosis. Ox-LDL Receptor 1 also binds to oxLDL, which acts as a receptor for the HSP70 protein involved in antigen cross-presentation to naive T-cells in dendritic cells, thereby participating in cell-mediated antigen cross-presentation. It also participates in inflammatory process, by acting as a leukocyte-adhesion molecule at the vascular interface in endotoxin-induced inflammation.

Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. This antibody detects acidic (Type I or LMW) and basic (Type II or HMW) cytokeratins: 67 kDa (CK1); 64 kDa (CK3); 59 kDa (CK4); 58 kDa (CK5); 56 kDa (CK6); 52 kDa (CK8); 56.5 kDa (CK10); 50 kDa (CK14); 50 kDa (CK15); 48 kDa (CK16); 40 kDa (CK19). Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. It is a broad spectrum anti pan-cytokeratin antibody, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer. It may be useful to characterize the source of various neoplasms and to study the distribution of cytokeratin containing cells in epithelia during normal development and during the development of epithelial neoplasms. This antibody stains cytokeratins present in normal and abnormal human tissues and has high sensitivity in the recognition of epithelial cells and carcinomas.

Interleukin-18 (IL-18) is a costimulatory factor for production of interferon-gamma (IFN-gamma) in response to toxic shock and shares functional similarities with IL-12. IL-18 is synthesised as a precursor 24kDa molecule without a signal peptide and must be cleaved to produce an active molecule. IL-1 converting enzyme (ICE; Caspase-1) cleaves pro-IL-18 at aspartic acid in the P1 position, producing the mature, bioactive peptide that is readily released from the cells. It is reported that IL-18 is produced from Kupffer cells, activated macrophages, keratinocytes, intestinal epithelial cells, osteoblasts, adrenal cortex cells and murine diencephalon. IFN-gamma is produced by activated T or NK cells and plays critical roles in the defense against microbiral pathogens. IFN-gamma activates macrophages and enhances NK activity and B cell maturation, proliferation and Ig secretion. IFN-gamma also induces expression of MHC class I and II antigens and inhibits osteoclast activation. IL-18 acts on T helper type-1 (Th1) T cells and in combination with IL-12 strongly induces them to produce IFN-gamma. Pleiotropic effects of IL-18 have also been reported, such as enhancement production of IFN-gamma and GM-CSF in peripheral blood mononuclear cells, production of Th1 cytokines, IL-2, GM-CSF, IFN-gamma in T cells and enhancement of Fas ligand expression by Th1 cells.

UGT1A1 is a UDP-glucuronosyltransferase, an enzyme of the glucuronidation pathway that transforms small lipophilic molecules, such as steroids, bilirubin, hormones, and drugs, into water-soluble, excretable metabolites. The preferred substrate of this enzyme is bilirubin, although it also has moderate activity with simple phenols, flavones, and C18 steroids. This gene encodes a UDP-glucuronosyltransferase, an enzyme of the glucuronidation pathway that transforms small lipophilic molecules, such as steroids, bilirubin, hormones, and drugs, into water-soluble, excretable metabolites. This gene is part of a complex locus that encodes several UDP-glucuronosyltransferases. The locus includes thirteen unique alternate first exons followed by four common exons. Four of the alternate first exons are considered pseudogenes. Each of the remaining nine 5' exons may be spliced to the four common exons, resulting in nine proteins with different N-termini and identical C-termini. Each first exon encodes the substrate binding site, and is regulated by its own promoter. The preferred substrate of this enzyme is bilirubin, although it also has moderate activity with simple phenols, flavones, and C18 steroids. Mutations in this gene result in Crigler-Najjar syndromes types I and II and in Gilbert syndrome. Tyrosine kinase that plays an essential role for the selection and maturation of developing T-cell in the thymus and in mature T-cell function. Is constitutively associated with the cytoplasmic portions of the CD4 and CD8 surface receptors and plays a key role in T-cell antigen receptor(TCR)-linked signal transduction pathways. Association of the TCR with a peptide antigen-bound MHC complex facilitates the interaction of CD4 and CD8 with MHC class II and class I molecules, respectively, and thereby recruits the associated LCK to the vicinity of the TCR/CD3 complex. LCK then phosphorylates tyrosines residues within the immunoreceptor tyrosines-based activation motifs (ITAMs) in the cytoplasmic tails of the TCRgamma chains and CD3 subunits, initiating the TCR/CD3 signaling pathway. In addition, contributes to signaling by other receptor molecules. Associates directly with the cytoplasmic tail of CD2, and upon engagement of the CD2 molecule, LCK undergoes hyperphosphorylation and activation. Also plays a role in the IL2 receptor-linked signaling pathway that controls T-cell proliferative response. Binding of IL2 to its receptor results in increased activity of LCK. Is expressed at all stages of thymocyte development and is required for the regulation of maturation events that are governed by both pre-TCR and mature alpha beta TCR.

alpha-Actinin 4 is an actin-bundling protein of ~100kDa that is associated with cell motility, endocytosis and cancer invasion. The alpha-actinin family comprises two non-muscle isoforms (alpha-actinin-1 and -4) and two skeletal muscle isoforms (alpha-actinin-2 and -3), with alpha-actinin-2 being also expressed in cardiac muscle. While alpha-actinin-4 is almost ubiquitously expressed, particularly high concentrations are found in glomeruli. On the subcellular level it is associated with actin stress fibers, but in certain cells it also localizes to the nucleus. Mutations in the alpha-actinin-4 gene cause an autosomal-dominant form of familial focal segmental glomerulosclerosis (FSGS), which is thought to result from a defect in glomerular podocyte function. A point mutation in the alpha-actinin-4 gene was found to generate an antigenic peptide that is recognized by autologous cytolytic T lymphocytes (CTL) on a human lung carcinoma. alpha-Actinin-4 interacts with a variety of proteins, including the ring finger protein BERP, the PDZ-LIM protein CLP-36, the hemidesmosomal and cell-cell contact protein BP180, and the tight junction protein MAGI-1. Moreover, alpha-actinin-4 forms a ternary complex with Ca2+/Calmodulin-dependent protein kinase II and densin-180, a protein of postsynaptic densities in CNS neurons. Ca2+-dependent association of alpha-actinin-4 with E3KARP is required for Ca2+-dependent inhibition of the Na+/H+ exchanger 3 (NHE3).
