Recognises a protein of about 50 kDa, which is identified as gamma-enolase/neuron specific enolase/enolase 2. Three isoenzymes of enolases are identified, alpha, beta and gamma. Alpha-isoform is expressed in most tissues, whereas beta-form is expressed predominantly in muscle tissue and gamma-enolase is found only in nervous tissue. These isoforms exist as both homodimers and heterodimers, and they play a role in converting phosphoglyceric acid to phosphenolpyruvic acid in the glycolytic pathway. NSE is a useful marker to identify peripheral nerves and tumors of neuro-endocrine origins, such as pheochromocytomas. It it be usually employed in combination with other markers such as Synaptophysin, Chromogranin A, and Neurofilament.

Mutations in this gene have been associated with epilepsy and febrile seizures. PEN2 Antibody: PEN2, in addition to presenilin, nicastrin, and APH-1 forms the gamma-secretase protein complex, a membrane-bound aspartyl protease that can cleave certain proteins at peptide bonds buried within the hydrophobic environment of the lipid bilayer. This cleavage is responsible for a key step in signaling from several cell-surface receptors and is thought to be required for the generation of the neurotoxic amyloid peptides that are central to the pathogenesis of Alzheimer's disease. Like the tumor necrosis factor-alpha-converting enzyme (TACE) and the beta-site cleavage enzyme (BACE) protease families, gamma-secretase will cleave the amyloid precursor protein (APP), but within the intramembrane region of APP, resulting in either the non-toxic p3 (from the alpha and gamma cleavage site) or the toxic Abeta amyloid peptide (from the beta and gamma cleavage site). It is thought that accumulation of the Abeta peptide is the precursor to Alzheimer's disease.

TMP21 Antibody: TMP21 is a ubiquitously expressed protein that is involved in vesicular targeting and protein transport. More recent experiments have shown that it is also a component in the presenilin complex and modulates the gamma-secretase but not the epsilon-secretase cleavage activity of the amyloid precursor protein. The presenilin complex is composed of the proteins APH1, nicastrin, and PEN2 in addition to presenilin-1. Together, these proteins cleave the amyloid precursor protein at what is known as the gamma- and epsilon-sites and can lead to the accumulation of the Abeta cleavage product that is associated with Alzheimer's disease. Co-immunoprecipitation experiments using antibodies against these proteins also yielded TMP21 indicating that TMP21 may play a role in the regulation of this complex. Suppression of TMP21 expression by siRNA in transfected cells caused increased gamma-secretase activity but not epsilon-secretase activity, and increased Abeta; production, demonstrating that TMP21 can modulate gamma-secretase activity.

APH1 Antibody: APH1 was initially identified as a component of the Notch pathway in C. elegans. Along with nicastrin, PEN2, and presenilin-1 APH1 is an essential component of the gamma-secretase complex which cleave the amyloid precursor protein (APP) at what are known as the gamma- and epsilon-sites and can lead to the accumulation of the Amyloid beta peptide (Abeta) cleavage product that is associated with Alzheimer's disease. APH1 exists in at least three distinct isoforms with APH1a as the principal isoform present in the gamma-secretase complex. Mice deficient in this isoform, but not the other two, were lethal at E10.5, with impaired vascular and neural development observed.

Stem cell factor (SCF), also known as cKit ligand (KL), mast cell growth factor (MGF), and steel factor (SLF), is a widely expressed 28-40 kDa type I transmembrane glycoprotein. It promotes the survival, differentiation, and mobilisaion of multiple cell types including myeloid, erythroid, megakaryocytic, lymphoid, germ cell, and melanocyte progenitors. SCF is a primary growth and activation factor for mast cells and eosinophils. Noncovalent dimers of transmembrane or soluble SCF interact with the receptor tyrosine kinase SCF R/cKit to trigger receptor dimerisaion and signalling. SCF assists in the recovery of cardiac function following myocardial infarction by increasing the number of cardiomyocytes and vascular channels.
