AHNAK is a 5,890 amino acid protein encoded by the human gene AHNAK. The intronless AHNAK gene is located on human chromosome 11q12 and has three main structural regions: the 251 amino acid N-terminus, a large central region of 4390 amino acids with multiple repeated units of about 128 amino acids in length, and the 1002 amino acid C-terminus. The central region seems to have antiparallel beta-strands connected by intervening loops. Several putative regulatory elements are clustered within the C-terminal region, including nuclear export localization signals, a leucine zipper, and potential phosphorylation sites for Akt1 and PKC. AHNAK is believed to be an important signalling molecule involved in a wide range of physiological activities and may be required for neuronal cell differentiation.. AHNAK also appears to influence b-adrenergic regulation of cardiac L-type Ca2+ channel function.

GPRC6A (G protein-coupled receptor family C group 6 member A) is a 928 amino acid protein encoded by the mouse Gprc6a gene. GPRC6A is a member of the G protein-coupled receptor 3 family. This receptor is activated by both amino acids and extracellular concentration of calcium ions. The activity of GPRC6A is mediated by a G protein that activates a phosphatidylinositol-calcium second messenger system. GPRC6A senses changes in the extracellular concentration of calcium ions, suggesting that it may mediate extracellular calcium-sensing responses in osteoblasts. Osteocalin stimulates the activity of GPRC6A in the presence of calcium. GPRC6A also acts as a receptor for amino acids, with a preference for basic amino acids such as L-Lys, L-Arg and L-ornithine. Its affinity for amino acids suggests that it may act as a regulatory component of the urea cycle. GPRC6A is expressed at high levels in liver, lung, spleen and heart, and at lower levels in kidney, skeletal muscle and brain.

AMPK is activated by high AMP and low ATP through a mechanism involving allosteric regulation, promotion of phosphorylation by an upstream protein kinase known as AMPK kinase and inhibition of dephosphorylation. Activated AMPK can phosphorylate and regulate in vivo hydroxy-methylglutaryl-CoA reductase and acetyl-CoA carboxylase, which are key regulatory enzymes of sterol synthesis and fatty acid synthesis, respectively. The human AMPKa1 and AMPKa2 genes encode 548 amino acid and 552 amino acid proteins, respectively. Human AMPKb1 encodes a 271 amino acid protein and human AMPKb2 encodes a 272 amino acid protein. The human AMPKg1 gene encodes a 331 amino acid protein.
