FRMD5 (FERM domain-containing protein 5) is a 570 amino acid single-pass membrane protein that contains one FERM domain and exists as two alternatively spliced isoforms. The gene encoding FRMD5 maps to human chromosome 15, which houses over 700 genes and comprises nearly 3% of the human genome. Angelman syndrome, Prader-Willi syndrome, Tay-Sachs disease and Marfan syndrome are all associated with defects in chromosome 15-localized genes.

The family of guanylin regulatory peptides, including guanylin and uroguanylin, are strongly expressed in intestinal mucosa and regulate intestinal fluid secretion during digestion. Guanylins are also involved in acid neutralization and the regulation of membrane-bound guanylate cyclase Signalling molecules. Guanylin and uroguanylin are secreted primarily in the stomach, intestine, and colon. Guanylin is also detected in plasma. Guanylin is an endogenous activator of intestinal guanylate cyclase. It stimulates intestinal guanylate cyclase through the same receptor binding region as the heat-stable enterotoxins. Gut enterochromaffin cells synthesize guanylin to be a prohormone of 115 amino acids which is then is processed to the molecular form of 94 amino acids. This 10 kDa form is found circulating in the blood.

The 3-HAO family of proteins consists of intramolecular dioxygenases that contain non-heme ferrous iron. HAAO (3-hydroxyanthranilate 3,4-dioxygenase), also known as HAO or 3-HAO, is a 286 amino acid monomeric cytoplasmic protein that belongs to the 3-HAO family. Utilizing Fe²⁺ ions as a cofactor, HAAO participates in the formation of quinolinic acid (QUIN), and excitotoxin, by catalyzing the oxidative ring opening of 3-hydroxyanthranilate to 2-amino-3-carboxymuconate semialdehyde. HAAO may play a role in disorders that are associated with varying levels of QUIN expression, and is widely expressed in peripheral organs, including kidney and liver. Existing as two alternatively spliced isoforms, the gene encoding HAAO maps to human chromosome 2p21 and mouse chromosome 17 E4.

The two ubiquitin C-terminal hydrolase (UCH) enzymes, UCHL1 and UCHL3, deubiquitinate ubiquitin-protein conjugates and control the cellular balance of ubiquitin. UCHL1 and UCHL3 are both small proteins of ~220 amino acids that share more than 40% amino acid sequence identity. UCHL3 is universally expressed in all tissues, while UCHL1 is expressed exclusively in neuronal tissue, testis and ovary. The activity of UCHL3 is more than 200 fold higher than UCHL1 when a fluorogenic ubiquitin substrate is used. UCHL1 associates with monoubiquitin and UCHL3 binds to Nedd8, ubiquitin-like protein. UCHL1 and UCHL3 play a role in the regulation of neuronal development and spermatogenesis. UCHL1 is involved in the pathogenesis of Parkinson’s disease (PD) and Alzheimer’s disease (AD).

Predominantly localized to the nucleolus, BOP1 (Block of proliferation 1 protein) is a 746 amino acid highly conserved non-ribosomal protein that is involved in ribosome biogenesis. Truncation of the amino terminus of BOP1 leads to cell growth arrest in the G1 phase and specific inhibition of 28S and 5.8S rRNA synthesis, as well as a deficit in the cytosolic 60S ribosomal subunit. This suggests that BOP1 is involved in the formation of mature rRNAs and in the biogenesis of the 60S ribosomal subunit. BOP1 physically interacts with pescadillo (a protein involved in cell proliferation) and enables efficient incorporation of pescadillo into the nucleolar preribosomal complexes, thereby affecting rRNA maturation and the cell cycle. The BOP1-pescadillo complex is also necessary for biogenesis of 60S ribosomal subunits. Deregulation of BOP1 may lead to colorectal tumorigenesis.

This antibody recognizes a polypeptide which is identified as insulin, a 51-amino acid polypeptide composed of A and B chains connected through the C-peptide. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Deficiency of insulin results in diabetes mellitus. The main storage site for insulin is the pancreatic islets. Antibodies to insulin are important as beta-cell and insulinoma marker.
