Beschreibung:   Pancreatic polypeptide (PP), neuropeptide Y (NPY), and peptide YY (PYY) are related 36-amino acid hormones. A number of structurally related receptors for these peptides have been isolated, NPY1-R, NPY2-R, NPY3-R, NPY4-R, NPY5-R, and NPY6-R. NPY4-R is expressed in several human tissues, including brain, coronary artery, and ileum. NPY4-R maps to human chromosome 10q11.2. NPY-5R, isolated from rat hypothalamus, encodes a 456-amino acid protein with less than 35% overall identity to known Y-type receptors. The human NPY5-R sequence is nearly identical to, but in the opposite orientation from, that of the human NPY1-R sequence. NPY5-R localizes to the paraventricular hypothalamic nucleus, the lateral hypothalamus, and other locations consistent with a role in the control of feeding behavior. The gene which encodes NPY5-R maps to human chromosome 4q32.2. NPY6-R is abundantly expressed in human heart and skeletal muscle and the gene which encodes NPY6-R maps to human chromosome 5q31.

Beschreibung:   The mammalian FXYD family maintains Na+ and K+ gradients between the intracellular and extracellular milieus of cells in processes such as renal Na+-reabsorption, muscle contraction and neuronal excitability. FXYDs are single-span membrane proteins that share a 35 amino acid signature domain, beginning with the sequence PFXYD and containing seven invariant and six conserved amino acids. Members of the FXYD family include FXYD1 (PLM, phospholemman), FXYD2 (the g subunit of the Na+/K+-ATPase), FXYD3 (Mat8, mammary tumor protein), FXYD4 (CHIF) and FXYD5 (RIC). FXYD6 is expressed in various epithelial cells bordering the endolymph space and in the auditory neurons. FXYD6 co-localizes with Na+/K+-ATPase in the stria vascularis and can be co-immunoprecipitated with Na+/K+-ATPase. After expression, FXYD6 associates with Na+/K+-ATPase alpha1-beta1 and alpha1-beta2 isozymes, which are preferentially expressed in different regions of the inner ear and also with gastric and non-gastric H+/K+-ATPase.

Beschreibung:   The mammalian homologues of the key cell death gene CED 4 in C. elegans has been identified recently from human and mouse and designated Apaf1 (for apoptosis protease activating factor 1). Apaf1 binds to cytochrome c (Apaf2) and caspase 9 (Apaf3), which leads to caspase 9 activation. Activated caspase 9 in turn cleaves and activates caspase 3 that is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis. A new Apaf1 Interacting Protein (APIP) also known as CG129 and MMRP19, has been identified as a negative regulator of ischemic injury. APIP competes with Caspase 9 binding site of Apaf1. APIP is predicted to code for a 204 amino acid. An isoform of APIP, APIP2 encodes a 242 amino acid protein, which is an alternative splicing variant differing in its N terminus from APIP. APIP transcript is ubiquitously expressed in most adult tissue with high expression in skeletal muscle, heart, and kidney.

Beschreibung:   Kelch domain-containing protein 8A (KLHDC8A) is a 350 amino acid protein. KLHDC8A contains seven kelch repeats, each of which is an approximately 50 amino acid long conserved region that forms a tertiary structure beta-propeller. The gene that encodes KLHDC8A is located on chromosome 1, which is the largest human chromosome, spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes Lamin A. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma.

Beschreibung:   Amines, including those present on proteins, spontaneously react with glucose to make fructosamines in a reaction termed glycation. Fructosamine 3-kinase (FN3K), a 309-amino acid enzyme initially identified in erythrocytes, catalyzes the ATP-dependent phosphorylation of the third carbon on both D- and L-fructosamines, leading to their destabilization and eventually, their removal from the protein. FN3K is a monomer that is ubiquitously expressed in mammalian tissue and phosphorylates both low molecular mass and protein-bound fructosamines which are formed as a result of glycation of glucose with primary amines. FN3K protects proteins from the harmful effects of nonenzymatic glycation, and may also be involved in peptide repair and cell metabolism. FN3KRP (fructosamine-3-kinase-related protein) is a 309 amino acid protein that is expressed in erythrocytes, bone marrow, spleen, brain and kidney and belongs to the fructosamine kinase family. FN3KRP functions to phosphorylate psicoamines and ribulosamines on the third carbon of their sugar moiety, thereby leading to the deglycation of the target amines.

Beschreibung:   The nuclear envelope separates the nucleoplasm from the cytoplasm in eukaryotic cells and includes the outer and inner nuclear membrane, nuclear pore complexes and the nuclear lamina. The nuclear lamina contains intermediate filament-type proteins called lamins that form a dense network to strengthen and stabilize the nuclear envelope. Lamina-associated polypeptide 2 (LAP2) is also known as thymopoietin. LAP2 is a nuclear envelope protein and contains an amino-terminal region called the LAP2-emerin-MAN1 or LEM motif. LAP2 also contains a unique DNA-binding amino-terminal domain. Alternative splicing produces six isoforms (, _,_, and ) of mammalian LAP2 and three isoforms in Xenopus LAP2. LAP2 and LAP2_ associate with chromosomal barrier-to-autointegration factor (BAF) and may play a role in stabilizing chromatin structure. LAP2_ also binds to lamin B. LAP2 is a non-membrane isoform of LAP2 that associates with the internal nucleoskeleton and binds lamin A. The gene encoding human LAP2 maps to chromosome 12q23.1.

Beschreibung:   Gamma-glutamyltranspeptidase (GGT) acts as a glutathionase and catalyzes the transfer of the glutamyl moiety of glutathione to a variety of amino acids and dipeptide acceptors. This enzyme is located on the outer surface of the cell membrane and is widely distributed in mammalian tissues involved in absorption and secretion. In humans, hepatic GGT activity is elevated in some liver diseases. GGT1 is released into the bloodstream after liver damage, and an elevated level of the enzyme may be a useful early sign of hepatocellular carcinoma. GGT5 converts leukotriene C4 to leukotriene D4; it does not, however, convert synthetic substrates that are commonly used to assay GGT. In human serum and in human tissues, there is a marked heterogeneity in GGT, but this heterogeneity can be attributed to different glycosylation of the same peptide rather than to the products of different genes. Belonging to the gamma-glutamyltranspeptidase family, GGT6 (gamma-glutamyltransferase 6), is a 493 amino acid single-pass type II membrane protein that cleaves glutathione conjugates. GGT6 exists as two alternatively spliced isoforms.

Beschreibung:   The MARVEL domain is a 130 amino acid motif that contains four transmembrane helices, both of which have cytoplasmic N- and C-terminal regions. MARVEL domain-containing proteins are thought to participate in tight junction regulation, the biogenesis of vesicular transport carriers and in cholesterol-rich membrane apposition events. Pantophysin, also known as SYPL1 (synaptophysin-like protein 1) or H-SP1, is a 259 amino acid multi-pass membrane protein that localizes to melanosomes and vesicles, as well as to the cytoplasm, and contains one MARVEL domain. Expressed as multiple alternatively spliced isoforms, pantophysin is present in tissues throughout the body where it may play a role in vesicle trafficking and protein transport. The gene encoding pantophysin maps to human chromosome 7, which houses over 1,000 genes and comprises nearly 5% of the human genome. Defects in some of the genes localized to chromosome 7 have been linked to Osteogenesis imperfecta, Williams-Beuren syndrome, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome.
