All collagens contain a triple helix domain and frequently show lateral self-association in order to form complex connective tissues. The Collagen Type XXV precursor produces the proteolytic fragment CLAC (collagenous Alzheimer amyloid plaque component), which deposits in senile plaques associated with amyloid beta peptides (Abeta) in the brains of patients with Alzheimer's disease. CLAC binds to the fibrillized form of Abeta, which disturbs the structure and function of plasma membranes.

Alternative splicing of this gene results in three tissue-specific forms of glucokinase, one found in pancreatic islet beta cells and two found in liver. The protein localises to the outer membrane of mitochondria. In contrast to other forms of hexokinase, this enzyme is not inhibited by its product glucose-6-phosphate but remains active while glucose is abundant. Mutations in this gene have been associated with non-insulin dependent diabetes mellitus (NIDDM), maturity onset diabetes of the young, type 2 (MODY2) and persistent hyperinsulinemic hypoglycemia of infancy (PHHI).

HGFA Inhibitor 1 is an endogenous inhibitor of the kunitz-type serine proteinase HGF-activator (HGFA). HGF was first described as a hepatocyte-specific mitogen and survival factor, and has since been shown to exert a variety of actions on many cell types by binding to its MET receptor. HGF is activated by cleavage of the single-chain form to form a two-chain version by HGFA. HGFA Inhibitor 1 also inhibits trypsin, and MTSP-1 (Matriptase), a serine proteinase that also activates HGF. HGFB Inhibitor 1 is a splice variant of HGFA Inhibitor 1, and contains a 16 amino acid insert after the first kringle domain, relative to the HGFA Inhibitor 1 sequence.

MTERF (mitochondrial transcription termination factor 1) is composed of 399 amino acids and contains three leucine zippers that form a three-stranded coiled-coil that binds to DNA. It has been suggested that only the phosphorylated form of MTERF has transcription termination activity. MTERFD1 is also thought to act as a mitochondrial transcription regulator and is expressed as two isoforms produced by alternative splicing. MTERFD3 is believed to be involved in cell cycle regulation and cell growth by modulating mitochondrial transcription. MTERFD3 is expressed in heart, skeletal muscle, pancreas and liver.
