Beschreibung:   VAV2 is a ubiquitously expressed structural homolog of the VAV protooncogene that is expressed preferentially in hematopoetic cells. Both proteins are comprised of a Dbl homology (DH) domain with guanosine nucleotide exchange (GEF) activity exclusively directed towards Rho/Rac GTPases, a pleckstrin homology (PH) domain, a calponin-homology (CH) region, an acidic domain (AD) a zinc finger butterfly motif, two SH3 regions and one SH2 domain. GEF activity of RhoA family G proteins is induced by tyrosine phosphorylation in wild type VAV2, and is constitutively activated in N terminus deleted oncogene forms. Constitutive expression of a VAV2 oncoprotein may result in morphological alterations including highly enlarged cells in which karyokinesis and cytokinesis frequently are uncoupled.

Beschreibung:   MPDZ is a 2042 amino acid peripheral membrane protein that colocalizes with SR-2C on the apical membrane of epithelial choroid plexus cells. Expressed in heart, brain, placenta, liver, skeletal muscle, kidney and pancreas, MPDZ causes clustering of SR-2C, a serotonin receptor, at the cell surface. MPDZ is member of the NMDAR Signalling complex that is involved in regulating AMPAR potentiation and synaptic plasticity in excitatory synapses. As a tight junction protein in epithelial cells, MPDZ interacts with SSTR3, a G-protein-coupled receptor, to regulate transepithelial permeability in a pertussis toxin sensitive manner. MPDZ, along with Kir4.2, may form a complex with other proteins in the nephron and regulate ion transport. MPDZ contains one L27 domain and thirteen PDZ domains.

Beschreibung:   B-ATF is a nuclear basic leucine zipper protein that belongs to the AP-1/ATF superfamily of transcription factors. The leucine zipper of B-ATF mediates dimerization with members of the Jun family of proteins. The B-ATF protein does not homodimerize efficiently, but rather forms a heterodimer preferentially with c-Jun. The B-ATF/c-Jun protein complex can interact with DNA containing a consensus binding site for AP-1, suggesting that B-ATF functions as a tissue-specific modulator of the AP-1 transcription complex in human cells. B-ATF also associates with IFP35, a leucine zipper protein that translocates to the nucleus following IFN treatment. The gene encoding B-ATF, also designated SFA-2, is strongly expressed in mature T and B lymphocytes, and is up-regulated after transformation by human T-cell leukemia virus type I.

Beschreibung:   14-3-3 proteins regulate many cellular processes relevant to cancer biology, notably apoptosis, mitogenic signaling and cell-cycle checkpoints. Seven isoforms comprise this family of signaling intermediates, denoted 14-3-3 b, g, e, z, h, q and s. 14-3-3 proteins form dimers that present two binding sites for ligand proteins, thereby bringing together two proteins that may not otherwise associate. These ligands largely share a 14-3-3 consensus binding motif and exhibit serine/threonine phosphorylation. 14-3-3 proteins function in broad regulation of these ligand proteins; by cytoplasmic sequestration, occupation of interaction domains and import/export sequences, prevention of degradation, activation/repression of enzymatic activity, and facilitation of protein modification. Loss of expression contributes to a vast array of pathogenic cellular activities.

Beschreibung:   Zinc-finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. ZDHHC20 (zinc finger, DHHC-type containing 20) is a 365 amino acid multi-pass membrane protein that contains one DHHC-type zinc finger and is thought to function as a palmitoyltransferase, catalyzing the transformation of palmitoyl-CoA and a cysteine-conjugated protein to an S-palmitoyl protein and free CoA. In response to DNA damage, ZDHHC20, which exists as multiple alternatively spliced isoforms, may be phosphorylated by ATM or ATR. The gene encoding ZDHHC20 maps to human chromosome 13, which houses over 400 genes, such as BRCA2 and RB1, and comprises nearly 4% of the human genome.

Beschreibung:   This gene encodes the acidic form of complement factor 4, part of the classical activation pathway. The protein is expressed as a single chain precursor which is proteolytically cleaved into a trimer of alpha, beta, and gamma chains prior to secretion. The trimer provides a surface for interaction between the antigen-antibody complex and other complement components. The alpha chain may be cleaved to release C4 anaphylatoxin, a mediator of local inflammation. Deficiency of this protein is associated with systemic lupus erythematosus and type I diabetes mellitus. This gene localizes to the major histocompatibility complex (MHC) class III region on chromosome 6. Varying haplotypes of this gene cluster exist, such that individuals may have 1, 2, or 3 copies of this gene. Two transcript variants encoding different isoforms have been found for this gene.

Beschreibung:   Enzymes known as phosphatidylserine decarboxylases (PSDs) catalyze the formation of phosphatidylethanolamine from phosphatidylserine via phosphatidylserine decarboxylation. Type I PSDs contain LGST motifs and are found in bacteria and eukaryotic mitochondria, whereas type II PSDs contain GGST motifs and are found in eukaryotic endomembrane systems. PISD (phosphatidylserine decarboxylase), also known as phosphatidylserine decarboxylase proenzyme, PSDC, PSD, PSSC, DJ858B16, dJ858B16.2 or DKFZp566G2246, is a 408 amino acid a type I phosphatidylserine decarboxylase that localizes to the inner mitochondrial membrane. PISD contains a conserved LGST motif which is cleaved to produce two isoforms known as PISD α and PISD β. PISD is capable of forming a heterodimer and is highly expressed in liver and testis. The gene encoding PISD maps to human chromosome 22q12.2.

Beschreibung:   The MEL-14 antibody is specific for mouse CD62L, also known as L-Selectin, a cell adhesion molecule which facilitates lymphocyte “rolling” on activated vascular endothelium and homing to high endothelial venules (HEV) as immune cells transmigrate from blood into peripheral tissues. L-Selectin is a member of a family of Selectin molecules which act together with the integrin family of adhesion molecules to mediate leukocyte-endothelial interactions. L-Selectin is characteristically expressed by neutrophils, and is also found on B cells, monocytes, granulocytes, and at varying levels on naive, effector and memory T cells. It is rapidly shed upon cell activation, releasing into the circulation a soluble form whose biological role is of particular interest in cancer biology research.

Beschreibung:   Best vitelliform macular dystrophy, known as Best disease, is an early-onset autosomal dominant condition in which accumulation of lipofuscin-like material within and beneath the RPE leads to progressive loss of central vision. Best disease is frequently a reflection of mutations in the Bestrophin gene, which encodes a protein containing four putative transmembrane domains and localizes to the basolateral plasma membrane of RPE cells. Human Bestrophin forms oligomeric chloride channels that are sensitive to intracellular calcium. Missense mutations at the Bestrophin locus reduces or abolishes Bestrophin protein mediated membrane current. Bestrophin Bestrophin 2,Bestrophin 3, and Bestrophin 4 are transmembrane proteins that contain a high percentage of aromatic residues, a conserved RFP (Arg-Phe-Pro) motif and they function as anion channels.

Beschreibung:   Made up of nearly 146 million bases, chromosome 8 encodes about 800 genes. Translocation of portions of chromosome 8 with amplifications of the c-Myc gene are found in some leukemias and lymphomas, and typically associated with a poor prognosis. Portions of chromosome 8 have been linked to schizophrenia and bipolar disorder. Trisomy 8, also known as Warkany syndrome 2, most often results in early miscarriage but is occasionally seen in a mosaic form in surviving patients who suffer to a varying degree from a number of symptoms including retarded mental and motor development, and certain facial and developmental defects. WRN is a DNA helicase encoded by chromosome 8 and shown defective in those with the early aging disorder Werner syndrome. Chromosome 8 is also associated with Pfeiffer syndrome, congenital hypothyroidism and Waardenburg syndrome.

Beschreibung:   May mediate activation of CASP1 via ASC and promote activation of NF-kappa-B via IKK.NALP proteins are cytoplasmic proteins that form a subfamily within the larger CATERPILLER family and are thought to play a crucial role in cell proliferation and reproduction. Like all other NALP family members, NALP12, also known as Monarch-1, has a C-terminal leucine-rich repeat (LRR) region, an N-terminal Pyrin domain (PYD) followed by a NACHT domain, and a NACHT-associated domain. NALP12 is thought to act as an attenuating factor of inflammation by suppressing inflammatory responses such as NF-kB activation by TLR-signaling molecules MyD88, IRAK-1, TRAF6 and RIPK1 in activated monocytes. Recent evidence suggests that mutations in NALP12 result in hereditary periodic fever syndromes.

Beschreibung:   The epitope of this MAb maps in between aa 1-34. PTH is a hormone produced by the parathyroid gland that regulates the concentration of calcium and phosphorus in extracellular fluid. This hormone elevates blood Ca2 levels by dissolving the salts in bone and preventing their renal excretion. It is produced in the parathyroid gland as an 84 amino acid single chain polypeptide. It can also be secreted as N-terminal truncated fragments or C-terminal fragments after intracellular degradation, as in case of hypercalcemia. Defects in this gene are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism.
