Mono-ADP-ribosylation is one of the posttranslational protein modifications regulating cellular metabolism (e.g. nitrogen fixation) in prokaryotes. Mono-ADP-ribosylation is a process in which the ADP-ribose moiety of nicotinamide adenine dinucleotide is transferred to an acceptor amino acid. Five mammalian ADP-ribosyltransferases (ART1-ART5) have been cloned, and each ART is expressed in different tissues. ART5 (ADP-ribosyltransferase 5), also known as Ecto-ADP-ribosyltransferase 5, is a 292 amino acid secretory protein that is expressed in testis, heart, skeletal muscle and lymphoma. Functionally, ART5 is implicatetd to play a role in cell signaling and metabolism cascades. Two isoforms of ART5 exist as a result of alternative splicing events.

GMEB-1 is a 573 amino acid protein that contains one SAND domain and is a member of the KDWK family of combinatorial transcription modulators. Localized to both the cytoplasm and the nucleus, GMEB-1 forms a heterodimer with GMEB-2 (Glucocorticoid modulatory element-binding protein 2) and, once associated with GMEB-2, plays a key role in parvovirus DNA replication. In addition, GMEB-1 functions alone as a trans-acting factor that, by binding to glucocorticoid modulatory elements (GMEs) in TAT (tyrosine aminotransferase) promoters, increases intracellular sensitivity to glucocorticoid concentrations. GMEB-1 also interacts with initiator procaspases and, via this interaction, can inhibit caspase-induced apoptosis. Due to alternative splicing events, GMEB-1 is expressed as two isoforms.

Recognizes the epsilon-chain of CD3 (Workshop V; Code: CD03.09), which consists of five different polypeptide chains (designated as γ, δ, ε, ζ, and η) with MW ranging from 16-28 kDa. The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. In cortical thymocytes, CD3 is predominantly intra-cytoplasmic. However, in medullary thymocytes, it appears on the T cell surface. CD3 antigen is a highly specific marker for T cells, and is present in majority of T cell neoplasms.

PIRT is a 137 amino acid multi-pass membrane protein. Highly conserved among vertebrates, PIRT consists of two transmembrane domains and one putative C-terminal phosphoinositide-binding domain. Although PIRT is expressed in peripheral nervous system, with highest levels in dorsal root ganglion and trigeminal neurons, and lowest levels in sympathetic and enteric neurons, it is not expressed in spinal cord. PIRT is a required component of the VR1 complex, which positively regulates VR1, a sensor of both noxious heat and capsaicin. Correspondingly, PIRT knockout results in impaired responses to noxious heat and capsaicin exposure, while VR1 remains unaltered. The gene that encodes PIRT maps to human chromosome 17p13.1.

Tuberous sclerosis complex (TSC) is an autosomal dominant genetic disorder characterised by mental retardation and the widespread development of distinctive tumors termed hamartomas. Two different genetic loci have been linked to TSC; one of these loci, the tuberous sclerosis-2 gene (TSC2), encodes a protein called tuberin and the other loci, tuberous sclerosis-1 gene (TSC1), encodes a protein called hamartin. Tuberin and hamartin interact with each other forming a cystoplasmic complex. Hamartin interacts with the ezrin-radixin-moesin (ERM) family of actin-binding proteins and inhibition of hamartin activity results in loss of cell adhesion. Hamartin is present in most adult tissues with strong expression in brain, heart, and kidney.

Copine 6 contains two N-terminal C2 domains and one C-terminal VWFA (von Willebrand factor A) domain, which is also referred to as the A domain or the core domain. As is characteristic of the copine family, copine 6 functions in membrane trafficking and is capable of binding phospholipids in a calcium-dependent manner. Copine 6 may also play a role in synaptic plasticity.
