Interleukin-21 (IL-21) is a key factor in the transition between innate and adaptive immune responses secreted by activated T cells. The IL-21 receptor (IL-21R) is expressed in lymphoid tissue, in particular by NK, B, T and dendritic cells, macrophages and endothelial cells. Recent evidence suggests that IL-21 plays a supportive role in the proliferation of T and B cells and influences the cytolytic activity of natural killer cells. IL-21 has been shown to up-regulate genes associated with innate immunity and to inhibit the differentiation of naive T helper cells. IL-21 specifically inhibits IFN-gamma production from developing TH1 cells and is preferentially expressed by TH2 cells. Furthermore IL-21 has been identified as a growth and survival factor for human myeloma cells. IL-21/IL-21R interactions have a unique role in sequentially activating both innate and adaptive immune responses against poorly immunogenic tumours, leading to tumour rejection that is perforin dependent but IFN-gamma independent.

Trace amines are endogenous molecules structurally related to classical biogenic amines that are linked to psychiatric conditions. A family of G-protein coupled receptors referred to as trace-amine-associated receptors (TAAR) are activated by trace amines and are present in very low levels in mammalian tissue. TAAR’s contain several structural features that are similar to the rhodopsin beta-adrenergic receptor superfamily, including the positions of the seven transmembrane regions that provide common ligand-binding pockets as well as the short N- and C-terminal domains. TAAR proteins are potential targets for drugs of abuse, such as amphetamine and MDMA, as well as neuropsychiatric disorders including schizophrenia, depression, and attention deficit disorder. TAAR-1 is a 340 amino acid protein that increases intracellular cAMP accumulation in response to beta-phenylethylamine and tyramine. TAAR-1 is associated with the detection of social cues, illustrating its significance as a therapeutic target.

Stimulates cell cycle progression at the G1-S and G2-M transitions by phosphorylation of CDC25A and CDC25C. Interleukin-21 receptor (IL-21R) is a type I transmembrane glycoprotein within the class I cytokine receptor family, type 4 subfamily. Complex formation between IL-21R and the common gamma chain (gammac) is required for signalling. IL-21R is expressed mainly on B cells (highest on mature, activated, follicular and germinal center B cells), NK cells and activated T cells, but is also found on dendritic cells, alternatively activated macrophages, intestinal lamina propria fibroblasts and epithelial cells and keratinocytes. B cell IL-21R engagement induces Blimp1 (which mediates plasma cell differentiation) and is important for memory responses. IL-21R engagement on mouse NK cells enhances their cytotoxic activity and IFN-gamma production. IL-21R engagement on CD8+ T cells aids control of viral infection and tumour growth; IL-21R is also necessary for sufficient numbers of regulatory T cells to combat chronic inflammation. IL-21R expression is often upregulated in allergic skin inflammation, systemic lupus erythematosus and diffuse large B cell lymphoma (DLBCL).
