Beschreibung:   The mouse monoclonal antibody recognizes CD1b, a 44 kDa type I glycoprotein associated with beta2-microglobulin. It is expressed on dendritic cells, Langerhans cells, thymocytes, and T acute lymphoblastic leukemia cells. The CD1 multigene family encodes five forms of the CD1 T-cell surface glycoprotein in human, designated CD1A, 1B, 1C, 1D and 1E. CD1, a type 1 membrane protein, has structural similarity to the MHC class I antigen and has been shown to present lipid antigens for recognition by T lymphocytes. Constitutive endocytosis of CD1B molecules and the differential sorting of MHC class II from lysosomes separate peptide- and lipid antigen-presenting molecules during dendritic cell maturation. CD1B is also expressed in interdigitating cells.

Beschreibung:   Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). CK8 is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Hepatocellular carcinomas are defined by the use of antibodies that recognize only cytokeratin 8 and 18. CK8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Anti-CK8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against CK8, CK18 and CK19). Anti-CK8 is useful for the differentiation of lobular (ring-like, perinuclear) from ductal (peripheral-predominant) carcinoma of the breast.

Beschreibung:   Histone methyltransferase that specifically monomethylates 'Lys-4' of histone H3. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation. Plays a central role in the transcriptional activation of genes such as collagenase or insulin. Recruited by IPF1/PDX-1 to the insulin promoter, leading to activate transcription. Has also methyltransferase activity toward non-histone proteins such as p53/TP53, TAF10, and possibly TAF7 by recognizing and binding the [KR]-[STA]-K in substrate proteins. Monomethylates 'Lys-189' of TAF10, leading to increase the affinity of TAF10 for RNA polymerase II. Monomethylates 'Lys-372' of p53/TP53, stabilizing p53/TP53 and increasing p53/TP53-mediated transcriptional activation. Also able to demethylated 'Lys-372' of p53/TP53 in vitro.

Beschreibung:   Complete remission of acute promyelocytic leukemia can be achieved by treating patients with retinoic acid, and PML-RAR-a (promyelocytic leukemia-retinoic acid receptor alpha fusion protein) plays a major role in mediating retinoic acid effects in leukemia cells. The retinoic acid-induced gene, PRAM-1 (PML-RAR-a target gene encoding an adaptor molecule 1) encodes an adaptor protein which is expressed and modulated during normal human myelopoiesis. PRAM-1 expression is hindered by expression of PML-RAR-a. The 718 amino acid PRAM-1 protein contains eight N-terminal proline-rich repeats and several proline residues that are clustered as type I or type II SH3 recognition motifs. PRAM-1 demonstrates expression in hematopoietic tissues and lung.

Beschreibung:   HDAC7 is a member of the class II mammalian histone deacetylases, which plays an important role in modulating the eukaryotic chromatin structure. Human HDAC7 is composed of 912 amino acid residues. Although HDAC7 is localized mostly to the cell nucleus, it is also found in the cytoplasm, suggesting nucleo-cytoplasmic shuttling. The histone deacetylase activity of HDAC7 maps to a carboxy-terminal domain and is dependent on interaction with class I HDACs in the nucleus. It is an active component of different transcriptional corepressor complexes that can be recruited to specific promoter regions via interactions with a growing number of sequence specific transcriptional factors. HDAC7 catalyzes removal of acetyl-groups from acetyl-lysines of histones and promotes compaction of chromatin in these regions, leading to the inhibition of gene transcription.

Beschreibung:   The hydroxyacylglutathione hydrolase-like protein (HAGHL) is a 290 amino acid protein that belongs to the glyoxalase II family. HAGHL binds two zinc ions per subunit and acts as a hydrolase on ester bonds. The gene encoding HAGHL maps to human chromosome 16, which encodes over 900 genes in approximately 90 million base pairs, makes up nearly 3% of human cellular DNA and is associated with a variety of genetic disorders. The rare disorder Rubinstein-Taybi syndrome is associated with chromosome 16, as is Crohn's disease, a gastrointestinal inflammatory condition that may involve the NOD2 gene. An association with systemic lupus erythematosis and a number of other autoimmune disorders with the pericentromeric region of chromosome 16 has led to the identification of SLC5A11 as a potential autoimmune modifier.

Beschreibung:   Defects in G6PD are the cause of chronic non-spherocytic hemolytic anaemia (CNSHA) . Deficiency of G6PD is associated with hemolytic anaemia in two different situations. First, in areas in which malaria has been endemic, G6PD-deficiency alleles have reached high frequencies (1% to 50%) and deficient individuals, though essentially asymptomatic in the steady state, have a high risk of acute hemolytic attacks. Secondly, sporadic cases of G6PD deficiency occur at a very low frequencies, and they usually present a more severe phenotype. Several types of CNSHA are recognized. Class-I variants are associated with severe NSHA; class-II have an activity <10% of normal; class-III have an activity of 10% to 60% of normal; class-IV have near normal activity.

Beschreibung:   Recognizes a protein of 33-55 kDa, identified as CD53 (Workshop V; Code CD53.1). CD53 is expressed on monocytes, and macrophages, granulocytes, dendritic cells, osteoblasts and osteoclasts, NK cells, and on T- and B-cells from every stage of differentiation but is absent from platelets, erythrocytes, and non-haemopoietic cells. CD53 is a member of a family of tetraspan transmembrane proteins, including CD9, CD37, CD63, CD81, and CD82. It associates with integrins, MHC class II molecules, and a tyrosine phosphatase and plays a role in cellular activation as part of a signal transduction complex involving other membrane glycoproteins. Defects of CD53 expression on neutrophils appear to be related with recurrent infectious diseases. Cross-linking CD53 using CD53 antibodies led to cytoplasmic calcium fluxes in B cells, monocytes, and granulocytes and activation of the monocyte oxidative burst.

Beschreibung:   CNOT2 (CCR4-NOT transcription complex subunit 2) is a ubiquitous protein encoded by the human gene CNOT2. CNOT2 belongs to the CNOT2/3/5 family and is part of the CCR4-NOT complex. The CCR4-NOT complex is an evolutionarily conserved, multi-component complex known to be involved in transcription as well as mRNA degradation. Various subunits (e.g. CNOT1, CNOT2) are involved in influencing nuclear hormone receptor activities. The CCR4-NOT complex is also involved in the regulation of Histone H3 lysine 4 methylation through a ubiquitin-dependent pathway that likely involves the proteasome. Increased expression of the CNOT2 subunit acts to strongly repress transcription by RNA polymerase II. This repressive effect is mediated by a conserved NOT-Box, which is located at the C-terminus of CNOT2 proteins. Repression by the NOT-Box is sensitive to treatment with the histone deacetylase (HDAC) inhibitor trichostatin A.

Beschreibung:   GLT8D2 (glycosyltransferase 8 domain-containing protein 2), also known as GALA4A, is a 349 amino acid single-pass type II membrane protein. A member of the glycosyltransferase 8 family, GLT8D2 is encoded by a gene that maps to human chromosome 12q23.3. Encoding over 1,100 genes within 132 million base pairs, chromosome 12 makes up about 4.5% of the human genome. A number of skeletal deformities are linked to chromosome 12, including hypochondrogenesis, achondrogenesis, Noonan syndrome, Kniest dysplasia and trisomy 12p. Chromosome 12 is also home to a homeobox gene cluster, which encodes crucial transcription factors for morphogenesis, as well as the natural killer complex gene cluster, which encodes C-type lectin proteins that mediate the NK cell response to MHC I interaction.

Beschreibung:   The apolipoprotein L gene family maps to a region on chromosome 22 and encodes six highly homologous proteins designated apoL-I, apoL-II, apoL-III, apoL-IV, apoL-V and apoL-VI, all of which function as components of plasma lipoproteins. ApoL-IV (apolipoprotein L-IV), also known as APOL4, is a 351 amino acid protein that exists as multiple alternatively spliced isoforms, one of which is secreted. Expressed in spleen, placenta, spinal cord, uterus, testis and trachea, apoL-IV is thought to play a role in lipid exchange and transport throughout the body and may be involved in reverse cholesterol transport, specifically from peripheral cells to the liver. Overexpression of apoL-IV is associated with schizophrenia, suggesting that apoL-IV may play a role in the pathogenesis of neural disorders.
