Interleukin-4 (IL-4) is a cytokine produced by type 2 helper T cells, the Th2 cells. These cells tends to make a specific set of lymphokines including IL-4, IL-5, IL-6, IL-10, IL-13, IL-3 and GM-CSF and fail to produce IL-2, IFN-gamma, and lymphotoxin (TNF-beta). In addition, mast cells can produce IL-4. IL-4 exerts numerous effects on various hematopoietic cell types. On B cells, IL-4 promotes immunoglobulin class switching to IgE and IgG1 isotypes and upregulates MHC class II and CD23 expression. IL-4 promotes survival, growth, and differentiation of both T and B lymphocytes, mast cells and endothelial cells. In addition, IL-4 inhibits the production of TNF, IL-1, and IL-6 by macrophages.

Kaposi's sarcoma-associated herpesvirus (KSHV) belongs to the gamma-(2)-herpesvirus subfamily and has been closely linked to the Kaposi’s sarcoma, primary effusion lymphoma (PEL) and multicentric Castleman’s disease. The genome of KSHV is 165-170 kb and contains at least 88 open reading frames. At least five major proteins are likely to be involved in the assembly of the HHV-8 capsid, including a protease (encoded by ORF17), the major capsid protein (encoded by ORF25), and three other smaller capsid proteins (encoded by ORF62, ORF26, and ORF65). Previous structural studies have shown that the HSV-1 triplex is a monomer of VP19c and a dimer of VP23 and that the HCMV triplex is similarly composed of a monomer and a dimer. By analogy, the KSHV triplexes are likely also composed of a monomer of the ORF62 protein and a dimer of the ORF26 protein, which are the respective homologs of VP19c and VP23.

PPARD is a member of the peroxisome proliferator-activated receptor (PPAR) family. PPARs are nuclear hormone receptors that bind peroxisome proliferators and control the size and number of peroxisomes produced by cells. PPARs mediate a variety of biological processes, and may be involved in the development of several chronic diseases, including diabetes, obesity, atherosclerosis, and cancer. This protein is a potent inhibitor of ligand-induced transcription activity of PPAR delta and PPAR gamma. It may function as an integrator of transcription repression and nuclear receptor signaling. The expression of this gene is found to be elevated in colorectal cancer cells. The elevated expression can be repressed by adenomatosis polyposis coli (APC), a tumor suppressor protein related to APC/beta-catenin signaling pathway. Knockout studies in mice suggested the role of this protein in myelination of the corpus callosum, lipid metabolism, and epidermal cell proliferation.

Interferon-gamma-inducible 10 kD protein (IP-10), is a CXC chemokine with chemoattractant properties for CD4-positive T cells and inhibits early normal and leukemic hemopoietic progenitor proliferation. IP-10 is produced by a wide variety of cell types ranging from neutrophils and monocytes to hepatocytes, endothelial cells and keratinocytes. The cytokine is reported to be involved in a scala of inflammatory pathologies such as HIV encephalitis, cutaneous T cell lymphoma, chronic hepatitis and acute anterior uveitis. Various observations strongly suggest a role for the CXC chemokines IL-8 and IP-10 in the regulation of angiogenic activity in cancer and in idiopathic pulmonary fibrosis.

IL-27 Antibody: Like interleukin-23 (IL-23), IL-27 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the p40-related protein EBI3 and an IL-12 p35-related protein termed p28. IL-27 is produced after activation by antigen-presenting cells and induces proliferation of naïve but not memory CD4+ T-cells. It acts by binding to its receptor WSX-1 and gp130 which results in the activation of a Jak/STAT signaling cascade, suggesting the IL-27 is involved in the regulation of immune processes. It has been suggested that IL-27 can also be used as a therapeutic agent against cancer as it can also induce tumor-specific anti-tumor activity mediated through CD8+ T-cells, IFN-gamma, and T-bet.

DAPK2 Antibody: Apoptosis is mediated by death domain containing adapter molecules and a caspase family of proteases. Certain serine/threonine protein kinases, such as RIP and DAP kinase, are mediators of apoptosis. DAP kinase (DAPK) is pro-apoptotic calcium-regulated serine/threonine kinase containing death domain. Ectopic expression of DAPK induces cell death and suppresses oncogenic transformation. DAPK mediates IFN gamma induced apoptosis. A novel DAP kinase-related protein was recently identified and designated DAPK2 and DRP-1. Ectopicly expressed DAPK2 induced apoptosis in various types of cells. DAPK has high sequence homology to ZIP kinase and DRAK1/2, and they represent a novel family of serine/threonine kinases, which mediates apoptosis through their catalytic activities. The messenger RNA of DAPK2 is expressed in multiple human tissues.
