SPP1 Antibody: The secreted protein 1 (SPP1), also known as osteopontin, is a major noncollagenous protein of bone, but is also found in the extracellular matrix of other mineralized tissues and in bodily fluids. In bone, SPP1 is produced by osteoblasts, osteocytes, macrophages, and osteoclasts (1,2). SPP1 binds to cells through integrin and non-integrin receptors, as well as the adhesion receptor CD44 in an RGD-independent manner, enabling SPP1 to induce a number of functional effects including macrophage chemotaxis, cytoprotection, and regulation of T helper type 1 cells. SPP1 can regulate biomineralization by inhibiting the formation of hydroxyapatite (3) and the growth of calcium oxalate crystals.

CHFR is an E3 ubiquitin-protein ligase required to transiently arrest cells in early prophase when they are exposed to microtubule poisons. It acts in early prophase before chromosome condensation, when the centrosome moves apart from each other along the periphery of the nucleus. CHFR probably promotes the formation of 'Lys-63'-linked polyubiquitin chains and functions with the specific ubiquitin-conjugating UBC13-MMS2 (UBE2N-UBE2V2) heterodimer. Substrates that are polyubiquitinated at 'Lys-63' are usually not targeted for degradation, but are rather involved in signaling cellular stress. This suggests that it may be involved in signaling the presence of mitotic stress caused by microtubule poisons.

This antibody is specific for a protein of 36 kDa, identified as Thymidylate Synthase (TS). Thymidylate synthase converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), which is essential for DNA biosynthesis. Thymidylate synthase is also a critical target for the fluoropyrimidines, an important group of anti-neoplastic drugs that are widely used in the treatment of solid tumors. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP which inactivates thymidylate synthase by formation of a ternary covalent complex in the presence of the folate cofactor 5,10-methylenetetrahydrofolate. Expression of thymidylate synthase protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas.

SHISA4 Antibody: SHISA4 plays an essential role in the maturation of presomitic mesoderm cells by individual attenuation of both FGF and WNT signaling. The Shisa family of single-transmembrane proteins is characterized by an N-terminal cysteine-rich domain and a proline-rich C-terminal region. Its founding member, Xenopus Shisa, promotes head development by antagonizing Wnt and FGF signaling. Shisa physically interacted with immature forms of the Wnt receptor Frizzled and the FGF receptor within the ER and inhibited their posttranslational maturation and trafficking to the cell surface. Loss of Shisa function sensitized the neuroectoderm to Wnt signaling and suppressed head formation during gastrulation.

PTRF Antibody: The polymerase I and transcript release factor (PTRF) enables the dissociation of paused ternary polymerase I transcription complexes from the 3' end of pre-rRNA transcripts, regulating rRNA transcription by promoting the dissociation of transcription complexes and the re-initiation of polymerase I on nascent rRNA transcripts. PTRF also localizes to caveolae at the plasma membrane and is thought to play a critical role in the formation of caveolae and the stabilization of caveolins, translocating from caveolae to the cytoplasm after insulin stimulation. PTRF is also thought to modify lipid metabolism and insulin-regulated gene expression. Mutations in this gene result in a disorder characterized by generalized lipodystrophy and muscular dystrophy.

This antibody reacts with the Ig domain of CD47 protein. It has been shown to inhibit polymorphonuclear neutrophil (PMN) transmigration across cell monolayers and matrix. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction.

Anosmin-1 has been shown to directly bind FGFR1 via its N-terminal cysteine-rich domain, whey-acidic protein-like domain, and its first FnIII repeat with the D2 and D3 ectodomains of FGFR1. It is thought that Anosmin-1 can modulate FGFR1 signaling and have opposing effects on the formation and activation of FGF2-FGFR1-heparing complex.

MCM7 encodes a protein that is one of the highly conserved mini-chromosome maintenance proteins (MCM) that are essential for the initiation of eukaryotic genome replication. The hexameric protein complex formed by the MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. The MCM complex consisting of this protein and MCM2, 4 and 6 proteins possesses DNA helicase activity, and may act as a DNA unwinding enzyme. Cyclin D1-dependent kinase, CDK4, is found to associate with this protein, and may regulate the binding of this protein with the tumorsuppressor protein RB1/RB.
