GCS is a cGMP-synthesizing enzyme, which is the major receptor for the neurotransmitter nitric oxide. It plays a crucial role in smooth muscle contractility, platelet reactivity and neurotransmission. GCS is a heme-containing heterodimer, consisting of one alpha subunit and one beta subunit. The heme moeity mediates NO activation, and this heme group also binds carbon monoxide (CO), which weakly stimulates the enzyme. Both NO and CO stimulation are enhanced by the allosteric activator 3-(5'-hydroxymethyl-2'furyl)-benzyl-indazole, YC-1. YC-1 can also stimulate GCS in a NO-independent manner. Both alpha and beta subunits are required for cGMP generation, and at least two isoforms exist for each subunit. Heterodimers consisting of alpha-1/beta-1 and alpha-2/beta-1 have been identified, and both display similar enzymatic activity. HGFA Inhibitor 1 is an endogenous inhibitor of the kunitz-type serine proteinase HGF-activator (HGFA). HGF was first described as a hepatocyte-specific mitogen and survival factor and has since been shown to exert a variety of actions on many cell types by binding to its MET receptor. HGF is activated by cleavage of the single-chain form to form a two-chain version by HGFA. HGFA Inhibitor 1 also inhibits trypsin and MTSP-1 (Matriptase), a serine proteinase that also activates HGF. HGFB Inhibitor 1 is a splice variant of HGFA Inhibitor 1 and contains a 16 amino acid insert after the first kringle domain, relative to the HGFA Inhibitor 1 sequence.

The proteasome is responsible for producing these antigens from the components of foreign pathogens. MHC class 1 molecules consist of an α heavy chain that contains three subdomains (α1, α2, α3) and a non-covalent associating light chain, known as β-2-Microglobulin. β-2-Microglobulin associates with the α3 subdomain of the α heavy chain and forms an immunoglobulin domain-like structure that mediates proper folding and expression of MHC class 1 molecules. The α1 and α2 domains of the α heavy chain form the peptide antigen-binding cleft. Mutations in the β-2-Microglobulin gene can enhance the progression of malignant melanoma phenotypes.
