Beschreibung:   Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. [provided by RefSeq, Jul 2008].

Beschreibung:   Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. These receptors are heteromeric protein complexes composed of multiple subunits, arranged to form ligand-gated ion channels. The classification of glutamate receptors is based on their activation by different pharmacologic agonists. The subunit encoded by this gene belongs to a family of AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate)-sensitive glutamate receptors, and is subject to RNA editing (AGA->GGA; R->G). Alternative splicing of this gene results in transcript variants encoding different isoforms, which may vary in their signal transduction properties.

Beschreibung:   The protein encoded by this gene is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein can be activated by various ligands including interferon-alpha, interferon-gamma, EGF, PDGF and IL6. This protein mediates the expression of a variety of genes, which is thought to be important for cell viability in response to different cell stimuli and pathogens. Two alternatively spliced transcript variants encoding distinct isoforms have been described. [provided by RefSeq].

Beschreibung:   The small leucine-rich proteoglycan (SLRP) family of proteins contains various proteins such as Decorin, Biglycan, Fibromodulin, Keratocan, Lumican, Osteoadherin and Osteoglycin. These proteins all have similar functions as they all mediate extracellular matrix organization and act as binding partners of TGF beta. Osteoglycin, which also may be designated osteoinductive factor (OIF), is a secreted protein detected in bone tissues. Osteoglycin induces the formation of bone in conjunction with either TGF-beta-1 or TGF-beta-2. The precursor form of the OGN gene product, designated Mimecan, is subject to in situ proteolytic cleavage to yield the mature Osteoglycin.

Beschreibung:   Voltage-dependent Ca2+ channels mediate Ca2+ entry into excitable cells in response to membrane depolarization, and they are involved in a variety of Ca2+-dependent processes, including muscle contraction, hormone or neurotransmitter release and gene expression.Ca2+ currents are characterized on the basis of their biophysical and pharmacologic properties and include L-, N-, T-, P-, Q-, and R- types. L-type Ca2+ currents initiate muscle contraction, endocrine secretion, and gene transcription, and can be regulated through second-messenger activated protein phosphorylation pathways. L-type calcium channels may form macromolecular signaling complexes with G protein-coupled receptors, thereby enhancing the selectivity of regulating specific targets.

Beschreibung:   This gene encodes a member of the RING zinc finger protein family found in striated muscle and iris. The product of this gene is localised to the Z-line and M-line lattices of myofibrils, where titin's N-terminal and C-terminal regions respectively bind to the sarcomere. In vitro binding studies have shown that this protein also binds directly to titin near the region of titin containing kinase activity. Another member of this protein family binds to microtubules. Since these family members can form heterodimers, this suggests that these proteins may serve as a link between titin kinase and microtubule-dependent signal pathways in muscle.

Beschreibung:   Extracellular glycoproteins fibrillin-1 and -2 are major components of connective tissue microfibrils. Fibrillin-2 containing microfibrils regulate the early process of elastic fiber assembly in tissue. Mutations in the fibrillin-2 gene resulting in impaired assembly of fibrillin-2 may lead to molecular congenital contractural arachnodactyly. Fibrillin-2 constitutes the backbone of microfibrils which insert directly into the lamina densa of basement membranes. Epithelial cells primarily deposit fibrillin into the extracellular matrix in a nonfibrillar form. Mutations in the 8-cysteine motif of Fibrillin-2 alters its binding to microfibril-associated glycoprotein-1 (MAGP-1), which may increase the severity of congenital contractural arachnodactyly.

Beschreibung:   GDE3, also known as glycerophosphodiester phosphodiesterase 3, glycerophosphodiester phosphodiesterase domain containing 2 (GDPD2) or osteoblast differentiation promoting factor (OBDPF), is a 539 amino acid protein belonging to the glycerophosphoryl diester phosphodiesterase family. Possessing glycerophosphoinositol inositolphosphodiesterase activity, GDE3 hydrolyzes glycerophosphoinositol to form inositol 1-phosphate and glycerol, and is suggested to play a role in Actin cytoskeleton remodeling and osteoblast differentiation and growth. A multi-pass membrane protein, GDE3 localizes to cell membrane and cytoplasm, and colocalizes with actin in the cytoskeleton. GDE3 contains one GDPD domain, binds calcium as a cofactor and is encoded by a gene mapping to human chromosome Xq13.1.
