There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH2 (protocadherin-2), also known as PCDHGC3 (protocadherin gamma subfamily C, 3) or PC43, is a 934 amino acid single-pass type I membrane protein that contains six cadherin domains and belongs to the protocadherin gamma family. Functioning as a calcium-dependent cell-adhesion protein, PCDH2 is thought to be involved in the establishment and maintenance of neuronal connections within the brain. Multiple isoforms of PCDH2 exist due to alternative splicing events.

PKA (or cAPK) is a cyclic AMP dependent protein kinase. When activated by the second messenger cAMP, PKA mediates diverse cellular mechanisms, including proliferation, ion transport, regulation of metabolism, plus gene transcription. PKA is comprised of two dimers of two subunits, R (regulatory) and C (catalytic). Two families of R subunit (RI and RII) and three C subunit isoforms (C alpha, C beta, and C gamma) have been identified each possessing distinct cAMP binding properties and resulting in different phosphorylation states. C subunit is activated through autophosphorylation and direct phosphorylation at Thr197 by PDK-1. Tissue specific expression of C gamma, indicates pressure on C gamma during evolution, acting to modulate it in a functionally specific way. Certain amino acid substitutions make C gamma a distinct member of the cAMP dependent subfamily of protein kinases, and suggest that C gamma may be distinct in its protein substrate specificity or its interaction with the different regulatory subunits.

Heterotrimeric G proteins function to relay information from cell surface receptors to intracellular effectors (1). Each of a very broad range of receptors specifically detects an extracellular stimulus (a photon, pheromone, odorant, hormone or neurotransmitter) while the effectors (e.g., adenyl cyclase), which act to generate one or more intracellular messengers, are less numerous. In mammals, G protein alpha, beta and gamma polypeptides are encoded by at least 16, 4 and 7 genes, respectively (2-5). Most interest in G proteins has been focused on their alpha subunits, since these proteins bind and hydrolyze GTP and most obviously regulate the activity of the best studied effectors. More recent evidence, however, has established an important regulatory role for the beta gamma subunits (6-8). The Gs subfamily of G alpha subunits includes two closely related proteins, Ga s and Ga olf, which respectively stimulate adenylate cyclase and mediate response to olfactory stimuli (9).

PCDHGA9 (Protocadherin gamma-A9) is a 932 amino acid that is one of 22 proteins encoded by the protocadherin gamma cluster. The protocadherein gamma cluster consists of three subfamilies (A, B and C) and PCDHGA9 is a member of the gamma subfamily A. PCDHGA9 is a type I transmembrane receptor containing six cadherin motifs and is expressed in the central nervous system where it localizes to synapses. Members of the gamma cluster of protocadherins are essential for neuronal survival. There are two isoforms of PCDHGA9 that are produced as a result of alternative splicing events.
