Beschreibung:   The GGA family of proteins (Golgi-localized, g-Adaptin ear-containing, ARF-binding proteins) are ubiquitous coat proteins that facilitate the trafficking of soluble proteins from the trans-Golgi network (TGN) to endosomes/lysosomes by means of interactions with TGN-sorting receptors, ARF (ADP-ribosylation factor), and clathrin. Members of the GGA family, GGA1,GGA2 (also known as VEAR) and GGA3, are multi-domain proteins that bind mannose 6-phosphate receptors (MPRs). GGAs have modular structures with an N-terminal VHS (VPS27, Hrs and STAM) domain followed by a GAT (GGA and Tom1) domain, a connecting hinge segment and a C-terminal GAE (g-Adaptin ear) domain. The amino-terminal VHS domains of GGAs form complexes with the cytoplasmic domains of sorting receptors by recognizing acidic-cluster di-leucine (ACLL) sequences. The human GGA3 gene maps to chromosome 17 and encodes a 723 amino acid protein that shares 46% sequence identity with GGA1 and 38% with GGA2.

Beschreibung:   GTP cyclohydrolase I (also designated dopa-responsive dystonia) catalyzes the conversion of GTP to D-erythro-7,8-dihydroneopterin triphosphate, the first and rate-limiting step in tetrahydrobiopterin (BH4) biosynthesis. Tetrahydrobiopterin is an essential cofactor for 3 aromatic amino acid monooxygenases: phenylalanine, tyrosine, and tryptophan hydroxylases. Animals can synthesize tetrahydrobiopterin in vivo from GTP through several enzymatic reactions.

Beschreibung:   The leucine-rich (LRR) repeat is a 20-30 amino acid motif that forms a hydrophobic å/∫ horseshoe fold, allowing it to accommodate several leucine residues within a tightly packed core. All LRR repeats contain a variable segment and a highly conserved segment, the latter of which accounts for 11 or 12 residues of the entire LRR motif. The primary function of these motifs is to provide a versatile structural framework to mediate the formation of protein-protein interactions. LRRs are present in a variety of proteins with diverse structure and function, including innate immunity and nervous system development. Several human diseases are associated with mutations in genes encoding LRR-containing proteins. LRRC23 (leucine-rich repeat-containing protein 23), also known as leucine-rich protein B7, is a 343 amino acid protein that contains eight LRR (leucine-rich) repeasts and one LRRCT domain. LRRC23 exists as two alternatively spliced isoforms and is encoded by a gene mapping to chromosome 12.

Beschreibung:   ILEI is a 227 amino acid, ubiquitously expressed protein containing an amino-terminal signal peptide. Elevated levels of ILEI translation are observed in oncogenic, Ras-transformed mammary epithelial cells and epithelial to mesenchymal transition (Emt) as well as tumor growth and metastasis. Also, overexpression of ILEI results in loss of ZO-1, a protein involved in tight junctions, and expression of cytoplasmic E-cadherin, which has been shown to influence loss of polarity and invasiveness. Due to this evidence, it is suspected that ILEI cooperates with oncogenic Ras to cause TGF∫-independent Emt and its overexpression is correlated with the invasion, metastasis and survival in a variety of cancers.

Beschreibung:   GUP1 is a 504 amino acid multipass membrane protein of the endoplasmic reticulum that functions as a membrane bound O-acyltransferase. With specific expression in heart, GUP1 negatively regulates amino-terminal palmitoylation of Shh by HHAT, a protein that is required for Shh signaling. Deletion of the gene encoding GUP1 results in higher sensibility to specific sphinogolipid biosynthesis inhibitors and resistance to ergosterol biosynthesis inhibitors, indicating that GUP1 is an essential component in lipid metabolism. Also, GUP1 also seems to be important for cell wall assembly and stability due to evidence in Saccharomyces cerevisiae GUP1 mutants, which exhibit altered plasma membrane lipid composition and membrane potential.

Beschreibung:   G protein-coupled receptors (GPRs), also known as seven transmembrane receptors, heptahelical receptors or 7TM receptors, comprise a superfamily of proteins that play a role in many different stimulus-response pathways. G protein coupled receptors translate extracellular signals into intracellular signals (G protein activation) and they respond to a variety of signaling molecules, such as hormones and neurotransmitters. GPR110 (G protein-coupled receptor 110), also known as PGR19, is a 911 amino acid protein that belongs to the G-protein coupled receptor 2 family and LN-TM7 subfamily. Characterized as an adhesion GPCR, GPR110 is a multipass membrane-bound protein with a long amino-terminus that contains multiple domains. One of these domains is the GPCR proteolytic site (GPS), which is essential for proteolytic cleavage of the amino-terminus and for cell surface expression.

Beschreibung:   GPR120, a member of the rhodopsin family of G protein-coupled receptors (GPCRs), is a 377 amino acid protein which is expressed in the intestine. GPR120 is a receptor for unsaturated long-chain FFAs (free fatty acids). FFAs act as signaling molecules and are an important energy source. They also employ various physiological responses through their GPCRs. One such response occurs when dietary FFAs stimulate GPR120. This stimulation promotes the secretion of glucagon-like peptide 1 (GLP-1) in vivo and in vitro. GLP-1 belongs to the class of molecules known as the incretins, which are associated with insulin secreted from the pancreas as a result of food intake. GLP-1 also inhibits glucagon and gastric acid secretion and gastric emptying. Consequently, the role of GPR120 in the secretion of GLP-1 is critical in the treatment of diabetes.

Beschreibung:   The UDP-Glucuronosyltransferases (UGT) comprise a family of enzymes that detoxify and enhance the urinary excretion of a wide variety of xenobiotic and endogenous substrates by transferring glucuronic acid to sulfhydryl, hydroxyl, aromatic amino, or carboxylic acid groups. They have been subdivided into two families, UGT1 and UGT2, based on the evolutionary divergence of their genes. The enzymes of the UGT1A family play an important role in the metabolism of dietary constituents, phenols, and therapeutic drugs, and also the glucuronidation of bilirubin and iodothyronines. The enzymes of the UGT2B family are involved in the metabolism of bile acids, phenol derivatives, catecholestrogens and steroids. Although it is widely recognized that the liver is the major site of glucuronidation, it is now clear that UGT enzymes are also found in extra-hepatic tissues.
