ZNF843 is a 348 amino acid protein that is encoded by a gene which maps to chromosome 16. Chromosome 16 encodes over 900 genes in approximately 90 million base pairs, makes up nearly 3% of human cellular DNA and is associated with a variety of genetic disorders. The GAN gene is located on chromosome 16 and, with mutation, may lead to giant axonal neuropathy, a nervous system disorder characterised by increasing malfunction with growth. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16 through the CREBBP gene which encodes a critical CREB binding protein. Signs of Rubinstein-Taybi include mental retardation and predisposition to tumour growth and white blood cell neoplasias. Crohn's disease is a gastrointestinal inflammatory condition associated with chromosome 16 through the NOD2 gene. An association with systemic lupus erythematosis and a number of other autoimmune disorders with the pericentromeric region of chromosome 16 has led to the identification of SLC5A11 as a potential autoimmune modifier. The ZNF843 gene product has been provisionally designated MGC46336 pending further characterisation.

Prion diseases or transmissible spongiform encephalopathies (TSEs) are manifested as genetic, infectious or sporadic, lethal neurodegenerative disorders involving alterations of the prion protein (PrP). Infectious PrPSc is highly expressed in the brain of animals affected by TSEs, including scrapie in sheep, BSE in cattle, and Cruetzfeldt-Jacob disease in humans. The PRND gene locus, located on human chromosome 20p, encodes for the doppel protein (Dpl), which exhibits approximately 25% sequence homology with PrP. Dpl is characterised by an alpha-helical conformation, intramolecular disulfide bonds, and two N-linked oligosaccharides, and it is presented on the cell surface by a glycosylphosphatidylinositol anchor. Dpl is highly expressed in adult testis and heart and is detectable in the brain of neonatal mice. Dpl does not appear to contribute to prion disease progression, but ectopic expression of Dpl is implicated in neuronal degeneration of ataxic PRP-deficient mice. Dpl is also thought to play a role in angiogenesis, specifically maturation of the blood-brain barrier.

Spinocerebellar ataxia (SCA) is an autosomal dominant neurodegenerative disorder characterised by ataxia and selective neuronal cell loss. SCA is caused by the expansion of a translated CAG repeat, encoding a polyglutamine tract in SCA gene products, known as ataxins. The ataxin proteins are ubiquitously expressed in nervous tissue, but are primarily detected in cerebellum, brain stem and spinal cord in the central nervous system. Ataxin-10 is a cytoplasmic protein that belongs to the family of armadillo repeat proteins. A loss of ataxin-10 in primary neuronal cells causes increased apoptosis of cerebellar neurons. Ataxin-10 interacts with p110, an O-Linked beta-N-acetylglucosamine transferase, and may be important in the regulation of intracellular glycosylation levels and homeostasis in the brain. Spinocerebellar ataxia type 10 (SCA10) is an autosomal dominant disorder that causes cerebellar ataxia and seizures. SCA10 is caused by an expansion of an ATTCT pentanucleotide repeat in intron 9 of the ataxin-10 gene.

In vitro, silences the attractive effect of NTN1 but not its growth-stimulatory effect and silencing requires the formation of a ROBO1-DCC complex. May be implicated in spinal chord midline post-crossing axon repulsion. In vitro, only commissural axons that crossed the midline responded to SLIT2. In the developing visual system appears to function as repellent for retinal ganglion axons by providing a repulsion that directs these axons along their appropriate paths prior to, and after passage through, the optic chiasm. In vitro, collapses and repels retinal ganglion cell growth cones. Seems to play a role in branching and arborization of CNS sensory axons, and in neuronal cell migration. In vitro, Slit homolog 2 protein N-product, but not Slit homolog 2 protein C-product, repels olfactory bulb (OB) but not dorsal root ganglia (DRG) axons, induces OB growth cones collapse and induces branching of DRG axons. Seems to be involved in regulating leukocyte migration.

HHCM is a 467 amino acid protein that has oncogenic transforming capabilities on a variety of different cell lines. When HHCM was transfected into BRL-1 (Buffalo rat liver) or NIH3T3 cells, the cells exhibited significant morphological changes, anchorage independent growth and loss of contact inhibition. When the cells were inoculated into rats and mice, they became highly tumorigenic. HHCM gene-related DNA sequences were found in at least nineteen hepatomas of Asian origin. The gene encoding HHCM maps to human chromosome 8, which is made up of nearly 146 million bases and encodes about 800 genes. Translocation of portions of chromosome 8 with amplifications of the c-Myc gene are found in some leukemias and lymphomas, and are typically associated with a poor prognosis.
