The OKT4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation (together with the MHC class II). The OKT4 and the RPA-T4 monoclonal antibodies recognize different epitopes of CD4 and they do not exhibit cross-block binding.

CNOT4 Antibody: CNOT4 is a component of the CCR4-NOT transcription complex, a complex that is implicated in the repression of RNA polymerase II transcription. In the CCR4-NOT complex, CNOT4 acts as an E3 ubiquitin-protein ligase and interacts with a subset of E2 ubiquitin-conjugating enzymes through a unique C4C4 RING domain. This E3 ligase activity was shown to be dependent on the selective and specific interaction with the ubiquitin conjugating enzyme UbcH5B. In yeast, mutations in CNOT4 that prevented its interaction with the UbcH5B homolog UBC4 caused increased sensitivity to hydroxyurea, heat shock, and hygromycin B, suggesting that CNOT4 and UbcH5B are involved in stress response in vivo. Multiple isoforms of CNOT4 are known to exist.

CD40 Ligand (CD40LG) is a type II transmembrane glycoprotein that belongs to the TNF superfamily. Like other TNF superfamily members, CD40LG exists as a trimer in membrane bound and soluble form, both of which are bioactive. CD40LG is a ligand for CD40; its ligation also initiates signal transduction in CD40LG expressing cells. CD40LG is a differentiation antigen that is expressed on the surface of T-cells. It stimulates B-cell proliferation and secretion of all immunoglobulin isotypes in the presence of cytokines. CD40LG has been shown to induce cytokine production and tumouricidal activity in peripheral blood monocytes. It also co-stimulates proliferation of activated T-cells and this is accompanied by the production of IFN-gamma, TNF-alpha, and IL2.

Napsin is a pepsin-like aspartic proteinase, in the A1 clan of the AA clade of proteinases. There are two closely related napsins, napsin A and napsin B. Immunohistochemical studies revealed high expression levels of napsin A in human lung and kidney but low expression in spleen. It is expressed as a single chain protein in type II pneumocytes and in adenocarcinomas of lung. The high specificity expression of napsin A in adenocarcinomas of lung is useful to distinguish primary lung adenocarcinomas from adenocarcinomas of other organs.<sup>1-5</sup>

Interleukin-4 (IL-4) is a cytokine produced by type 2 helper T cells, the Th2 cells. These cells tends to make a specific set of lymphokines including IL-4, IL-5, IL-6, IL-10, IL-13, IL-3 and GM-CSF and fail to produce IL-2, IFN-gamma, and lymphotoxin (TNF-beta). In addition, mast cells can produce IL-4. IL-4 exerts numerous effects on various hematopoietic cell types. On B cells, IL-4 promotes immunoglobulin class switching to IgE and IgG1 isotypes and upregulates MHC class II and CD23 expression. IL-4 promotes survival, growth, and differentiation of both T and B lymphocytes, mast cells and endothelial cells. In addition, IL-4 inhibits the production of TNF, IL-1, and IL-6 by macrophages.

CD19 associates with CD21, CD81 and MHC class II to form a multi-molecular complex that initiates the mature B lymphocyte activation by interaction with the B-cell receptors. CD 19 enhances the B cell proliferation, development and activation, and the maturation of memory B cells. In CD19-deficient mice, the generation and maturation of B lymphocytes in the bone marrow and periphery are affected.
