Mouse Programmed cell death 1 ligand 1(Cd274，PD-L1), is a member of the growing B7 family of immune proteins.It involved in the costimulatory signal essential for T-cell proliferation and IFNG production in a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation by blocking cell cycle progression and cytokine production.B7-H1 has been identified as one of two ligands for programmed death1 (PD1), a member of the CD28 family of immunoreceptors. B7-H1 is constitutively expressed in several organs such as heart, skeletal muscle B7-H1 expression is upregulated in a small fraction of activated T and B cells and a much larger fraction of activated monocytes. The costimulatory function of B7-H1 is critical for enhancing maturation and differentiation of T-cells in lymphoid organs.B7-H1 expression is also induced in dendritic cells and keratinocytes after IFN gamma stimulation. Interaction of B7-H1 with PD1 results in inhibition of TCR-mediated proliferation and cytokine production. The B7-H1:PD1 pathway is involved in the negative regulation of some immune responses and may play an important role in the regulation of peripheral tolerance.

Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17 kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25 kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. TNF alpha is currently being evaluated in treatment of certain cancers and AIDS Related Complex.

GABA (gamma-aminobutyric acid) is the primary inhibitory neurotransmitter in the central nervous system and interacts with three different receptors: GABA(A), GABA(B) and GABA(C) receptor. The ionotropic GABA(A) and GABA(C) receptors are ligand-gated ion channels that produce fast inhibitory synaptic transmission. In contrast, the metabotropic GABA(B) receptor is coupled to G proteins that modulate slow inhibitory synaptic transmission. Functional GABA(B) receptors form heterodimers of GABA(B)R1 and GABA(B)R2 where GABA(B)R1 binds the ligand and GABA(B)R2 is the primary G protein contact site. Two isoforms of GABA(B)R1 have been cloned: GABA(B)R1a is a 130 kD protein and GABA(B)R1b is a 95 kD protein. G proteins subsequently inhibit adenyl cylase activity and modulate inositol phospholipid hydrolysis. GABA(B) receptors have both pre- and postsynaptic inhibitions: presynaptic GABA(B) receptors inhibit neurotransmitter release through suppression of high threshold calcium channels, while postsynaptic GABA(B) receptors inhibit through coupled activation of inwardly rectifying potassium channels. In addition to synaptic inhibition, GABA(B) receptors may also be involved in hippocampal long-term potentiation, slow wave sleep and muscle relaxation.

It exists as a heterodimer composed of the p40-related protein EBI3 and an IL-12 p35-related protein termed p28. IL-27 is produced after activation by antigen-presenting cells and induces proliferation of naïve but not memory CD4+ T-cells. It acts by binding to its receptor WSX-1 (also known as TCCR) and gp130 which results in the activation of a Jak/STAT signaling cascade, suggesting the IL-27 is involved in the regulation of immune processes. It has been suggested that IL-27 can also be used as a therapeutic agent against cancer as it can also induce tumor-specific anti-tumor activity mediated through CD8+ T-cells, IFN-gamma, and T-bet. IL-35 is composed of EBI3 and the p35 subunit of IL-12 and has been reported to have therapeutic effects against collagen-induced arthritis by expanding the population of regulatory T cells and suppressing Th17 cells. At least two isoform of EBI3 are known to exist.

The complement system provides innate defense against microbial infection and is a "complement" to antibody mediated immunity. The complement system consists of thirty five interacting plasma and membrane associated proteins which contribute to host-defense and initiate and amplify inflammation, even in the preimmune state where specific antibodies and lymphocytes are not available. In addition to the complement components themselves, this system also contains several soluble factors that prevent spontaneous complement activation from occurring in solution, as well as, several regulatory membrane associated proteins that protect host cells from accidental complement attack. Cleavage of the C3 component by either the classical pathway or the alternative pathway releases C3a and C3b. C3a appears to be important in many inflammatory responses while the C3b fragment covalently binds to the cell or bacterial surface and plays a role in opsonisation. Binding of C3b to the C4b component of the C3 convertase, results in C5 convertase (C4b3b2a) formation
The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer is standardized to an in-house standard by equivalence-point precipitation. Each IgG fraction is tested for purity and specificity at a minimum of 40 mg/mL using immunoelectrophoresis. The product is mostly goat IgG; no trace of albumin is detected. This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. Note: F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. The product has been successfully used in Immunoblotting. It detects C3 from human, equine, hamster, Guinea pig, murine, rabbit, rat, pig, and non-human primate samples. It does not cross react wtih bovine, canine, chicken, feline and sheep samples. Interleukin-23 (IL-23) is a heterodimeric cytokine composed of two disulfide-linked subunits, a p19 subunit that is unique to IL-23, and a p40 subunit that is shared with IL-12 (1-5). Although p19 is expressed by activated macrophages, dendritic cells, T cells, and endothelial cells, only activated macrophages and dendritic cells express p40 concurrently to produce IL-23. The functional IL-23 receptor complex consists of two receptor subunits, the IL-12 receptor beta 1 subunit (IL-12 Rbeta1) and the IL-23-specific receptor subunit (IL-23 R). IL-23 has biological activities that are similar to, but distinct from IL-12. Both IL-12 and IL-23 induce proliferation and IFN-gamma production by human T cells. While IL-12 acts on both naie and memory human T cells, the effects of IL-23 is restricted to memory T cells. In mouse, IL-23 but not IL-12, has also been shown to induce memory T cells to secret IL-17, a potent proinflammatory cytokine. IL-12 and IL-23 can induce IL-12 production from mouse splenic DC of both the CD8-and CD8+ subtypes, however only IL-23 can act directly on CD8+ DC to mediate immunogenic presentation of poorly immunogenic tumour/self peptide.

Programmed cell death 1 ligand 2 (PD-L2), also known as butyrophilin B7-DC or PDCD1 ligand 2, belongs to the member of B7 family which can regulate the activation and tolerance of T cells. PD-L2 is one ligand for Programmed cell death 1(PD-1), and the other is PD-L1. These two ligands shares 34% aa sequence identity. Mouse PD-L2 gene encodes a 273 amino acids (aa) protein with a putative 19 aa signal peptide, a 201 aa extracellular region , a 21 aa transmembrane domain and a 32 aa cytoplasmic region. The mouse PD-L2 gene is highly expressed in heart, placenta, pancreas, lung and liver while expressed weakly in spleen, lymph nodes and thymus. Besides, the expression of PD-L2 gene can be induced on dendritic cells grown from peripheral blood mononuclear cells under CSF2 and IL4/interleukin-4 treatment, and up-regulated by IFNG/IFN-gamma stimulation in monocytes. PD-L2 usually functions in a PDCD1-independent manner and is involved in regulating costimulatory signal which is essential for T-cell proliferation and IFNG production. Recent studies demonstrate that the expression of PD-L2 on the tumour cells promotes CD8 T cell–mediated rejection of tumour cells, at both the induction and effector phase of antitumour immunity. Moreover, PD-L2 binds to PD-1 cells and enhances T cell killing in a PD-1–independent mechanism.
