This gene encodes a polypeptide hormone precursor that undergoes extensive, tissue-specific, post-translational processing via cleavage by subtilisin-like enzymes known as prohormone convertases. There are eight potential cleavage sites within the polypeptide precursor and, depending on tissue type and the available convertases, processing may yield as many as ten biologically active peptides involved in diverse cellular functions. The encoded protein is synthesized mainly in corticotroph cells of the anterior pituitary where four cleavage sites are used; adrenocorticotrophin, essential for normal steroidogenesis and the maintenance of normal adrenal weight, and lipotropin beta are the major end products. In other tissues, including the hypothalamus, placenta, and epithelium, all cleavage sites may be used, giving rise to peptides with roles in pain and energy homeostasis, melanocyte stimulation, and immune modulation. These include several distinct melanotropins, lipotropins, and endorphins that are contained within the adrenocorticotrophin and beta-lipotropin peptides. Mutations in this gene have been associated with early onset obesity, adrenal insufficiency, and red hair pigmentation. Dimethyl Sulfoxide is a dipola, aprotic solvent. It has been shown to accelerate strand renaturation (1-10% concentration) and is believed to give the nucleic acid thermal stability against depurination. Dimethyl Sulfoxide is used to enhance dermal absorption of many chemicals, as a solvent for many organic and inorganic compounds including fats, carbohydrates, dyes, resins, and polymers, in antifreeze or hydraulic fluids, as a cryopreservative for cell cultures, oxidation of thiols and disulfides to sulfonic acids, as a PCR cosolvent to help improve yields, especially in long PCR. DMSO is routinely used in polymerase chan reaction (PCR), amplification of cDNA libraries, DNA sequencing, column-loading buffers for poly (A)+ RNA selection, buffers for the transformation of competent E. coli, and transfection protocols. To prepare sterile solutions use a teflon or nylon membrane to sterile-filter the DMSO - do not use a cellulose acetate membrane.

The 11B11 antibody binds to mouse Interleukin-4 (IL-4), a 14 kDa cytokine that is largely secreted by activated T cells of the Th2 subset, and to some degree by NKT and mast cells. This cytokine acts as a stimulatory factor for B cells, inducing their proliferation and differentiation, as well as playing a role in immunoglobulin class-switching. IL-4 may also provide autocrine stimulation for T cells, and affect the function of antigen presenting cells such as macrophages and dendritic cells. IL-4 can bind and signal via three cell surface receptor types: CD124 by itself, CD124 in combination with the common gamma chain (type I complex), or CD124 combined with CD213a1 (type II complex).

Guanylate cyclases belong to the adenylyl cyclase class-4/guanylyl cyclase family. There are two forms of guanylate cyclase. The soluble form, known as GCS or sGC, act as receptors for nitric oxide (NO). The membrane-bound receptor form, known as GC, are peptide hormone receptors. GCS is a cGMP-synthesizing enzyme, which is the major receptor for the neurotransmitter nitric oxide. It plays a crucial role in smooth muscle contractility, platelet reactivity and neurotransmission. GCS is a heme-containing heterodimer, consisting of one alpha subunit and one beta subunit. The heme moeity mediates NO activation, and this heme group also binds carbon monoxide (CO), which weakly stimulates the enzyme. Both NO and CO stimulation are enhanced by the allosteric activator 3-(5'-hydroxymethyl-2'furyl)-benzyl-indazole, YC-1. YC-1 can also stimulate GCS in a NO-independent manner. Both alpha and beta subunits are required for cGMP generation, and at least two isoforms exist for each subunit. Heterodimers consisting of alpha-1/beta-1 and alpha-2/beta-1 have been identified, and both display similar enzymatic activity. The distribution of the beta-2 subunit seems to be much more restricted than the beta-1 subunit, with predominant expression in kidney and liver.
