iASPP (inhibitor of apoptosis stimulating protein of p53, RelA-associated inhibitor (RAI), NFKap B-interacting protein 1, protein phosphatase 1 regulatory subunit 13-like, PPP1R13L) is the third member of the ASPP family of proteins. Unlike ASPP1 and ASPP2, which interact with p53 and enhance its ability to induce apoptosis, iASPP inhibits apoptosis induced by p53 overexpression. iASPP has been identified in a yeast-two hybrid screen as an interacting protein of the p65 subunit of NFKap B (RelA),3 interacts with NFKap B in vivo, and acts as an efficient inhibitor of HIV-1 gene expression in which both NFKap b and Sp1 play major roles. iASPP expression is upregulated in human breast carcinomas expressing wild-type p53, and gene overexpression may play an important role in the leukemogenesis and/or progression of acute leukemia. The RING finger motif is a specialized DNA-binding zinc finger domain found in many transcriptional regulatory proteins. The ring finger protein (RNF) family includes any protein containing the signature RING finger motif. RNF8 is a ubiquitously expressed nuclear RING finger protein that acts as an E3 ubiquitin-protein ligase. It is required for the ubiquitination of some nuclear proteins and promotes their subsequent degradation. The heterodimeric ubiquitin-conjugating enzyme UBC13 interacts with RNF8, and they co-localize in the nucleus. RNF8 may regulate mediation of UBC13 polyubiquitylation by elongating the ubiquitin chains. RNF8 also binds to Retinoid X receptor alpha (RXR?, a member of the steroid hormone receptor superfamily. It increases RXR?mediated transactivation of the RXR?responsive element (RXRE) promoter in a dose-dependent manner, suggesting that RNF8 is a regulator of RXR?mediated transcriptional activity.

Can phosphorylate C1D not only in the presence of linear DNA but also in the presence of supercoiled DNA. Ability to phosphorylate p53/TP53 in the presence of supercoiled DNA is dependent on C1D. Contributes to the determination of the circadian period length by antagonizing phosphorylation of CRY1 'Ser-588' and increasing CRY1 protein stability, most likely through an indirect machanism. Interacts with CRY1 and CRY2; negatively regulates CRY1 phosphorylation.

This receptor, known as kinase insert domain receptor, is a type III receptor tyrosine kinase. The signalling and trafficking of this receptor are regulated by multiple factors, including Rab GTPase, P2Y purine nucleotide receptor, integrin alphaVbeta3, T-cell protein tyrosine phosphatase, etc.. Calcyclin, also known as Protein S100-A6, growth factor-inducible protein 2A9, S-100 calcium-binding protein A6 (S-100A6) or MLN 4, is a homodimeric member of the S-100 calcium-binding protein family whose expression is upregulated in proliferating and differentiating cells. Calcyclin is inducible by growth factors and overexpressed in acute myeloid leukemias. It is expressed in a cell-specific manner in subpopulations of neurons and astrocytes and in epithelial cells and fibroblasts. Calcyclin is a specific target of S-100B protein in vivo. The binding of Calcyclin to S-100B is stabilized by S-100B-bound calcium and zinc. Calcyclin associates with both Annexin XI and CacyBP (calcyclin-binding protein). It functions to activate several processes along the calcium signal transduction pathway including the regulation of cell growth, proliferation, secretion and exocytosis.

The Interferon family of proteins are able to alter the expression of a variety of target genes, thereby controlling various events within the cell. IFI-35 (Interferon-induced 35 kDa protein), also known as IFP35, is a 286 amino acid interferon-induced protein. Localized to the nucleus and expressed in macrophages, fibroblasts and epithelial cells, IFI-35 is a leucine zipper protein that can form homodimers, but, unlike most leucine zipper proteins, cannot bind DNA. Upon induction by IFN-? IFI-35 associates with Nmi (N-Myc-interacting protein), resulting in the formation of a high molecular weight complex that is thought to play a role in IFN-?signaling and cellular responses. Once complexed with Nmi, IFI-35 is unable to be degraded by the proteasome, suggesting that IFI-35 is protected from degradation only when needed by IFN-? Two isoforms of IFI-35 exist due to alternative splicing events.
