Fibroblast growth factors (FGFs) produce mitogenic and angiogenic effects in target cells by signaling through the cellular surface tyrosine kinase receptors. There are four members of the FGF receptor family: FGFR-1 (flg), FGFR-2 (bek, KGFR), FGFR-3 and FGFR-4. Each receptor contains an extracellular ligand binding domain, a transmembrane region and a cytoplasmic kinase domain (1). Following ligand binding and dimerization, the receptors are phosphorylated at specific tyrosine residues (2). Seven tyrosine residues in the cytoplasmic tail of FGFR-1 can be phosphorylated: Tyr463, Tyr583, Tyr585, Tyr653, Tyr654, Tyr730 and Tyr766. Tyrosine 653 and 654 are important for catalytic activity of the activated FGFR and are essential for signaling (3). The other phosphorylated tyrosine residues may provide docking sites for downstream signaling components such as Crk and PLCgamma.

The protein encoded by this gene is a smooth muscle myosin belonging to the myosin heavy chain family. The gene product is a subunit of a hexameric protein that consists of two heavy chain subunits and two pairs of non-identical light chain subunits. It functions as a major contractile protein, converting chemical energy into mechanical energy through the hydrolysis of ATP. The gene encoding a human ortholog of rat NUDE1 is transcribed from the reverse strand of this gene, and its 3' end overlaps with that of the latter. The pericentric inversion of chromosome 16[inv(16)(p13q22)] produces a chimeric transcript that encodes a protein consisting of the first 165 residues from the N terminus of core-binding factor beta in a fusion with the C-terminal portion of the smooth muscle myosin heavy chain. This chromosomal rearrangement is associated with acute myeloid leukemia of the M4Eo subtype. Alternative splicing generates isoforms that are differentially expressed, with ratios changing during muscle cell maturation. The sodium-coupled neutral amino acid transporters (SNAT) of the SLC38 gene family include System A subtypes SNAT1, SNAT2 and SNAT4 and System N subtypes SNAT3 and SNAT5. The SLC38 transporters are essential for the uptake of nutrients, energy production, metabolism, detoxification, and the cycling of neurotransmitters. SNAT2, also designated ATA2, PRO1068 and SAT2 is encoded by the human gene SLC38A2. The functional role of SNAT2 in the nervous system is unclear. Protein expression is notably enriched in the spinal cord and brain stem nuclei of the auditory system. System A transport proteins are also present in placental tissue. These SNAT proteins may play a significant role in fetal development and inhibition of the transport system has been associated with fetal growth retardation.

Zinc-finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. The majority of Zinc-finger proteins contain a Kr_ppel-type DNA binding domain and a KRAB domain, which is thought to interact with KAP1, thereby recruiting histone modifying proteins. HIC-2 (hypermethylated in cancer 2) possesses Zinc finger motifs that are thought to be important for DNA-binding and also has a BTB/POZ domain at the N-terminus, which is thought to be important for protein-protein binding, as well as for the binding of transcription factors. HIC-2 is also known as Hic-3, HIC1-related gene on chromosome 22 or Zinc finger and BTB domain-containing protein 30, and is a 615 amino acid protein that is expressed as two isoforms produced by alternative splicing. HIC-2 is highly expressed in cerebellum and is localised to the nucleus in cells. HIC-2 contains a short amino acid sequence that is thought to interact with CtBP, a transcriptional repressor. The gene sequence associated with HIC-2 is thought to be a target for miRNAs (microRNAs) which are expressed in many cancers, suggesting that HIC-2 could possess tumor suppressor capabilities.

G protein-coupled receptors (GPRs) are a protein family of transmembrane receptors that transmit an extracellular signal (ligand binding) into an intracellular signal (G protein activation). GPR signaling is an evolutionarily ancient mechanism used by all eukaryotes to sense environmental stimuli and mediate cell-cell communication. All of the receptors have seven membrane-spanning domains and the extracellular parts of the receptor can be glycosylated. These extracellular loops also contain two highly conserved cysteine residues which create disulfide bonds to stabilize the receptor structure. GPR103 is a 455-amino acid protein with highest expression in the brain, retina, trigeminal ganglion, hypothalamus and vestibular nucleus. In peripheral tissues, GPR103 is expressed only in the heart, kidney and testis. GPR103 may regulate adrenal function. A hypothalamic neuropeptide of the RFamide family (26RFa) acts as an endogenous ligand for GPR103.

In kidneys, FOXD1 controls the production of signals required for the normal transition of induced mesenchyme into tubular epithelium and full growth and branching of the collecting system. Deletion of FOXD1 results in renal abnormalities. FOXD2 acts as a modulator of T cell activation.

Glycophorins A, B and C are sialoglycoproteins of the human erythrocyte membrane, which bear the antigenic determinants for the MN, Ss and Gerbich blood groups, respectively. Glycophorins span the membrane once and present their amino-terminal end to the extracellular surface of the human erythrocyte. The genetic array of expressed glycophorin surface antigens on erythrocytes defines the blood group phenotype of the individual. The human Glycophorin A gene maps to chromosome 4q31.21, contains seven exons which are 97% homologous to Glycophorin B, and encodes a 150 amino acid protein. The human Glycophorin B gene maps to chromosome 4q31.21 and encodes a 91 amino acid protein. The human Glycophorin C gene maps to chromosome 2q14.3 and contains four exons. Glycophorin C transcript can generate two protein isoforms. Isoform 1 includes all 4 exons and encodes the full length 128 amino acid protein. Isoform 2 is missing exon 2 and encodes a 109 amino acid protein, which specifies the Yus subtype of the Gerbich phenotype.

This protein is located in the endoplasmic reticulum, and has been shown to possess ubiquitin ligase activity. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.
