Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHB1 (Protocadherin beta 1) is an 818 amino acid protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whose genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB1) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. PCDHB1 is a single-pass type I membrane protein containing six cadherin domains.

KLHL13 (kelch like 13), also known as BKLHD2 (BTB and kelch domain containing protein 2), is a 604 amino acid protein that contains six Kelch repeats and one BTB/POZ domain. Expressed predominantly in brain, KLHL13 is believed to play a role in protein ubiquitination and may function as a substrate specific adapter of an E3 ubiquitin protein ligase complex. E3 ligases accept a ubiquitin residue from an E2 ubiquitin conjugating enzyme and immediately transfer that residue to a protein that is targeted for degradation. Specifically, KLHL13 interacts with KLHL9 and CUL3, a member of the cullin family of mediators that participate in the selective targeting of proteins for ubiquitin mediated proteolysis. Defects in the gene encoding SNAP 29 are the cause of CEDNIK syndrome, a neurocutaneous syndrome that is associated with cerebral dysgenesis, neuropathy, ichthyosis and palmoplantar keratoderma. The gene encoding SNAP 29 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

TSH is a glycoprotein hormone composed of two subunits which are non covalently bound to one another. Each of these hormones also has a unique beta subunit, which provides receptor specificity. Expressed ubiquitously, MARCH9 exists as a homodimer and functions as an E3 ubiquitin-protein ligase that accepts a ubiquitin residue from an E2 ubiquitin-conjugating enzyme and is thought to promote the degradation of target proteins, such as CD4 and MHC-I. Multiple isoforms of MARCH9 exist due to alternative splicing events.
