RREB1 (also known as RAS-responsive element binding protein 1 and Raf responsive zinc finger protein) is a transcription factor that binds specifically to the distal RAS-responsive element (RRE) in the calcitonin gene promoter and augments the Ras/Raf-mediated transcriptional response of that promoter. RREB1 may be involved in Ras/Raf-mediated cell differentiation. RREB is a nuclear protein expressed in heart, placenta, lung, liver, skeletal muscle, kidney and pancreas and is not found in the brain. The protein contains 4 C2H2-type zinc fingers.

Sprouty-4 (also known as SPRY4) is an inhibitor of the insulin receptor and EGFR-transduced mitogen-activated protein kinase (MAPK) signaling pathway downstream of FGF and EGF receptor tyrosine kinase activation. It is positioned upstream of RAS activation and impairs the formation of active GTP-RAS. SPRY4 is widely expressed, with different isoforms. The protein consists of 322 amino acids, with a cysteine-rich region, Src homology 3 binding regions, proline-rich regions, and a PEST sequence. It is expressed predominantly in the cytoplasm. Northern results show bands across most tissues, with strongest expression in heart, brain, placenta, lung, and intestine.

PACRG (also known as Parkin coregulated gene protein and PARK2 coregulated) is a gene located very close to parkin, in reverse orientation on the chromosome. It is thought to be co-transcribed with parkin by a bi-directional promoter between the two genes. PACRG is expressed in all immune tissues, spleen, lymph nodes, thymus, tonsils, leukocyte and bone marrow and is also expressed in heart, brain, skeletal muscle, kidney, lung and pancreas. PACRG is expressed in primary Schwann cells and very weakly by monocyte-derived macrophages, which are the primary host cells of Mycobacterium leprae, the causative agent of leprosy. Splice variants have been described for this protein.

Glycogen synthase kinase-3 (GSK 3) is a proline-directed serine-threonine kinase that was initially identified as a phosphorylating and inactivating glycogen synthase. Two isoforms, alpha (GSK 3A) and beta, show a high degree of amino acid homology. GSK 3B is involved in energy metabolism, neuronal cell development, and body pattern formation. GSK 3B participates in the Wnt signaling pathway and has been implicated in the hormonal control of several regulatory proteins including glycogen synthase, MYB and the transcription factor JUN. GSK 3B phosphorylates JUN at sites proximal to its DNA-binding domain, thereby reducing its affinity for DNA.

Nestin, is a large intermediate filament protein (class Type VI) that was first identified with a monoclonal antibody by Hockfield and McKay (1985). Nestin is expressed predominantly in stem cells of the central nervous system in the neural tube. Nestin is expressed during development and in myotendinous and neuromuscular junctions. Nestin expression is restricted, typically disappearing by E18. Nestin is thought to be a reasonable neuronal marker; however, recent studies have found nestin expression in other cell types such as endothelial cells. Nestin expression is seen in almost all GBMs (Glioblastoma mulitformes) and many melanomas (both primary and metastatic) but not in any metastatic carcinomas. Upon terminal neural differentiation, nestin is downregulated and replaced by neurofilaments.
