Chemokines also play fundamental roles in the development, homeostasis, and function of the immune system, and they have effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis. Chemokines are divided into 2 major subfamilies, CXC and CC, based on the arrangement of the first 2 of the 4 conserved cysteine residues; the 2 cysteines are separated by a single amino acid in CXC chemokines and are adjacent in CC chemokines. Mice lacking this gene displayed an underdeveloped uterus and reduced estrogen responsiveness demonstrating that this protein is essential for estrogen dependent proliferation. TRIM25 expression is increased in breast carcinoma where it promotes the growth of breast tumor by functioning as an E3 ubiquitin ligase that targets the negative cell cycle checkpoint 14-3-3s. Recent reports have indicated that TRIM25 E3 ubiquitin ligase is essential for the retinoic-acid-inducible gene 1 (RIG-1) signaling pathway to elicit host antiviral innate immunity.

SNCG(also designated gamma-synuclein or breast cancer-specific protein 1),with 127-amino acid protein(about 14 kDa), belongs to the synuclein family, which also includes alpha- and beta- synuclein.Three synucleins are located in the neuronal cytosol and enriched in presynaptic terminals,while SNCG is also expressed in many other non-neuronal tissues. SNCG is abnormally expressed in a high percentage of tumor tissues of diversified cancer types, including liver, esophagus, colon, gastric, lung, prostate, cervical, and breast cancer, but rarely expressed in tumor-matched nonneoplastic adjacent tissues. High levels of SNCG have been identified in advanced breast carcinomas suggesting a correlation between overexpression of SNCG and breast tumor development.

Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

This antibody recognises HMW Keratin 1, 5, 10 and 14. In normal epithelia, it stains stratified epithelia, myoepithelial cells and basal cells in the prostate gland and bronchi. The HMW Cytokeratin antibody shows no reactivity with hepatocytes, pancreatic acinar cells, proximal renal tubules, or endometrial glands; there is no reactivity with cells derived from simple epithelia. Mesenchymal tumors, lymphomas, melanomas, neural tumors, and neuroendocrine tumors are negative with this mAb. It stains myoepithelial cells and has been shown to be useful in distinguishing prostate adenocarcinoma from benign prostate. It has also been useful in separating benign from malignant intraductal breast proliferations.

Recognises a protein of 36 kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.
