Dyrk is the homolog of the Drosophila mnb (minibrain) gene, which is required for neurogenesis (1–3). Dyrk is a dual-specificity tyrosine kinase and serine/threonine kinase, which is itself regulated by tyrosine phosphorylation (1). Several mammalian Dyrk related proteins have been identified and are thought to compose a family of dual specificity protein kinases (4). Dyrk family members, including Dyrk1A (originally Dyrk), Dyrk1B, Dryk1C, Dyrk2, Dyrk3, Dyrk4A and Dyrk4B, are thought to be involved in diverse cellular functions (4). Dyrk1A is a candidate gene that may be involved in Downs syndrome, and it has been found to be somewhat overexpressed in Downs syndrome (1,5). Two isoforms of human fetal brain Dyrk2 exist: a deduced 528-amino acid protein and a protein containing 73 additional amino acids at the amino terminus (4). Dyrk3 is strongly expressed in testis, only after the onset of spermatogenesis, and very weakly expressed in spleen and adrenal gland (1). The genes which encode Dyrk2 and Dyrk3 map to human chromosomes 12 and 1q32, respectively (4).

Outer dense fibers are filamentous structures located on the outside of the axoneme in the midpiece and principal piece of the mammalian sperm tail. May help to maintain the passive elastic structures and elastic recoil of the sperm tail.Constituting the main cytoskeletal structure of spermatid flagella, outer dense fibers (ODFs) add elastic recoil, stiffness and protection against shear forces during sperm movement. Human ODFs consist of approximately 10 major and at least 15 minor proteins. The major proteins of the ODF include Odf1, Odf2, and Odf3, which compose a family of proteins that are preferentially expressed during mammalian spermiogenesis. Odf3 (outer dense fiber protein 3), also known as sperm tail protein SHIPPO 1 and TISP50 (transcript induced in spermiogenesis protein 50), is a 254 amino acid protein that is expressed during the latter part of spermatogenesis in flagella of elongated spermatids and mature sperm. Odf proteins are directed to their exact subcellular location by Spags, which are characterized as chaperone-like Odf-binding molecules. There are two isoforms of Odf3 that are produced as a result of alternative splicing events.

CCT2 is one of eight largely unrelated subunit proteins found in a protein chaperone complex known as the chaperonin-containing TCP-1 (CCT) or TRiC complex. The CCT complex is an abundanct cytoslic component that is credited with helping newly synthesised polypeptides adopt the correct conformation. Proteins that fold and assemble with the help of CCT include the cytoskeletal proteins actin and tubulin as well as up to 15% of newly synthesised eukaryotic proteins. CCT2 is the _-subunit of the chaperone complex and is one of several CCT proteins that exhibit increased expression in response to stress. This implies that the CCT complex helps cells recover from protein damage by assisting in protein folding and assembly. CCT subunit levels also change throughout the cell cycle, with lower proteins levels (and reduced chaperone activity) found during induced cell cycle arrest during at M phase. Each CCT subunit is thought to perform a specific function during protein folding and assembly; CCT2 exhibits both actin and tubulin binding activities but the exact molecular function on this subunit remains uncertain.
