Sorting nexin (SNX) proteins are members of a large family of hydrophilic PX (phospholipid-binding motif) domain-containing proteins that interact with a variety of receptor types. SNXs are widely expressed, although the tissue distribution of each SNX mRNA varies. The ability of SNXs to bind specific phospholipids, as well as their tendency to form protein-protein complexes, suggests a role for these proteins in cellular membrane trafficking and protein sorting. SNXs may also function specifically in pro-degradative sorting, internalization, endosomal recycling or simply in endosomal sorting. SNX10 (Sorting nexin-10) is a 201 amino acid protein that contains one phox domain and belongs to the SNX family. Like other members of the SNX family, SNX10 is thought to play a role in intracellular trafficking events throughout the cell.

The mature form of Factor X (Factor X A) is generated by Factor IX A- or Factor VII A-mediated cleavage at the tripeptide sequence, Arg-Lys-Arg, to yield a disulfide linked dimer. Together with the cofactor Factor V A and Ca2+ on the surface of platelets or endothelial cells, Factor X A coordinates as part of the prothrombinase complex, which mediates proteolysis of Prothrombin into active Thrombin. Mutations at the Factor X locus resulting in Factor X deficiencies can contribute to hemorrhagic diathesis.

Synaptotagmins are a large gene family of synaptic vesicle type III integral membrane proteins that function as regulators of both exocytosis and endocytosis and are involved in neurotransmitter secretion from small secretory vesicles. Synaptotagmin XIV, also known as SytXIV, is a 555 amino acid single-pass type III membrane protein belonging to the Synaptotagmin family. With the ability to form heterodimers, Synaptotagmin XIV mainly exists as a homodimer and contains two C2 domains, an N-terminal transmembrane domain and a putative fatty-acylation site. Synaptotagmin XIV is Ca2+-independent and may function in the trafficking and exocytosis of secretory vesicles to tissues outside the brain. Disruption of Synaptotagmin XIV may be affiliated with neurodevelopmental abnormalities. Synaptotagmin XIV exists as six alternatively spliced isoforms and is encoded by a gene on human chromosome 1q32.2.

TAK1 (or MAP3K7) was shown to participate in regulation of transcription by transforming growth factor beta (TGF beta). TAK1 is stimulated in response to TGF beta and bone morphogenetic protein. TAB1 induced TAK1 activation promoted the dissociation of active forms of IKKa and IKK b from active TAK1, whereas the IKK mutants remained to interact with active TAK1. TNF a activated endogenous TAK1, and the kinase negative TAK1 acted as a dominant negative inhibitor against TNF a induced NFkB activation. TAK1 was suggested to act as a regulatory kinase of IKKs.

Additionally, PIST is thought to regulate ionic currents via membrane channel modification and may also play a role in autophagy. Chromosomal aberrations in the gene encoding PIST are found in glioblastoma multiform (GBM), a common and aggressive form of brain tumor, suggesting a role for mutated PIST in carcinogenesis. Three isoforms of PIST exist due to alternative splicing events.

G6Pase-Beta, also known as SCN4, UGRP or G6PC3 (glucose 6 phosphatase, catalytic, 3), is a 346 amino acid endoplasmic reticulum multi-pass membrane protein that is involved in carbohydrate biosynthesis and the gluconeogenesis pathway. Inhibited by vanadate, G6Pase-Beta hydrolyzes GP6 to glucose in the endoplasmic reticulum. Due to its necessary involvement in normal glucose metabolism, G6Pase-Beta may play an integral role in diabetes and glycogen storage diseases (GSDs).

Infection by HPVs lead to persistent wart-like or macular lesions. TMC8 is expressed in placenta, prostate and testis, and three named isoforms exist as a result of alternative splicing events.

Recognizes a protein of 40 kDa, identified as CD7 (Workshop IV; Code T155). CD7 is a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. CD7 ligation induces production of D-3 phosphoinositides and tyrosine phosphorylation.

BAF53b is a member of a family of actin-related proteins (ARPs), which shares significant sequence identity with conventional actins. Both actins and ARPs comprise an actin fold, which forms an ATP binding cleft. BAF53b is a component of the BAF (BRG/brm-associated factor) complex in mammals, which is functionally related to the SWI/SNF complex in S. cerevisiae and Drosophila, and is associated with chromatin remodelling to facilitate transcriptional activation. As neural progenitors exit the cell cycle, the highly homologous BAF53a subunit of the BAF complex is replaced by BAF53b. This subunit switch is essential for the transition from from neural stem cells / progenitors to postmitotic neurons. BAF53b also plays a role in dendritic outgrowth, during which it is required to recruit the BAF complex and the dendritic regulator CREST to specific gene promoters. This function can not be performed by BAF53a.
