Nck is a highly conserved, oncogenic protein. It is a common target for the action of different surface receptors, encoding one SH2 and three SH3 domains, the Src homology motifs found in nonreceptor tyrosine kinases, Ras GTPase activating protein, phosphatidylinositol 3 kinase, and phospholipase Cg. Nck is widely expressed in various tissues and in cell lines from human, murine, and rat origins. Nck is phosphorylated on tyrosine, serine, and threonine residues in response to stimulation of EGF and PDGF in A431 and NIH 3T3 cells respectively. Like other SH2 containing proteins, Nck is associated with tyrosine autophosphorylated EGF or PDGF receptors via its SH2 domain. Overexpression of Nck leads to transformation of NIH 3T3 cells.

NGL-1 is a single pass type I membrane protein that acts as a cell adhesion molecule. It contains nine leucine-rich repeats (LRR) and one Ig-like C2-type domain. NGL-1 is predominantly expressed in the striatum and the cerebral cortex of both the embryonic and adult brain. NGL-1 specifically interacts with Netrin G1 (a molecule involved in axon guidance in the developing central nervous system) via its LRR region. NGL-1 plays a role in the regulation of neurite outgrowth of developing thalamic neurons. Soluble NGL-1 inhibits thalamic axon outgrowth while NGL-1 that is bound to the surface of developing thalamocortical axons stimulates growth. NGL-1 also interacts with Whirlin possibly stablising interstereociliar links.

Bloom’s syndrome is an autosomal recessive disorder characterized by pre- and post-natal growth deficiencies, sun sensitivity, immunodeficiency and a predisposition to various cancers. The gene responsible for Bloom’s syndrome, BLM, encodes a protein homologous to the RecQ helicase of E. coli and is mutated in most Bloom’s syndrome patients. One characteristic of Bloom’s syndrome is an increased frequency of sister chromatid exchange (SCE). BLM has been shown to unwind G4 DNA, and a failure of this function is thought to be responsible for the increased rate of SCE. BLM is known to be translocated to the nucleus, where its ATPase activity is stimulated by both single- and double-stranded DNA. Mutations in the yeast SGS1, a homolog of BLM, are known to cause mitotic hyperrecombination similiar to that observed in Bloom’s cells.

Constitutive expression of a VAV2 oncoprotein may result in morphological alterations including highly enlarged cells in which karyokinesis and cytokinesis frequently are uncoupled.

There are three protocadherin gene clusters designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. The protocadherein gamma cluster consists of three subfamilies (A, B and C). As a member of the gamma subfamily B, PCDHGB1 (Protocadherin gamma B1) is a 927 amino acid protein that is one of 22 proteins encoded by the protocadherin gamma cluster. Typical of gamma protocadherins, PCDHGB1 contains six cadherin motifs and is a type I transmembrane receptor expressed in the central nervous system. With localisation to synapses, members of the gamma cluster of protocadherins are essential for neuronal survival. There are two isoforms of PCDHGB1 that are produced as a result of alternative splicing events.

Results demonstrate that transmembrane semaphorins, like the secreted ones, can act as repulsive axon guidance cues. This gene encodes a class 6 vertebrate transmembrane semaphorin that demonstrates alternative splicing. Several transcript variants have been identified and expression of the distinct encoded isoforms is thought to be regulated in a tissue- and development-dependent manner.

The protein encoded by this gene is one of the highly conserved mini-chromosome maintenance proteins (MCM) that are essential for the initiation of eukaryotic genome replication. The hexameric protein complex formed by the MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. The MCM complex consisting of this protein and MCM2, 4 and 6 proteins possesses DNA helicase activity, and may act as a DNA unwinding enzyme. Cyclin D1-dependent kinase, CDK4, is found to associate with this protein, and may regulate the binding of this protein with the tumorsuppressor protein RB1/RB. Staphylococcal enterotoxins represent a group of proteins, which are secreted by Staphylococcus aureus and cause staphylococcal food poisoning syndrome. The illness is characterised by high fever, hypotension, diarrhea, shock, and in some cases death. Their molecular masses range between 27 and 30 kDa. At present, seven enterotoxins are known, namely A, B, C1, C2, C3, D and E. Their amino acid sequences have been determined and it was shown that all are single chain polypeptides containing one disulfide bond formed by two half cystines located in the middle of the polypeptide chain, which form the so called cysteine loop. SEB is an extremely potent activator of T cells, stimulating the production and secretion of various cytokines which mediate many of the toxic effects of SEB.
