The HOOK proteins, HOOK1, HOOK2 and HOOK3, comprise a family of cytosolic coiled-coil proteins that contain conserved N-terminal domains, which attach to microtubules; and more divergent C-terminal domains, which mediate binding to organelles. HOOK1, a cytoskeletal linker protein, may play a role in endocytic membrane trafficking. It exists as a homodimer, most likely mediated through its central coiled-coil domain. HOOK1 interacts with VPS18 and is required for spermatid differentiation, in which it is most likely involved in the positioning of the manchette microtubules and the flagellum. HOOK1 localizes primarily to the cytoplasm and does not associate with the Golgi complex, unlike HOOK3, which participates in the organization of the cis-Golgi compartment.

Contactin 2 is a neuronal cell adhesion molecule (CAM) that influences the formation of axon connections in the developing nervous system. Contactin 2 is a member of the immunoglobulin superfamily (IgSF) and contains a glycosylphosphatidylinositol-anchor, six immunogobulin (Ig)-like and four Fibronectin type III (FNIII)-like domains. Contactin 2 is expressed predominantly during neural development on the cell membrane of axons in nerve fiber tracts in order to guide commissural axons without promoting their growth. Contactin 2 binds with NgCAM in the plane of the same membrane (cis-binding). The Contactin 2 heterophilic (Contactin 2/NgCAM and Contactin 2/NrCAM) binding sites are localized to the first four Ig domains. The Contactin 2 homophilic (Contactin 2/Contactin 2) binding site is localized to the FNIII domain.

Eukaryotic gene transcription is regulated by sequence-specific transcription factors that bind modular cis acting promoter and enhancer elements. The ATF/CREB transcription factor family binds the palindromic cAMP response element (CRE) octanucleotide TGACGTCA. The best characterized members of this gene family include CREB-1, CREB-2 (also designated ATF-4), CRE-BPa, LZIP (also designated CREB3 and Luman), CREM-1, CREM-2, ATF-1, ATF-2, ATF-3, ATF-5, ATF-6 and ATF-7. This family of proteins contain highly divergent N-terminal domains, but share a C-terminal leucine zipper for dimerization and DNA binding. ATF-5 (ATFx), which can localize to the cytoplasm or the nucleus, binds DNA as a dimer. It interacts with CCND3 and PTP4A1.

The multiple human cytosolic dihydrodiol dehydrogenases are involved in the metabolism of xenobiotics, such as polycyclic aromatic hydrocarbons, pesticides and steroid hormones, and are responsible for the reduction of ketone-containing drugs by using NADH or NADPH as a cofactor. The 20alpha-HSD catalyzes the reaction of progesterone to the inactive form 20alpha-hydroxyprogesterone. The 3alpha-HSD is a cytosolic, monomeric, NADPH-dependent oxidoreductase that reduces 3-keto-5-dihydrosteroids to their tetrahydro products. DD1 and DD2 are ubiquitously expressed, whereas DD4 mRNA is restricted to the liver. DD3 is a unique enzyme that can specifically catalyze the dehydrogenation of trans-benzenedihydrodiol and trans-naphthalenedihydrodiol.
