Insulin is a pancreatic hormone that regulates glucose and is involved in the synthesis of protein and fat. It increases cell permeability to monosaccharides, amino acids and fatty acids. The insulin-link growth factors, IGF-I and IGF-II (also desinated somatomedin C and multiplication stimulating activator, respectvely), share approximatly 76% sequence identity and are 50% related to pro-insulin.IGF-I and IGF-II are nonglycosylated, single chain proteins of 70 and 76 amino acids in length, respectivelly. IGF-I functions as an autocrine regulator of growth in vaious, whereas the function of IGF-II is less well defined.

The family of guanylin regulatory peptides, including guanylin and uroguanylin, are strongly expressed in intestinal mucosa and regulate intestinal fluid secretion during digestion. Guanylins are also involved in acid neutralization and the regulation of membrane-bound guanylate cyclase Signalling molecules. Guanylin and uroguanylin are secreted primarily in the stomach, intestine, and colon. Guanylin is also detected in plasma. Guanylin is an endogenous activator of intestinal guanylate cyclase. It stimulates intestinal guanylate cyclase through the same receptor binding region as the heat-stable enterotoxins. Gut enterochromaffin cells synthesize guanylin to be a prohormone of 115 amino acids which is then is processed to the molecular form of 94 amino acids. This 10 kDa form is found circulating in the blood.

Indo-1, AM ester is membrane-permeant and thus can be loaded into cells via incubation. Because of the relatively low water solubility of the AM ester, Pluronic F-127, a mild detergent, is often used as a dispersing agent to facilitate cell loading. Indo-1-AM Ester itself does not bind Ca2+ , but it is readily hydrolyzed to indo-1 by endogenous esterases once it enters cells.

Arginase I which is expressed almost exclusively in the liver, catalyzes the conversion of arginine to ornithine and urea . The human arginase I gene, which maps to chromosome 6q23, encodes a 322 amino acid protein. Arginase I exists as a homotrimeric protein and contains a binuclear manganese cluster. Arginase II catalyzes the same reaction as arginase I, but differs in its tissue specificity and subcellular location. Specifically, arginase II localizes to the mitochondria. Arginase II is expressed in non-hepatic tissues, with the highest levels of expression in the kidneys, but, unlike arginase I, is not expressed in liver. The human arginase II gene, which maps to chromosome 14q24.1-q24.3, encodes a 354 amino acid protein. In addition, arginase II contains a putative amino-terminal mitochondrial localization sequence.
