EMAP II is a cytokine that is specifically induced by apoptosis. The release of this cytokine renders the tumor associated vasculature sensitive to tumor necrosis factor. Its precursor is identical to the p43 subunit, which is associated with the multi tRNA synthetase complex. Therefore, proEMAP II may function in binding RNA as part of the tRNA synthetase complex in normal cells and in stimulating inflammatory responses after proteolytic cleavage in tumor cells.
EMAP II inhibits angiogenesis of vascular beds and suppresses the growth of primary and secondary tumors without affecting normal tissues.

Recognises proteins of 33, 35 and 41 kDa, which are identified as various isoforms of CD74/CLIP. Its epitope is localized in the extracellular domain of CD74. CD74/CLIP is a type II transmembrane protein which binds to the peptide binding groove of newly synthesised MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. The molecule plays a critical role in the presentation of peptides, by the MHC class II antigens, to CD4 positive lymphocytes. CD74/CLIP is expressed on MHC class II positive cells including B cells, a subset of activated T cells, monocytes, and dendritic cells and by various types of carcinomas. It is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74/CLIP stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas and has been shown to be useful in differentiating atypical fibroxanthoma (-) from malignant fibrous histiocytoma (+).

Type I IFNs consist of IFN alpha, beta , τ, and ω and bind to the type I IFN receptor, whereas IFN- gamma is the only type II IFN and is specific for the type II IFN receptor. IFNW1 is a recently discovered protein structurally related to IFN-alpha and –beta. It has been shown that IFN-omega 1 similar to that of other human class I IFNs; potent antiviral activity was also observed on cells of bovine and ovine but not of equine or murine origin.

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play distinct roles in cells. The protein encoded by this gene is one of the PKC family members. There have been at least 12 different PKC isoforms identified in humans to date including alpha, beta I, beta II, gamma, delta, epsilon, zeta, eta, theta, iota, lambda, and mu. PKC gamma is expressed solely in the brain and spinal cord and its localization is restricted to neurons. It has been demonstrated that several neuronal functions, including long term potentiation (LTP) and long term depression (LTD), specifically require this kinase. Knockout studies in mice also suggest that this kinase may be involved in neuropathic pain development. Defects in this protein have been associated with neurodegenerative disorder spinocerebellar ataxia-14 (SCA14).

The SMAD2-SMAD4 complex is subsequently translocated to the nucleus where it modulates the transcription of the TGF-beta-regulated genes. This constitutes the canonical SMAD-dependent TGF-beta signaling cascade. Also involved in non-canonical, SMAD-independent TGF-beta signaling pathways. For instance, TGFBR1 induces TRAF6 autoubiquitination which in turn results in MAP3K7 ubiquitination and activation to trigger apoptosis. Also regulates epithelial to mesenchymal transition through a SMAD-independent signaling pathway through PARD6A phosphorylation and activation.
