RILPL2 is a 211 amino acid protein that belongs to the RILPL family. RILPL2 does not regulate lysosomal morphology or distribution. RILPL2 shares 32% and 18% amino acid identity with RILPL1 and RILP, respectively. RILPL2 as a novel interacting partner for the actin-based molecular motor MyoVa, and has a novel role for RILPL2 in controlling neuronal morphogenesis. It has been suggested that there is also a novel role for RILPL2 in the regulation of cellular shape and dendritic-spine morphogenesis, probably via the Rac1-Pak pathway. PCR analysis of human tissues detects highest RILPL2 expression in lung, followed by placenta. Lower expression is detected in liver, kidney, pancreas, heart and brain, but no expression is detected in skeletal muscle. The RILPL2 gene is conserved in chimpanzee, dog, cow, mouse, rat, chicken and zebrafish, and maps to human chromosome 12q24.31.

Napsin A is an aspartic proteinase that belongs to the peptidase A1 family and plays a role in pneumocyte surfactant processing. It is a 420-amino acid polypeptide consisting of a 24-residue signal peptide, a 40-amino acid propart, the mature enzyme of 336 amino acids, and a C-terminal extension of 18 residues. The mature Napsin A protein contains 3 predicted disulfide bonds, 3 potential N-linked oligosaccharide attachment sites, an RGD motif, a recognition motif for integrin binding, in the C terminus, immediately before a 4-amino acid insert that is unique to aspartic proteinases. Highest levels of Napsin A have been detected in adult lung (type II pneumocytes), fetal lung, and kidney tissues. Napsin A is also expressed at lower levels in adult spleen and at very low levels in peripheral blood leukocytes. Human napsin A shares 72.6% sequence identity with the mouse homolog.

SLITRK family proteins are integral membrane proteins that have a C-terminal domain that is partially similar to TRK neurotrophin receptor proteins and two leucine-rich repeat (LRR) domains that are similar to those of SLIT proteins. SLITRK4 (SLIT and NTRK-like protein 4) is a 837 amino acid single-pass type I membrane protein that contains 18 LRR (leucine-rich) repeats and is expressed in neural tissues, specifically in the thalamus, hypothalamus, subventricular zone, CA3 region of the hippocampus and cortical plate. SLITRK4 may be upregulated in some astrocytic brain tumors such as glioblastomas, astrocytomas and primitive neuroectodermal tumors. As compared with its family member SLITRK2, SLITRK4 only weakly suppresses neurite outgrowth. A study using genome-wide transcriptional profiling suggested that the gene encoding SLITRK4, as well as the ARL5B and PLA2G7 genes, may be involved in the pathogenesis of preeclampsia.

AT-motif binding factor 1 (ATBF1) binds to the AT-rich core sequence element in the human a-fetoprotein enhancer (1). Alternative splicing generates the ATBF1-A and ATBF1-B (2,3). While ATBF1-A contains a 920-amino acid extension at the N-terminus, both ATBF1-A and ATBF1-B contain 4 DNA-binding homeobox domains (2,3). Additionally, ATBF1-A contains 23 zinc finger motifs while ATBF1-B contains 18 zinc finger motifs (1–3). The N-terminal extension unique to ATBF1-A has transcriptional repressor activity (4). In the small intestine, ATBF1-A inhibits expression of the brushborder enzyme aminopeptidase-N through direct binding to the AT motif element (5). Besides functioning in transcription regulation, ATBF1 also functions in ATPase activity (6). ATPase activity associated with ATBF1-A is DNA/RNA-dependent and requires both homeobox domains and zinc finger motifs (6). ATBF1 is highly expressed in spleen and brain tissues (7). The gene encoding human ATBF1 maps to chromosome 16q22.3-q23.1 (8).
