The forkhead domain-containing gene family (Fox) comprises over 20 members in mammals and is defined by a conserved 110 amino-acid motif containing a winged helix structure DNA-binding domain. The members of this gene family have been implicated as key regulators of embryogenesis, cell cycling, cell lineage restriction and cancer. As such, FOXN2 contains a domain with homology to the forkhead DNA binding domain. FOXN2, or Human T-cell leukemia virus enhancer factor, is a 341 amino acid protein mapping to human gene FOXN2, which has been localized to human chromosome 2p16-p22. This protein, encoded by a 1239-bp cDNA isolated from the Jurkat cDNA library, is capable of binding to a region of the human T-cell leukemia virus long terminal repeat (HTLV-I LTR) located between amino acids 155 and 117. This purine-rich region is important in the regulation of gene expression by the Ets family of transcription factors. FOXN2 is a unique cellular gene that may function in the transcriptional regulation of HTLV-I LTR.

All the BAG proteins have an approximately 45-amino acid BAG domain near the C terminus but differ markedly in their N-terminal regions. The predicted BAG2 protein contains 211 amino acids. OLFM3 is a 478 amino acid protein that interacts with myocilin. Myocilin is an extracellular protein that plays a key role in the actomyosin system and is responsible for controlling intraocular pressure. OLFM3 is a secreted protein that contains an olfactomedin-like (OLF) domain, an approximately 260 amino acid motif commonly found in secreted glycoproteins. OLFM3 localizes to the Golgi apparatus of the cell and is highly expressed in both eye and brain tissue. Mutations in the gene that encodes OLFM3 may cause severe glaucoma, a condition in which increased intraocular pressure within the eyeball causes loss of eye sight.

Band 3, also designated AE1, is an erythrocyte membrane glycoprotein that contributes to cell stuctural integrity and mediates exchange of chloride and bicarbonate across the phospholipid bilayer. The diverse functions of the approximately 900 amino acid protein are mediated by two distinct domains. The amino terminal domain, also known as cdb3 for cytoplasmic domain of erthrocyte membrane band 3, acts as an attachment site for the erythrocyte skeleton by binding ankyrin. The carboxy-terminal, membrane-associated domain carries out exchange transport of anions. Degradation of band 3 can generate an aging antigen known as senescent cell antigen, or SCA, which is expressed on old cells and marks them for removal by the immune system. An isoform of band 3, which lacks the first 65 amino acids and does not bind ankryin, is expressed in kidney.

Members of the class-III pyridoxal-phosphate-dependent aminotransferase family, such as AGXT2, catalyze the conversion of glyoxylate to glycine using L-alanine as the amino donor. AGXT2 protects from asymmetric dimethylarginine (ADMA)-induced inhibition in nitric oxide (NO) production. Elevated blood concentrations of ADMA, a methyl derivate of the amino acid arginine and an endogenous inhibitor of nitric oxide (NO) synthase, is produced by the physiological degradation of methylated proteins and is found in association with diabetes, hypertension, congestive heart failure and atherosclerosis. AGXT2L2 (alanine-glyoxylate aminotransferase 2-like 2) is a 450 amino acid pyridoxal phosphate that exists as a homotetramer. Belonging to the class-III pyridoxal-phosphate-dependent aminotransferase family, AGXT2L2 localizes to the mitochondria and exists as three alternatively spliced isoforms. Encoded by a gene located on human chromosome 5q35.3, AGXT2L2 may have similar functions as AGXT2.
