Tumor necrosis factor receptor superfamily member 12A(Tnfrsf12a) is a single-pass type I membrane protein and contains 1 TNFR-Cys repeat. It is weak inducer of apoptosis in some cell types.It promotes angiogenesis and it is the proliferation of endothelial cells. It may modulate cellular adhesion to matrix proteins.TNFR binds specifically to tumor necrosis factor (TNF) and blocks its interaction with cell surface TNF receptors. TNF is a naturally occurring cytokine that is involved in normal inflammatory and immune responses. It plays an important role in the inflammatory processes of rheumatoid arthritis (RA), polyarticular-course juvenile rheumatoid arthritis (JRA), and ankylosing spondylitis and the resulting joint pathology. In addition, TNF plays a role in the inflammatory process of plaque psoriasis. Elevated levels of TNF are found in involved tissues and fluids of patients with RA, psoriatic arthritis, ankylosing spondylitis (AS), and plaque psoriasis. Two distinct receptors for TNF (TNFRs), a 55 kilodalton protein (p55) and a 75 kilodalton protein (p75), exist naturally as monomeric molecules on cell surfaces and in soluble forms. Biological activity of TNF is dependent upon binding to either cell surface TNFR.

NFX1 may play an important role in regulating the duration of an inflammatory response by limiting the period in which MHC class II molecules are induced by interferon-gamma. Isoform 3 binds to the X-box motif of TERT promoter and represses its expression. Together with PABPC1 or PABPC4, isoform 1 acts as a coactivator for TERT expression. NFX1 mediates E2-dependent ubiquitination.MHC class II gene expression is controlled primarily at the transcriptional level by transcription factors that bind to the X and Y boxes, two highly conserved elements in the proximal promoter of MHC class II genes. The protein encoded by this gene is a transcriptional repressor capable of binding to the conserved X box motif of HLA-DRA and other MHC class II genes in vitro. The protein may play a role in regulating the duration of an inflammatory response by limiting the period in which class II MHC molecules are induced by IFN-gamma. Three alternative splice variants, each of which encodes a different isoform, have been identified.

CIKS Antibody: Nuclear factor kappa B (NF-kappa B) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kappa B mediates the expression of a great variety of genes in response to extracellular stimuli. NF-kappa B associates with I kappa B proteins in the cell cytoplasm, which inhibit NF-kappa B activity. I kappa B is phosphorylated by I kappa B kinase (IKK) complex that contains IKK alpha , IKK beta , and IKK gamma. A novel molecule that associates with and activates IKK was recently identified and designated CIKS (for connection to IKK and SAPK/JNK) and Act1 (for NF-kappa B activator 1). CIKS directly interacts with IKK gamma. CIKS/Act1 also activates activating transcription factor (ATF) and activator protein 1 (AP-1) through Jun kinase (JNK). These results indicate that CIKS/Act1 is involved in the inflammation and stress responses. CIKS/Act1 is ubiquitously expressed in human tissues.

Interleukin-22(IL-22) is a member of a group of the IL-10 family, a class of potent mediators of cellular inflammatory responses. IL-22 is produced by activated DC and T cells. IL-22 and IL-10 receptor chains play a role in cellular targeting and signal transduction. It can initiate and regulate innate immune responses against bacterial pathogens especially in epithelial cells such as respiratory and gut epithelial cells. IL-22 along with IL-17 likely plays a role in the coordinated response of both adaptive and innate immune systems. IL-22 also promotes hepatocyte survival in the liver and epithelial cells in the lung and gut similar to IL-10. Biological activity of IL-22 is initiated by binding to a cell-surface complex consisting of IL-22R1 and IL-10R2 receptor chains. IL-22 biological activity is further regulated by interactions with a soluble binding protein, IL-22BP. IL-22BP and an extracellular region of IL-22R1 share sequence similarity. In some cases, the pro-inflammatory versus tissue-protective functions of IL-22 are regulated by cytokine IL-17A.

IL-22 Receptor Antibody: A novel cytokine, designated IL-TIF for IL-10 related T cell-derived inducible factor and IL-22, was recently identified. The receptor for IL-22 (IL-22R, also termed CRF2-9 and IL-TIF-R1 chain) is a new member of the class II cytokine receptor family. IL-22R forms a complex with IL-10 receptor beta chain and mediates IL-22 signaling. IL-22 and its receptor activate JAK-STAT signaling pathway. IL22R is expressed in normal liver and kidney and their cell lines HepG2 and TK-10. A soluble form of IL-22 receptor, also termed IL-22 binding protein (IL-22BP) and IL-22 receptor-alpha 2 (IL-22RA2), was identified very recently. IL-22BP prevents binding of IL-22 to the functional cell surface IL-22R complex and neutralizes IL-22 activity. LPS induces IL-22 expression, which indicates the role of IL-22 in inflammatory response.

Interleukin-21 (IL-21) is a key factor in the transition between innate and adaptive immune responses secreted by activated T cells. The IL-21 receptor (IL-21R) is expressed in lymphoid tissue, in particular by NK, B, T and dendritic cells, macrophages and endothelial cells. Recent evidence suggests that IL-21 plays a supportive role in the proliferation of T and B cells and influences the cytolytic activity of natural killer cells. IL-21 has been shown to up-regulate genes associated with innate immunity and to inhibit the differentiation of naive T helper cells. IL-21 specifically inhibits IFN-gamma production from developing TH1 cells and is preferentially expressed by TH2 cells. Furthermore IL-21 has been identified as a growth and survival factor for human myeloma cells. IL-21/IL-21R interactions have a unique role in sequentially activating both innate and adaptive immune responses against poorly immunogenic tumours, leading to tumour rejection that is perforin dependent but IFN-gamma independent.

Renin is also known as REN and angiotensinogenase, is a circulating enzyme that participates in the body's renin-angiotensin system (RAS), and plays an essential role in the elevation of arterial blood pressure and increased sodium retention by the kidney. Renin activates the renin-angiotensin system by cleaving angiotensinogen, produced by the liver, to yield angiotensin I, which is further converted into angiotensin II by ACE, the angiotensin-converting enzyme primarily within the capillaries of the lungs. Renin is secreted from kidney cells, which are activated via signaling from the macula densa, which responds to the rate of fluid flow through the distal tubule, by decreases in renal perfusion pressure (through stretch receptors in the vascular wall), and by sympathetic nervous stimulation, mainly through beta-1 receptor activation. Renin can bind to ATP6AP2, which results in a fourfold increase in the conversion of angiotensinogen to angiotensin I over that shown by soluble renin. In addition, renin binding results in phosphorylation of serine and tyrosine residues of ATP6AP2. The level of renin mRNA appears to be modulated by the binding of HADHB, HuR and CP1 to a regulatory region in the 3' UTR. An over-active renin-angiotension system leads to vasoconstriction and retention of sodium and water. These effects lead to hypertension. Therefore, renin inhibitors can be used for the treatment of hypertension.

Receptor-binding studies have demonstrated that GRO-alpha, -beta, and -gamma signal mainly through G protein-coupled receptors CXCR1 and CXCR2 (Ahuja and Murphy). GRO-beta is expressed in epithelial cells, monocytes, fibroblasts, and melanocytes and is further induced during inflammatory, epithelialization, and angiogenic processes, for example during the wound healing process of human burn wounds (Zaja-Milatovic and Richmond). GRO-beta also stimulates mitogenesis in certain human melanoma cells (Unemori <i>et al.</i>).
