The UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase (GalNAc-T) family of enzymes are substrate-specific proteins that catalyze the transfer of GalNAc (N-acetylgalactosaminyl) to serine and threonine residues of various proteins, thereby initiating mucin-type O-linked glycosylation in the Golgi apparatus. GalNAc-T10 (Polypeptide N-acetylgalactosaminyltransferase 10), also known as UDP-GalNAc:polypeptide N-acetylgalactosaminyltransferase 10, is a 603 amino acid single-pass type II membrane protein that prefers Muc5Ac and EA2 peptide substrates. The N-terminal domain is involved in substrate binding and manganese coordination, while the C-terminal domain is involved in UDP-Gal binding and catalytic reaction. GalNAc-T10 is widely expressed, with highest levels found in small intestine. There are four isoforms of GalNAc-T10 that are produced as a result of alternative splicing events.

Sp1 is a sequence-specific transcription factor that recognizes GGGGCGGGGC and closely related sequences, which are often referred to as GC boxes. Sp1 was initially identified as a HeLa cell-derived factor that selectively activates in vitro transcription from the SV40 promoter and binds to the multiple GC boxes in the 21-bp repeated elements in SV40. The sequence specificity of DNA binding is conferred by Zn (II) fingers, whereas a different region of Sp1 appears to regulate the affinity of DNA binding. Sp1 belongs to a subgroup of transcription factors that are phosphorylated upon binding to promoter sequences. Evidence suggests that the early growth response gene, Erg-1 (also known as Zif268 or NGF1-A) (7), may downregulate certain mammalian gene promoters by competing with Sp1 for binding to an overlapping binding motif. The gene encoding human Sp1 maps to chromosome 12q13.1.

Casein kinase I (also designated CKI) and casein kinase II (CKII) compose a family of serine/threonine protein kinases which are present in all eukaryotes examined to date. Casein kinase I family members, which include casein kinase I Alpha, I Gamma, I Delta and I Epsilon, have been implicated in the control of cytoplasmic and nuclear processes, including DNA replication and repair, membrane trafficking, circadian rhythm, cell cycle progression, chromosome segregation, apoptosis and cellular differentiation. Casein kinase I isoform alpha-like (CSNK1A1L) is a 337 amino acid protein that shares a high degree of sequence similarity with the alpha isoform of casein kinase 1. CSNK1A1L resides in the cytoplasm and participates in the Wnt signaling pathway. By utilizing ATP within its protein kinase domain, CSNK1A1L phosphorylates a large number of proteins.

A human fibrosarcoma cell line, HT1080, expresses both STC1 and STC2 as secreted phosphoproteins in vivo, with STC2 being phosphorylated by an ecto-CK2-like enzyme. STC1 and STC2 have opposite effects on calcium and phosphate homeostasis, namely anti-hypercalcemic and anti-hypocalcemic actions, respectively. STC1 and STC2 are detected in human adrenal tumors, such as pheochromocytoma, differentiated neuroblastoma aldosterone-producing adenoma, and in cultured adrenal tumor cells (rat pheochromocytoma PC-12 cells and human neuroblastoma NB-1 cells).

Mutations in the gene encoding GDAP1 is the cause of many forms of Charcot-Marie-Tooth disease, a common inherited disorder of the peripheral nervous system that is characterized by reduced nerve conduction velocities, slow progressive distal muscle atrophy and absent deep tendon reflexes.

CNOT2 (CCR4-NOT transcription complex subunit 2) is a ubiquitous protein encoded by the human gene CNOT2. CNOT2 belongs to the CNOT2/3/5 family and is part of the CCR4-NOT complex. The CCR4-NOT complex is an evolutionarily conserved, multi-component complex known to be involved in transcription as well as mRNA degradation. Various subunits (e.g. CNOT1, CNOT2) are involved in influencing nuclear hormone receptor activities. The CCR4-NOT complex is also involved in the regulation of Histone H3 lysine 4 methylation through a ubiquitin-dependent pathway that likely involves the proteasome. Increased expression of the CNOT2 subunit acts to strongly repress transcription by RNA polymerase II. This repressive effect is mediated by a conserved NOT-Box, which is located at the C-terminus of CNOT2 proteins. Repression by the NOT-Box is sensitive to treatment with the histone deacetylase (HDAC) inhibitor trichostatin A.

Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). CK8 is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Hepatocellular carcinomas are defined by the use of antibodies that recognize only cytokeratin 8 and 18. CK8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Anti-CK8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against CK8, CK18 and CK19). Anti-CK8 is useful for the differentiation of lobular (ring-like, perinuclear) from ductal (peripheral-predominant) carcinoma of the breast.

Binding to the intracellular domain of the ∫-Amyloid precursor protein, Fe65L2 is thought to modulate the internalization and, therefore, the accessibility and function of ∫-Amyloid. The PK136 antibody is specific for mouse NK1.1, a type II transmembrane lectin-like receptor and member of the killer cell lectin-like receptor (KLR) family. NK1.1 is prominently expressed on natural killer (NK) cells, and is correlated with NK cytotoxic effects toward virus-infected cells and tumor cells. NK1.1 is expressed on subsets of NKT cells in certain mouse strains (C57BL/6, FVB/N, and NZB), yet absent from others (AKR, BALB/c, CBA/J, C3H, DBA/1, DBA/2, NOD, SJL, and 129). Putative subsets of NK cells and their expression of NK1.1 antigen are of continuing interest, including NK1.1+/CD117+ (c-Kit) cells reported to be immunosuppressive for CD8+ T cells in a mechanism involving PD-1 and PD-L1 (Ehlers et al. 2012. Endocrinology. 10: 1247).
